TaEXPB5 functions as a gene related to pollen development in thermo-sensitive male-sterility wheat with Aegilops kotschyi cytoplasm.

The thermo-sensitive cytoplasmic male-sterility line with Aegilops kotschyi cytoplasm (K-TCMS) is completely male sterile under low temperature (< 18 ℃) during Zadoks growth stages 45-52, whereas its fertility can be restored under hot temperature (≥ 20 ℃). The K-TCMS line may facilitate hybrid breeding and hybrid wheat production. Therefore, to elucidate the molecular mechanisms of its male sterility/fertility conversion, we conducted the association analysis of proteins and transcript expression to screen fertility related genes using RNA-seq, iTRAQ, and PRM-based assay. A gene encoding expa

Increased expression of the MALE STERILITY1 transcription factor gene results in temperature-sensitive male sterility in barley.

Understanding the control of fertility is critical for crop yield and breeding; this is particularly important for hybrid breeding to capitalize upon the resultant hybrid vigour. Different hybrid breeding systems have been adopted; however, these are challenging and crop specific. Mutants with environmentally reversible fertility offer valuable opportunities for hybrid breeding. The barley HvMS1 gene encodes a PHD-finger transcription factor that is expressed in the anther tapetum, which is essential for pollen development and causes complete male sterility when overexpressed in barley. This m

Breeding Utilization of the White-Grained Pre-Harvest Sprouting-Resistant Wheat Landrace Tuotuomai Through Marker-Assisted Selection.

Improving pre-harvest sprouting (PHS) resistance in white-grained wheat is challenging because reduced grain pigmentation is often associated with weaker seed dormancy. Here, we evaluated the breeding value of the white-grained, PHS-resistant landrace Tuotuomai (TTM) and developed a molecular marker for tracking TTM-derived material during breeding. Based on phenotypic segregation and sequence polymorphism analysis, a KASP marker targeting a polymorphic site in the TaMyb10-D1 region was developed. The marker co-segregated with white grain color in the TTM × CS population, and the TTM-type mark

Unlocking the future of crop improvement with crop wild relatives.

Crop wild relatives (CWRs) retain extensive genetic diversity lost during crop domestication, yet their translation into breeding outcomes remains constrained by fundamental quantitative-genetic and population-genetic bottlenecks. Here, we propose an integrative framework that evaluates CWR deployment through three interconnected genetic lenses: (i) the architecture of domestication bottlenecks and their consequences for allelic diversity, linkage disequilibrium, and selectable genetic variation in modern breeding pools; (ii) the genetic determinants of successful allele transfer, distinguishi

Decoding the autotetraploid origin of the stress-tolerant wild beet Beta corolliflora.

BACKGROUND: Most crop plants, including sugar beet (Beta vulgaris subsp. vulgaris), suffer from domestication bottlenecks and low genetic diversity caused by extensive selection for few traits. However, crop wild relatives (CWRs) harbour useful traits relevant for crop improvement, including enhanced adaptation to biotic and abiotic stresses. Especially polyploids are interesting from an evolutionary perspective as the genome undergoes reorganisation after the polyploidisation event. Through neo- and subfunctionalisation, novel gene functions emerge, which enable plants to cope with changing e

BiMSGP: A Bidirectional Mamba-Based Model for Scalable and Accurate Genomic Prediction in Plants.

Background/Objectives: Accurate prediction of complex traits from high-dimensional genomic markers remains difficult in plant breeding. Linear models have limited capacity for non-linear interactions, and many deep learning models scale poorly on long marker sequences. This study introduces BiMSGP, a bidirectional Mamba-based model for scalable genomic prediction. Methods: BiMSGP extends a selective state space model with forward and backward processing so that each marker can integrate sequence context from both directions while retaining linear computational complexity; this is a modeling de…

Engineering cold stress resilience in capsicum annuum through functional genomics and precision breeding.

This review synthesizes the molecular mechanisms of cold tolerance in pepper, integrating multi-omics data, genome editing, and precision breeding strategies to accelerate the development of cold-resilient cultivars. Cold stress is a significant environmental factor that affects the growth, productivity, and fruit quality of Capsicum annuum by impairing membrane integrity photosynthesis and cellular redox homeostasis. Although pepper has several endogenous cold-responsive regulators such as CaNAC035 and CabHLH035, along with antioxidant defense systems, its cold tolerance remains limited due to

Effects of a 28-day feeding trial of grain-containing versus pulse-based diets on cardiac function, taurine levels and digestibility in domestic dogs.

In 2018, the US Food and Drug Administration reported a link between canine dilated cardiomyopathy (DCM) and grain-free diets. Evidence to support a link has emerged, but the specific ingredients responsible and the role of taurine or other causative factors remain unclear. We hypothesized dogs fed pulse-based, grain-free diets for 28 days will show decreased macronutrient digestibility, increased fecal bile acid excretion, and reduced plasma cystine, cysteine, methionine and taurine, causing sub-clinical cardiac or blood changes indicative of early DCM. Three diets were formulated using white

New PCR-specific markers for pollen fertility restoration QRfp-4R in rye (Secale cereale L.) with Pampa sterilizing cytoplasm.

