It has been suggested that elevated luteal phase prolactin (PRL) levels may have an important role in causing some of the symptoms of the premenstrual tension syndrome (PMTS). Thirty-seven women suffering from severe premenstrual dysphoria were selected for this study. During the last four years 12 Prolactin (PRL) secreting adenomas whose first clinical manifestation appeared before the age of 15 years have been studied. The first signs were a decreased growth rate and arrest of pubertal development in 4 girls and 2 boys in whom the adenoma occurred before or during puberty. To clarify the controversy about the effect of prolactin (PRL) on dehydroepiandrosterone sulfate (DHEA-S), this study was undertaken to investigate the effects of alterations in plasma PRL on plasma DHEA-S concentrations in hyperprolactinemic women, as well as in normal male subjects. The plasma concentrations of prolactin and nortriptyline were determined in 17 patients with endogenous depression treated for 4 weeks with nortriptyline once a day. None of the patients developed galactorrhoea. A moderate but statistically significant increase (about 25%) in the plasma prolactin was found after 3 weeks of treatment. There was no correlation between the plasma levels of prolactin and of nortriptyline. The results indicate that nortriptyline does not belong to the group of psychotropics which produce considerable increases in the plasma concentration of prolactin.
