Salicylic acid-induced changes in physiological parameters and genes of the flavonoid biosynthesis pathway in Artemisia vulgaris and Dendranthema nankingense during aphid feeding.

Phloem-feeding aphids cause serious damage to plants. The mechanisms of plant-aphid interactions are only partially understood and involve multiple pathways, including phytohormones. In order to investigate whether salicylic acid (SA) is involved and how it plays a part in the defense response to the aphid Macrosiphoniella sanbourni, physiological changes and gene expression profiles in response to aphid inoculation with or without SA pretreatment were compared between the aphid-resistant Artemisia vulgaris 'Variegata' and the susceptible chrysanthemum, Dendranthema nankingense. Changes in lev…

Ectopic expression of foxtail millet zip-like gene, SiPf40, in transgenic rice plants causes a pleiotropic phenotype affecting tillering, vascular distribution and root development.

Plant architecture determines grain production in rice (Oryza sativa) and is affected by important agronomic traits such as tillering, plant … tecture is modified by the promotion of lateral root development, and the distribution of xylem and phloem in the vascular bundle is affected. Analysis of hormone levels showed that the ratios of indole-3-acetic acid/zeatin (IAA/ZR) and IAA/gibberellic acid (IAA/GA) decreased in SiPf40-transgenic plants compared with wild-type plants. These findings stro …

Elemental distribution and chemical speciation of copper and cobalt in three metallophytes from the copper-cobalt belt in Northern Zambia.

Three metallophyte species, Persicaria capitata, P. puncata (Polygonaceae), Conyza cordata (Asteraceae) from mineral wastes in the Zambian c … ions of Cu-Co are in the epidermal cells, whereas in Persicaria puncata Co was also enriched in the phloem. The Co coordination chemistry shows that an aqueous Co(ii)-tartrate complex was the predominant component identified in all plants and tissues, along with a minor component of a Co(iii) compound with oxygen donor ligands. For Cu, there was consider …

Relative Infestation Level and Sensitivity of Grapevine Cultivars to the Leafhopper Empoasca vitis (Hemiptera: Cicadellidae).

The leafhopper Empoasca vitis (Göthe) (Hemiptera: Cicadellidae) feeds on veins of grapevine leaves, mainly on the phloem, causing physiological injury, color change and drying of leaf margins, yield and sugar content reduction. The relative infestation level (i.e., the probability that a plant is attacked by herbivores) of E. vitis on different grapevine cultivars and their sensitivity (i.e., the incidence of symptoms expression in response to herbivore feeding or other stimuli) to this pest were studied over four years in two vineyards located in northeastern Italy. Some cultivars (e.g., Carm…

PDK1 regulates auxin transport and Arabidopsis vascular development through AGC1 kinase PAX.

The 3-phosphoinositide-dependent protein kinase 1 (PDK1) is a conserved master regulator of AGC kinases in eukaryotic organisms. pdk1 loss of function causes a lethal phenotype in animals and yeasts, but only mild phenotypic defects in Arabidopsis thaliana (Arabidopsis). The Arabidopsis genome contains two PDK1-encoding genes, PDK1 and PDK2. Here, we used clustered regularly interspaced short palindromic repeat (CRISPR)/CRISPR-associated protein 9 (Cas9) to generate true loss-of-function pdk1 alleles, which, when combined with pdk2 alleles, showed severe developmental defects including fused c

Exploring plant responses to aphid feeding using a full Arabidopsis microarray reveals a small number of genes with significantly altered expression.

The aim of this study was to determine which Arabidopsis thaliana (L.) genes had significantly altered expression following 2-36 h of infestation by the aphid Myzus persicae (Sulzer). Six biological replicates were performed for both control and treatment at each time point, allowing rigorous statistical analysis of any changes. Only two genes showed altered expression after 2 h (one up- and one down-regulated) while two were down-regulated and twenty three were up-regulated at 36 h. The transcript annotation allowed classification of the significantly altered genes into a number of classes, i

SlMES1 modulates methyl salicylate to influence fruit volatile profiles in tomato.