Pampa cytoplasmic male sterility phenomenon is used extensively in the rye hybrid breeding programs. It relies on sterilizing action of the cytoplasm resulting in non-viable pollen of female lines. The sterilizing effect is problematic for reversion, and efficient restores are needed. The most promising QTL is located on chromosome 4R, but other chromosomes may also code the trait. Advanced recombinant inbred lines formed bi-parental mapping population genotyped with DArTseq markers. Genetic mapping allowed the seven linkage groups to construct with numerous markers and represent all rye chrom

Domestication of aromatic medicinal plants in Mexico: Agastache (Lamiaceae)-an ethnobotanical, morpho-physiological, and phytochemical analysis.

BACKGROUND: Most reports of domesticated plants that involve a domestication gradient or inter-specific hybridization in Mexico have focused on those used as food. This study provides knowledge about these processes in two aromatic medicinal plants, Agastache mexicana (Lamiaceae) and A. m. subsp. xolocotziana, widely used in Mexican traditional medicine for the treatment of gastrointestinal ailments and for their sedative effect. Different populations of A. mexicana along a gradient of domestication are found in the foothills of the Popocatepetl volcano of central Mexico, while in this same ar

Disease resistance in maize and the role of molecular breeding in defending against global threat.

Diseases are a potential threat to global food security but plants have evolved an extensive array of methodologies to cope with the invading pathogens. Non-host resistance and quantitative resistance are broad spectrum forms of resistance, and all kinds of resistances are controlled by extremely diverse genes called "R-genes". R-genes follow different mechanisms to defend plants and PAMP-induced defenses in susceptible host plants are referred to as basal resistance. Genetic and phenotypic diversity are vital in maize (Zea mays L.); as such, genome wide association study (GWAS) along with cer

Phenotypic Characterization and Transcriptome Analysis of the Dwarf Mutant zmbrd1 in Maize.

Background: Maize (Zea mays L.) is a vital global crop, and yield improvement through dwarfing breeding-inspired by the Green Revolution-holds promise for addressing food security challenges. Despite the identification of over 60 dwarf genes in maize, their genetic diversity remains limited. Brassinosteroids (BRs) are key phytohormones that regulate plant height, and mutations in BR-related genes often result in dwarf phenotypes. Methods: The zmbrd1 mutant was generated via EMS mutagenesis in the B73 background. Phenotypic traits (plant height, root length) and histological features (e.g., mes

SlJMJ12 overexpression confers improved cadmium tolerance in Arabidopsis and tomato.

Cadmium (Cd) is a toxic heavy metal threatening ecosystems and crop production. The functions of histone demethylases in plant Cd tolerance remain poorly understood. We functionally characterized the tomato histone demethylase SlJMJ12 using Arabidopsis and tomato. Cd rapidly induces leaf-expressed SlJMJ12, with peak expression at 3 h post-treatment. Compared with the wild type, SlJMJ12 loss-of-function mutants show Cd sensitivity in both Arabidopsis and tomato, with reduced biomass, higher plant Cd and ROS levels, and decreased antioxidant enzyme activities, whereas overexpression improves Cd

Infrared Microimaging of Sucrose Distribution in Plant Vascular Tissues.

Sucrose is the primary transport sugar in plants, serving as an essential energy source and signaling molecule. Detection, visualization, and quantification of sucrose in various plant tissues are essential for understanding the metabolic and physiological processes that sustain plant life. Traditional metabolite-mapping techniques have struggled to visualize the quantitative distribution of sucrose at sufficient resolution to distinguish vascular bundles from surrounding tissues. Here, we present a Fourier-transform infrared (FTIR) imaging approach that can visualize sucrose in plant tissues

Genetic Dissection of Sorghum Dwarfism Through Systematic Screening of Dw1-Dw3 Alleles in Chinese Germplasm.

Sorghum dwarfing genes (Dw1, Dw2, Dw3) are crucial determinants of plant architecture and yield potential; however, their genetic characteristics and distribution patterns in Chinese sorghum landraces remain poorly understood. This study systematically analyzed their allelic distribution across 241 Chinese landrace accessions. Through rigorous PCR-based genotyping and detailed phenotypic characterization, we identified that only approximately 7% of the surveyed landraces carried natural dwarfing alleles, with mutations in the dw3 locus being the most frequently observed. Plant height statistic

A mutation in LacDWARF1 results in a GA-deficient dwarf phenotype in sponge gourd (Luffa acutangula).

A dwarfism gene LacDWARF1 was mapped by combined BSA-Seq and comparative genomics analyses to a 65.4 kb physical genomic region on chromosome 05. Dwarf architecture is one of the most important traits utilized in Cucurbitaceae breeding because it saves labor and increases the harvest index. To our knowledge, there has been no prior research about dwarfism in the sponge gourd. This study reports the first dwarf mutant WJ209 with a decrease in cell size and internodes. A genetic analysis revealed that the mutant phenotype was controlled by a single recessive gene, which is designated Lacdwarf1 (

Identification and mapping of a new recessive dwarfing gene dw9 on the 6RL rye chromosome and its phenotypic effects.

The introduction of high-yielding semi-dwarf varieties of wheat into cultivation has led to a "green revolution." This has required intensive research into various sources of dwarfism in wheat. However, there has been very little advancement in research on dwarfing genes in rye in comparison to wheat or barley. So far, three dominant dwarfing genes (Ddw1, Ddw3, and Ddw4) and three recessive genes (ct1, ct2, and np) have been characterized and precisely mapped in rye. There is no complete catalog of dwarfing genes available in rye. This paper presents an identification of the source of dwarfism
