An emerging role of Prevotella histicola on estrogen deficiency-induced bone loss through the gut microbiota-bone axis in postmenopausal women and in ovariectomized mice.

BACKGROUND: The gut microbiota (GM)-bone axis has emerged as a crucial mediator of bone homeostasis. Estrogen deficiency-induced bone loss is closely associated with an altered GM. However, the underlying mechanisms remain unclear. OBJECTIVES: We sought to explore the putative effects of GM on estrogen deficiency-induced bone loss and determine a potential mechanism. METHODS: Fecal samples collected from postmenopausal women with osteoporosis (PMO) and with normal bone mass (PMN) were examined by 16S ribosomal RNA (rRNA) gene sequencing and analysis. Prevotella histicola, a typical species o…

Prevotella copri exhausts intrinsic indole-3-pyruvic acid in the host to promote breast cancer progression: inactivation of AMPK via UHRF1-mediated negative regulation.

Emerging evidence has revealed the novel role of gut microbiota in the development of cancer. The characteristics of function and composition in the gut microbiota of patients with breast cancer patients has been reported, however the detailed causation between gut microbiota and breast cancer remains uncertain. In the present study, 16S rRNA sequencing revealed that Prevotella, particularly the dominant species Prevotella copri, is significantly enriched and prevalent in gut microbiota of breast cancer patients. Prior-oral administration of P. copri could promote breast cancer growth in speci…

Supernatants from DNase-deficient Prevotella intermedia strains enhance oral squamous cell carcinoma cell migration and invasion by activating inflammatory and epithelial-mesenchymal transition pathways.

INTRODUCTIONS: Oral squamous cell carcinoma (OSCC) has been increasingly associated with dysbiosis of the oral microbiome. Among oral pathogens, Prevotella intermedia (P. intermedia) is frequently enriched in patients with OSCC; however, the role of its virulence factors-particularly its deoxyribonuclease (DNase) activity-remains poorly understood. METHODS: We compared the effects of culture supernatants from wild-type P. intermedia OMA14 and DNase-deficient mutant strains (nucA, nucD and nucA nucD) on the migration and invasion of the OSCC cell line SAS, using wound healing and Matrigel inva…

Salivary Prevotella qPCR Signal as an Exploratory Non-Invasive Adjunct for Rotterdam Phenotype Stratification in Women with Polycystic Ovary Syndrome: A Proof-of-Concept Cross-Sectional Study.

Background/Objectives: Polycystic ovary syndrome (PCOS) is a heterogeneous endocrine-metabolic disorder. Rotterdam-defined phenotypes may need adjunctive, non-invasive signals for research-level phenotypic stratification. Saliva deserves attention because it is easy to collect repeatedly, acceptable in outpatient settings, and may reflect oral-systemic inflammatory and endocrine-metabolic interactions. This study evaluated whether salivary qPCR signals for Lactobacillus, Prevotella, and Bifidobacterium differ across Rotterdam-defined PCOS phenotypes and controls, with emphasis on the explorato…

Enzymatic tools for the treatment of caries-associated biofilms: Investigating the enzymes of a putative polysaccharide utilization locus from Prevotella melaninogenica and their application against Streptococcus mutans biofilm.

We identified a potential cluster of genes in the organism Prevotella melaninogenica which was predicted to represent a polysaccharide utilization locus (PUL). Based on the knowledge that PUL glycoside hydrolase (GH) enzymes are frequently co-expressed to saccharify a complex polysaccharide, and that one of the enzymes (PmGH87) is known to hydrolyse a major linkage of the extracellular polymeric substance (EPS) of cariogenic biofilms, we hypothesized that the predicted PUL (pPmPUL) GHs may be relevant to the degradation of biofilms associated with oral disease. To test this, the pPmPUL GHs wer…