Methyl salicylate (MeSA), known as phloem-based mobile signal, has been identified as undesirable volatile compounds for tomato fruits due to its medicinal and wintergreen aroma properties. However, the response of most volatile compounds to endogenous MeSA are still unclarified. In this work, we found the concentration of MeSA can be regulated by salicylic acid methyl esterase 1 (SlMES1). We used CRISPR/Cas9 and GC-MS strategies to investigate the effect of SlMES1 on the biosynthesis of flavor compounds during tomato fruit ripening. Our results showed that the loss of function of SlMES1 signi…

A Constitutively Active Cytokinin Receptor Variant Increases Cambial Activity and Stem Growth in Poplar.

The cambial meristem is responsible for bark and wood formation in woody plants. The activity of the cambial meristem is controlled by vario … eading to increased radial stem growth. The effectivity of ROCK3 when expressed in either secondary phloem cells or in cambial cells is consistent with a dual, tissue-autonomous and non-autonomous activity of cytokinin in regulating cambial activity. We propose ROCK3 as a novel gene to enhance biomass formation in woody plants.

Evidence for Nonstomatal Uptake of Hg by Aspen and Translocation of Hg from Foliage to Tree Rings in Austrian Pine.

To determine whether trees are reliable biomonitors of air mercury (Hg) pollution concentrations were measured in bark, foliage, and tree ri … ing Hg accumulation into tree rings is by way of the stomata and subsequent translocation by way of phloem. Populus leaves and Pinus rings can be used as biomonitors for TGM concentrations over space. However, the use of trees as temporal proxies requires further investigation due to radial translocation observed in active sapwood tree rings.

Multiple phytohormone signals control the transcriptional response to soybean aphid infestation in susceptible and resistant soybean plants.

The soybean aphid (Aphis glycines) is a major phloem-feeding pest of soybean (Glycine max). A. glycines feeding can cause the diversion of photosynthates and transmission of plant viruses, resulting in significant yield losses. In this study, we used oligonucleotide microarrays to characterize the long-term transcriptional response to soybean aphid colonization of two related soybean cultivars, one with the Rag1 aphid-resistance gene and one aphid-susceptible cultivar (without Rag1). Transcriptome profiles were determined after 1 and 7 days of aphid infestation. Our results revealed a suscepti…

Effects of phosphorus availability and genetic variation of leaf terpene content and emission rate in Pinus pinaster seedlings susceptible and resistant to the pine weevil, Hylobius abietis.

We studied the effects of phosphorus fertilisation on foliar terpene concentrations and foliar volatile terpene emission rates in six half-sib families of Pinus pinaster Ait. seedlings. Half of the seedlings were resistant to attack of the pine weevil Hylobius abietis L., a generalist phloem feeder, and the remaining seedlings were susceptible to this insect. We hypothesised that P stress could modify the terpene concentration in the needles and thus lead to altered terpene emission patterns relevant to plant-insect signalling. The total concentration and emission rate ranged between 5732 and …

Interplay of auxin, KANADI and Class III HD-ZIP transcription factors in vascular tissue formation.

Class III HD-ZIP and KANADI gene family members have complementary expression patterns in the vasculature and their gain-of-function and loss-of-function mutants have complementary vascular phenotypes. This suggests that members of the two gene families are involved in the establishment of the spatial arrangement of phloem, cambium and xylem. In this study, we have investigated the role of these two gene families in vascular tissue differentiation, in particular their interactions with the plant hormone auxin. We have analyzed the vasculature of plants that have altered expression levels of Cl…

Armet, an aphid effector protein, induces pathogen resistance in plants by promoting the accumulation of salicylic acid.

Effector proteins present in aphid saliva are thought to modulate aphid-plant interactions. Armet, an effector protein, is found in the phloem sap of pea-aphid-infested plants and is indispensable for the survival of aphids on plants. However, its function in plants has not been investigated. Here, we explored the functions of Armet after delivery into plants. Examination of the transcriptomes of Nicotiana benthamiana and Medicago truncatula following transgenic expression of Armet or infiltration of the protein showed that Armet activated pathways associated with plant-pathogen interactions, …
