Protein S deficiency in men with long-term human immunodeficiency virus infection.

Decreases in protein S levels have recently been reported in some human immunodeficiency virus (HIV)-infected patients. To examine predisposing factors, 25 men randomly selected from a long-term study of HIV-infected patients were studied. The minimum mean duration of HIV seropositivity in this group was 106.6 months (range 15 to 143 months). No patients were anticoagulated at the time of the study. Three of the 25 randomly selected patients gave a history of thrombosis, in each instance occurring after the onset of HIV positivity. Two of the 3 patients with thrombosis had more than one episod

Juvenile chronic myeloid leukemia. A malignancy of S-100 protein-positive histiocytes.

Three cases of juvenile chronic myeloid leukemia (JCML) are reported. The patients were aged 3-4.5 years and presented with generalized lymphadenopathy, hepatosplenomegaly, anemia, thrombocytopenia, elevated white blood cell count with monocytosis, and high fetal hemoglobin level. Philadelphia chromosome was absent in two cases studied. The bone marrow showed myeloid hyperplasia with increased monocytoid cells and blasts. Biopsy or postmortem material available in two cases revealed malignant infiltration of lymph nodes, liver, spleen, lungs, intestines, and skin. The neoplastic cells ranged f

Primary eosinophilic granuloma of frontal lobe. Diagnostic use of S-100 protein.

Solitary eosinophilic granuloma of the frontal lobe of the brain is a rare lesion. A 30-year-old Jordanian man with a history of recurrent bilateral chronic middle ear infections and tuberculosis presented with focal seizures. A solitary right frontotemporal mass was diagnosed by brain biopsy as an eosinophilic granuloma. Immunoperoxidase stains of virtually all histiocytes in the lesion were strongly positive for S-100. Four cases of eosinophilic granuloma and one of Letterer-Siwe disease were also examined, and virtually all histiocytes demonstrated strong S-100 positivity. In comparison, 24

Circulating antigens in schistosomiasis: detection of 31/32-kDa proteins in sera from patients infected with Schistosoma japonicum, S. mansoni, S. haematobium, or S. intercalatum.

A sandwich enzyme-linked immunosorbent assay (ELISA) was developed to detect 31/32-kDa schistosome proteins as circulating antigens in sera from schistosomiasis patients. A monoclonal antibody was used as a capture antibody and rabbit antiserum raised against purified 31/32-kDa proteins was the detecting antibody. Positive results were obtained with patients infected with Schistosoma japonicum (88%; n = 69), S. mansoni (80%; n = 56), S. haematobium (100%; n = 40), or S. intercalatum (94%; n = 65). Sera from uninfected Chinese and African individuals and from Chinese patients with trichinosis,

Reversible S-palmitoylation of C4 protein encoded by TYLCCxV orchestrates geminiviral pathogenesis.

Cysteine palmitoylation (S-palmitoylation or S-acylation) is a reversible post-translational modification dynamically controlled by opposing enzymes: palmitoyl acyltransferases (PATs) and depalmitoylases. Despite its established roles in other systems, the mechanistic details of S-acylation in plants, particularly its spatiotemporal regulation during plant-virus interactions, remain poorly understood, largely due to the lack of validated enzyme-substrate pairs. Using the geminivirus tomato yellow leaf curl Chuxiong virus (TYLCCxV) as a model, we show that the viral effector C4 undergoes S-palm

S-nitrosylation of the scaffold protein STRAP enhances oxidative stress-induced apoptosis.

Serine-threonine kinase receptor-associated protein (STRAP) functions as a negative regulator of apoptosis by inhibiting apoptosis signal-regulating kinase 1 (ASK1) activity. STRAP is consistently present in the inducible nitric oxide synthase (iNOS) interactome and contains two essential cysteine residues, Cys152 and Cys270, which are required for its interaction with ASK1. However, the role of the STRAP-iNOS interaction remains unclear. In this study, we found that STRAP specifically interacts with iNOS but not with endothelial NOS or neuronal NOS. iNOS mediates the S-nitrosylation of STRAP,

Hydrogen sulfide-mediated protein s-persulfidation: a regulatory mechanism in cardiovascular homeostasis.

