Hippuric acid metabolism by Corynebacteriumglucuronolyticum-like bacteria promotes calcium oxalate crystallization.

Nephrolithiasis (kidney stone disease) incidence is rising, and microbes have been implicated in kidney stone formation, yet the underlying mechanisms remain incompletely defined. Here, urinary microbiome analysis reveals enrichment of Corynebacterium glucuronolyticum-like (C. glucuronolyticum-like) bacteria in calcium oxalate (CaOx) stone patients. In vivo and in vitro experiments suggest that C. glucuronolyticum-like bacteria promote CaOx crystal nucleation and aggregation in experimental models. Mechanistically, hippuric acid (HA) is identified as a key urinary anti-crystallization metaboli

Salidroside attenuates calcium oxalate-induced renal injury via suppression of PANoptosis.

Calcium oxalate (CaOx), the primary constituent of most urinary calculi, triggers inflammatory responses and tissue damage upon entry into renal tubules, playing a pivotal role in nephrolithiasis pathogenesis. Salidroside (SAL), a phenolic glycoside isolated from Rhodiola plants, exhibits diverse pharmacological activities. This study integrated network pharmacology with experimental validation to investigate the therapeutic potential and mechanistic basis of SAL against urinary stones. In both in vitro and in vivo models of CaOx crystal-induced kidney injury, SAL treatment markedly reduced re

Accumulation of heavy metals in the mycelium of Abortiporus biennis and their effect on oxalate oxidase activity.

Abortiporus biennis belongs to a basidiomycete fungi with the unique ability to degrade oxalate using enzyme oxalate oxidase. Oxalate oxidase (OXO) from Abortiporus biennis is an intracellular oxalic acid-induced enzyme, which catalyses the degradation of oxalic acid to carbon dioxide and hydrogen peroxide. OXO activity was stimulated by all the heavy metals tested (copper, manganese, lead and cadmium) after addition as water-soluble salts to A. biennis fungal cultures. Manganese ions stimulated OXO activity to the greatest extent among the tested metal ions. In the presence of manganese ions,

Therapeutic potential of oxalate decarboxylase (OxDC) in the treatment of urolithiasis (kidney stone diseases).

Kidney stone disease is a devastating medical condition that affects 12 % of the global population. The pathophysiology of renal stones is significantly influenced by calcium oxalate, which is present in most renal stones. Oxalate is obtained by humans from increasing dietary oxalate intake as well as from endogenous synthesis. High amounts of oxalate cause hyperoxaluria, which is a significant risk factor for recurrent nephrolithiasis. The available treatments for people with primary and secondary hyperoxaluria are insufficient and do not always result in a noticeable decrease in the excretio

Effects of calcium oxalate crystals on neutrophil cellular proteome and functions: implications for nephrolithiasis.

BACKGROUND: The majority of stone formers (87.5-95.9%) exhibit mild to moderate interstitial inflammation surrounding the stone. Neutrophils and neutrophil-derived genes/proteins have been found in renal papillae, stone matrix and urine of calcium oxalate monohydrate (COM) stone formers. However, neutrophil-crystal interactions, especially responses of neutrophils to COM crystals, remained unknown. METHODS: This study addressed the effects of COM crystals on neutrophil cellular proteome and functions, including phagocytosis, activation/degranulation, neutrophil extracellular traps (NETs) form

Harnessing Solvent Displacement Crystallization for Actinide Synthesis: Insights from Uranyl Oxalate.

To address the challenge of actinide crystallization in systems with a low chemical potential, solvent displacement crystallization (SDC) techniques are applied to synthesize uranyl oxalate in a series of alcohols with varying solvent polarity. This work demonstrates the simplicity of applying SDC to actinides and indicates that solvent polarity affects crystallizations. Uranyl oxalate trihydrate was synthesized from methanol, ethanol, 1-propanol, and isopropanol as additive solvents, with characterization indicating an absence of solvent influence on the bulk structure. The choice of solvent

Proposing Oxalic Acid as Chemical Storage of Carbon Dioxide to Achieve Carbon Neutrality.

