Four species of malaria parasites are known to infect humans. A fifth species, Plasmodium knowlesi, has been reported to infect humans in Malaysian Borneo. Here we report for the first time the incrimination of Anopheles latens as the vector of P. knowlesi among humans and monkeys in Sarawak, Malaysia.

OBJECTIVE: To confirm the diagnosis of a human case with atypical vivax-malaria from Yunnan Province by molecular technique.
METHODS: DNA was extracted from blood films of unidentified sample, and of four known Plasmodium species (P. vivax, P. falciparum, P. knowlesi, and P. cynomolgi). A DNA-based diagnosis with the polymerase chain reaction (PCR) method targeting the small subunit ribosomal RNA (SSU rRNA) genes of genus- and species-specific (two human malaria species and P. knowlesi) was introduced.
RESULTS: The PCR amplification with primer pair specific for P. knowlesi produced a single

BACKGROUND: About a fifth of malaria cases in 1999 for the Kapit division of Malaysian Borneo had routinely been identified by microscopy as Plasmodium malariae, although these infections appeared atypical and a nested PCR assay failed to identify P malariae DNA. We aimed to investigate whether such infections could be attributable to a variant form of P malariae or a newly emergent Plasmodium species.
METHODS: We took blood samples from 208 people with malaria in the Kapit division between March, 2000, and November, 2002. The small subunit ribosomal RNA and the circumsporozoite protein genes

The development of erythrocytic stages of Plasmodium knowlesi separated from their host cells has been determined in terms of the capacity of the isolated organisms to carry out the synthesis and secretion of proteins. P. knowlesi trophozoites and schizonts were released from host cells by nitrogen decompression and cultivated in a medium consisting of 20 mM Na+; 120 mM K+; 1 mM Mg2+; no Ca2+; 100 mM Cl-; 20 mM HCO3-; 5 mM Hepes [pH 6.73], glucose, vitamins, amino acids and 10% fetal calf serum. The yield was about 97% intact parasites, judging by their ability to maintain a membrane potential…

In this report, we demonstrate that the variable antigens present on the surface of Plasmodium knowlesi-infected erythrocytes could not be found on the surface of merozoites. A number of technical problems had to be solved to make such a comparative study possible, including the purification of merozoites by affinity columns and gradient centrifugation, the use of hyperimmune rabbit sera instead of monkey sera, and the use of immunocytological methods (indirect immunofluorescence antibody test and electron microscopy with ferritin-labeled antibodies) instead of the schizont-infected cell agglu…

Material from various steps obtained in the French pressure cell technic of preparing antigen from Plasmodium knowlesi-infected red cells, was examined by elctron microscopy. A positively charged colloidal iron solution was used to differentiate between membranes of host red cells and parasites. Red cell membranes take the stain, wheras parasite membranes do not. This antigen which has been used previously to protect monkeys against P. knowlesi appears to consist almost entirely of membrane-bounded vesicles. Some of these vesicles contain a fine granular material, whereas others appear empty. …

RNA extracted from purified parasites of Plasmodium knowlesi was fractionated using agarose gel electrophoresis. Preparations from para sites grown both in vivo and in vitro contained species of RNA with sedimentation coefficients of 4-0S, 5-0S, 16-6S, 24-2S, 31-4S, 38-0S and 48-3S. There was less RNA present in parasites grown in vitro than the equivalent stage parasites grown in vivo but the proportional amounts of the various species of RNA was similar in both cases. It is suggested that the 24-2S and 16-6S species of RNA are ribosomal and that the high molecular weight 31-4S, 38-0S and 48-…

Experiments were performed on healthy (non-infected) and Plasmodium knowlesi-infected rhesus monkeys (Macaca mulatta) to determine the activity of the kinin-destroying enzymes (kininases) present in the circulation. Methods for the studies and estimation of kininases in the circulating blood of control and P. knowlesi-infected animals are described. The activity of the enzymes, present in the blood in all animals investigated, was evaluated from both methods employed by incubation with synthetic bradykinin. The isolated rat uterus served as the test organ. Studies were made on protein peak fra…

After publication of the article [1], it has been brought to our attention that two of the labels on Figure 4 have transposed. The labels "S-type SSU rRNA" and "A-type SSU rRNA" should be in opposite places.