BACKGROUND: Cardiovascular diseases represent a primary cause of global mortality. Hydrogen sulfide operates as a major endogenous gasotransmitter that contributes to cardiovascular homeostasis through multiple biochemical mechanisms, among which protein S-persulfidation, represents a major redox-sensitive post-translational mechanism. This dynamically reversible modification converts cysteine sulfhydryl groups into highly nucleophilic persulfide groups to prevent irreversible oxidative damage. AIM OF REVIEW: This review aims to systematically synthesize the current molecular understanding of

H2S-mediated protein S-sulfhydration: a novel regulatory module in lipid metabolism.

H2S-mediated protein S-sulfhydration is an emerging post-translational modification that regulates key biological processes via regulating enzyme activities, controlling protein-protein interactions, and modulating signal transduction. Lipid metabolism represents an important target of S-sulfhydration-mediated regulation, which fine-tunes lipid metabolic networks including fatty acid turnover, triglyceride metabolism, and cholesterol homeostasis. This review aims to systematically summarize current knowledge on the regulation of lipid metabolism with a focus on S-sulfhydration, and highlight n

Protein S-acylation dynamics provide metabolic plasticity to acute myeloid leukemia cells.

Though cancer cells' altered metabolism has been recognized for a century, the clinical success of metabolic targeting remains limited due to metabolic plasticity. Here, we use acute myeloid leukemia (AML) as a model to investigate this adaptability through combinatorial metabolic compound screening. Synthetic lethality emerged when AML cells were simultaneously treated with a glutaminase inhibitor and TOFA, a hypolipidemic agent. Sensitivity to this combination was also seen in primary patient samples and in other cancer types, while healthy hematopoietic progenitors were not affected. Unexpe

Protein S-nitrosylation in the heart: Mechanisms, physiological roles, and pathological remodeling.

Protein S-nitrosylation (SNO), a reversible nitric oxide (NO)-dependent post-translational modification to reactive cysteine residues, finely tunes cardiac excitation-contraction coupling, mitochondrial function, and stress responses. In cardiomyocytes, compartmentalized NO signaling via neuronal (nNOS), endothelial (eNOS), and inducible (iNOS) isoforms regulates SNO of key proteins (e.g., RyR2, LTCC, SERCA2a, mitochondrial Complex I). Under physiological conditions, SNO modulates Ca2+ handling, myofilament sensitivity, and bioenergetics, while pathological SNO dysregulation, hypo-nitrosylatio

PROS1 Cys475Tyr mutation associated with a type III protein S deficiency pattern.

A 35-year-old woman presented with a deep vein thrombosis (DVT) and was diagnosed with protein S deficiency. She was treated with parenteral anticoagulation before switching to a direct oral anticoagulant. She presented to our practice 5 years later, where testing was repeated, demonstrating a type III deficiency pattern and a variant of uncertain significance in PROS1: c.1424G>A; p.Cys475Tyr. This cysteine residue participates in a disulfide bond in the sex hormone-binding globulin-like domain, which interacts with C4b-binding protein and is involved in several antithrombotic roles of protein

Internal thoracic artery aneurysm coincidental with protein S deficiency: a case report.

BACKGROUND: Internal thoracic artery aneurysms are exceptionally rare and carry a risk of fatal rupture. Although pseudoaneurysms are commonly associated with trauma or surgical interventions, the development of true aneurysms promoted by thrombotic disorders has not been previously reported. CASE PRESENTATION: A 61-year-old man undergoing hemodialysis for end-stage renal disease due to untreated nephrotic syndrome presented with hemoptysis. He had a medical history of deep vein thrombosis and was taking warfarin. Imaging studies revealed pulmonary embolism, systemic collateral circulation, a

[PROSI Mutation With Clinical Heterogeneity in Protein S Deficiency:Report of One Case].