Increasing emissions of carbon dioxide into the atmosphere due to the use of fossil fuels and ongoing deforestation are affecting the global climate. To reach the Paris climate agreement, in the coming decades low emission technologies must be developed, which allow for carbon removal on a Gt per year-scale. In this regard, we propose the electrochemical conversion of carbon dioxide to oxalic acid as a potentially viable pathway for large scale CO2 utilization and storage. Combined with water oxidation, in principle this transformation does not need stoichiometric amounts of co-reagents and mi

Oxalate Nephropathy After Kidney Transplantation: Risk Factors and Outcomes of Two Phenotypes.

Describing risk factors and outcomes in kidney transplant recipients with oxalate nephropathy (ON) may help elucidate the pathogenesis and guide treatment strategies. We used a large single-center database to identify patients with ON and categorized them into delayed graft function with ON (DGF-ON) and late ON. Incidence density sampling was used to select controls. A total of 37 ON cases were diagnosed between 1/2011 and 1/2021. DGF-ON (n = 13) was diagnosed in 1.05% of the DGF population. Pancreatic atrophy on imaging (36.4% vs. 2.9%, p = 0.002) and gastric bypass history (7.7% vs. 0%; p =

Multiplex gene editing reduces oxalate production in primary hyperoxaluria type 1.

Targeting key enzymes that generate oxalate precursors or substrates is an alternative strategy to eliminate primary hyperoxaluria type I (PH1), the most common and life-threatening type of primary hyperoxaluria. The compact Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR) from the Prevotella and Francisella 1 (Cpf1) protein simplifies multiplex gene editing and allows for all-in-one adeno-associated virus (AAV) delivery. We hypothesized that the multiplex capabilities of the Cpf1 system could help minimize oxalate formation in PH1 by simultaneously targeting the hepatic hydr

Clinical and Metabolic Correlates of Calcium Oxalate Stone Subtypes: Implications for Etiology and Management.

Introduction: Calcium oxalate (CaOx) is the predominate component within renal calculi and can be divided into two subtypes: CaOx-monohydrate (COM) and CaOx-dihydrate (COD). COM and COD form in differing urinary environments, which suggest differential underlying metabolic abnormalities associated with each subtype. We compared clinical and metabolic findings in CaOx stone formers to delineate factors differentiating COD and COM stone formers and the implication this holds in terms of etiology and treatment. Patients and Methods: We identified CaOx stone formers that had passed their stones or

Case report of oxalate nephropathy in a patient with pancreatic metastases from renal carcinoma.

BACKGROUND: Patients with metastatic renal carcinoma frequently have pre-existing renal impairment and not infrequently develop worsening renal function as a complication of their treatment. The presence of pancreatic metastases in patients with metastatic renal carcinoma, often confers a more favourable prognosis and as a consequence this patient group may be exposed to such treatments for more prolonged periods of time. However, the development of renal failure may also be a consequence of the cancer itself rather than its treatment. CASE PRESENTATION: We present an 84-year-old patient rece

The influence of metal centres on the exchange interaction in heterometallic complexes with oxalate-bridged cations.

The reaction of bis(phenanthroline)metal(ii) cations (M = Mn2+, Co2+, Ni2+, Cu2+ and Zn2+) with bis(oxalato)chromium(iii) anions in a water/ethanol solution gives rise to a series of compounds with oxalate-bridged cations, [{M(phen)2}2(μ-C2O4)][Cr(phen)(C2O4)2]2·4H2O [Mn2Cr2 (1), Co2Cr2 (2), Ni2Cr2 (3), Cu2Cr2 (4) and Zn2Cr2 (5)]. Their structural analysis reveals that all the prepared compounds crystallize in the triclinic system, space group P1[combining macron], having similar unit cell parameters, molecular structures and crystal packing features. All metal centres in 1-5 are octahedrally