Malaria is a global public health concern, where Plasmodium knowlesi contributes to most human malaria cases in Malaysia. The leaves of Vernonia amygdalina (Delile) were found to exhibit significant anti-plasmodial properties and are commonly used in sub-Saharan Africa countries for malaria treatment. Nonetheless, studies are lacking in the effect of this plant against P. knowlesi specifically. This study aimed to investigate the in vitro anti-plasmodial activity of crude extracts from V. amygdalina (Delile) against P. knowlesi. The fresh leaves of V. amygdalina were sequentially extracted in …

Plasmodium knowlesi has been recently identified as the "fifth human malaria species" following the discovery in Malaysian Borneo of a large focus of this simian malaria parasite in humans. Even though it shares microscopic similarities with Plasmodium malariae, it may cause severe illness with risk of fatality. We describe a case of P. knowlesi infection causing multi-organ failure in a patient who was successfully managed due to early recognition of the infection. Clinicians in this region should be more aware of the infection as it is not as rare as previously thought. This case write up hi…

We report seven cases of naturally acquired human Plasmodium knowlesi infections which were admitted to our centre from July 2007 till June 2008. Diagnosis was confirmed by nested PCR. Cases of P. knowlesi infections, dubbed the fifth type of human malaria, have been reported in East Malaysia (Sabah and Sarawak) as well as in the state of Pahang in Peninsula Malaysia. These seven patients appear to be the first few reported cases of P. knowlesi infection in the Klang valley, Peninsula Malaysia. We then discuss the characteristics of human P. knowlesi infections, which include its natural hosts…

Eighteen Rhesus monkeys were immunized with a fresh or lyophilized antigen preparation obtained from erythrocytes infected with Plasmodium knowlesi. Sixteen of these monkeys showed a positive delayed hypersensitivity response when subsequently skin tested with the immunizing antigen. Ten of the 16 positive reactors survived a P, knowlesi challenge, the remaining 6 showed several indications of protection. None of the 18 control monkeys gave a positive skin reaction or survived the challenging inoculum. Macro- and microscopical evidence suggests that the dermal reaction is of a delayed type. Pr…

Accurate molecular tools are essential for estimating zoonotic malaria transmission in Southeast Asia. This study applied ultrasensitive reverse-transcriptase real-time polymerase chain reaction (PCR) to detect zoonotic malaria in febrile patients from health facilities across three mainland districts (Batubara, Tanjung Balai, and Central Tapanuli) and separately on Mursala Island, North Sumatra, Indonesia. Among 64 participants on Mursala, 7 (10.9%) adults had Plasmodium knowlesi infections (including 5 agricultural workers and 2 adults with severe WHO anemia), and 2 (3.1%) adults had Plasmod…

BACKGROUND: Plasmodium knowlesi is the most common cause of malaria in Malaysia. However, microscopic diagnosis is inaccurate and rapid diagnostic tests (RDTs) are insufficiently sensitive. PCR is sensitive and specific but not feasible at a district level. Loop-mediated isothermal amplification (LAMP) shows potential with only basic requirements. A commercially available LAMP assay, the Eiken Loopamp™ MALARIA Pan Detection kit, is sensitive for Plasmodium falciparum and Plasmodium vivax, but has not previously been evaluated for P. knowlesi. This study aims to determine the sensitivity of thi…

Paracetamol may improve renal function in patients with severe Plasmodium knowlesi malaria, particularly in those with acute kidney injury and hemolysis, via inhibition of cell-free hemoglobin mediated oxidative kidney damage. We developed a population pharmacokinetic/pharmacodynamic (PK/PD) model to assess effects of paracetamol on creatinine, hepatotoxicity, fever clearance, and parasite clearance among Malaysian patients with predominantly non-severe knowlesi malaria using data from the PACKNOW trial (Clinical Trials Registration: NCT03056391). A total of 372 patients were included in the P…

Although Malaysia is considered free of human malaria, there has been a growing number of Plasmodium knowlesi cases. This alarming trend highlighted the need for our understanding of this parasite and its associated vectors, especially considering the role of genetic diversity in the adaptation and evolution among vectors in endemic areas, which is currently a significant knowledge gap in their fundamental biology. Thus, this study aimed to investigate the genetic diversity of Anopheles balabacensis, Anopheles cracens, Anopheles introlatus, and Anopheles latens-the vectors for P. knowlesi mala…

A 55-year-old man sought treatment for an uncomplicated febrile illness after returning to Canada from the Philippines. A suspected diagnosis of Plasmodium knowlesi infection was confirmed by PCR, and treatment with atovaquone/proguanil brought successful recovery. We review the evolving epidemiology of P. knowlesi malaria in the Philippines, specifically within Palawan Island.

BACKGROUND: Malaria parasites such as Plasmodium knowlesi, P. inui, and P. cynomolgi are spread from macaques to humans through the Leucosphyrus Group of Anopheles mosquitoes. It is crucial to know the distribution of these vectors to implement effective control measures for malaria elimination. Plasmodium knowlesi is the most predominant zoonotic malaria parasite infecting humans in Malaysia.
METHODS: Vector data from various sources were used to create distribution maps from 1957 to 2021. A predictive statistical model utilizing logistic regression was developed using significant environmen…