Reduced protein S activity is one of the high-risk factors for venous thromboembolism.Hereditary protein S deficiency is an autosomal dominant disorder caused by mutations in the PROS1 gene.We reported a female patient with a mutation of c.292 G>T in exon 3 of the PROS1 gene, which was identified by sequencing. The genealogical analysis revealed that the mutation probably originated from the patient's mother. After searching against the PROS1 gene mutation database and the relevant literature,we confirmed that this mutation was reported for the first time internationally.

A Vaccine of SARS-CoV-2 S Protein RBD Induces Protective Immunity.

The pandemic of the novel severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has posed great threat to the world in many aspects. There is an urgent requirement for an effective preventive vaccine. The receptor binding domain (RBD), located on the spike (S) gene, is responsible for binding to the angiotensin-converting enzyme 2 (ACE2) receptor of host cells. The RBD protein is an effective and safe antigen candidate. The six-helix bundle (6HB) "molecular clamp" is a novel thermally-stable trimerization domain derived from a human immunodeficiency virus (HIV) gp41 protein segment. We

Direct Zinc Finger Protein Persulfidation by H2 S Is Facilitated by Zn2.

H2 S is a gaseous signaling molecule that modifies cysteine residues in proteins to form persulfides (P-SSH). One family of proteins modified by H2 S are zinc finger (ZF) proteins, which contain multiple zinc-coordinating cysteine residues. Herein, we report the reactivity of H2 S with a ZF protein called tristetraprolin (TTP). Rapid persulfidation leading to complete thiol oxidation of TTP mediated by H2 S was observed by low-temperature ESI-MS and fluorescence spectroscopy. Persulfidation of TTP required O2  , which reacts with H2 S to form superoxide, as detected by ESI-MS, a hydroethidine

Investigation of Glutathione S-Transferase Isoenzyme Protein Expression in Patients With Pterygium.

PURPOSE: We investigated glutathione S-transferase (GST) enzymes in terms of their potential effects on the pathogenesis of pterygium. METHODS: Twenty-six pterygium specimens and 15 normal conjunctival specimens of 15 control subjects were investigated. Expressions of GST (alpha, mu, pi, and theta) enzymes were assessed by immunohistochemical staining. A brown color in the cytoplasm and/or nuclei of epithelial cells was evaluated as positive staining for GST enzymes. For each antibody, the intensity of the reaction [negative (-), weak (1+), moderate (2+), or strong (3+)] was determined to des

Coronary artery bypass grafting in 2 thrombophilic patients with protein s deficiency.

In this report, we review 2 cases of coronary revascularization in patients with a diagnosis of coronary artery disease and preoperative protein S deficiency, an established hypercoagulable condition. In an attempt to normalize protein S levels, fresh frozen plasma was used as the priming fluid for the cardiopulmonary bypass circuit before the initiation of extracorporeal circulation. On the basis of a low risk of bleeding and the theoretical risk of thrombosis, neither patient received intraoperative antifibrinolytic treatment nor did they develop perioperative thrombotic complications.

Multiple abdominal veins thrombosis secondary to protein s deficiency - a case report.

Abdominal venous thrombosis may present either as Budd-Chiari syndrome (BCS) caused by hepatic vein or proximal inferior vena cava (IVC) obstruction or as an extra hepatic portal obstruction (EHPVO) caused by Portal vein thrombosis or mesenteric vein thrombosis, but a mixed involvement is uncommon. Multiple abdominal venous obstructions presenting with thrombosis of hepatic vein, IVC, portal vein and renal vein are very rarely seen . We are reporting a rare case with thrombosis of IVC, hepatic vein, portal vein and renal vein, with protein S and protein C deficiencies, which was managed by giv

[A case of portal vein thrombosis associated with congenital protein S deficiency].

A 25-year-old man was admitted to our hospital because of abdominal pain, nausea and low-grade fever. An abdominal CT showed remarkable thickening of the wall of the small intestine and extensive thrombosis of the mesenteric, portal and splenic veins. Because neither intestinal infarction nor peritonitis was seen, anticoagulation therapy was chosen. Heparin was administered intravenously and was used alternatively with warfarin later. The patient's symptoms and clinical data improved gradually. Concerning the etiological factors of the thrombosis, only protein S activity definitely decreased.
