Amyotrophic lateral sclerosis (ALS) is characterized by degeneration of the upper motor neurons in the frontal cortex and the motor neurons of the brain stem and spinal cord. Cranial nerve nuclei III, IV, VI are usually normal histopathologically, as are the eye movements clinically. Using electro-oculography, we found decreased saccadic or smooth pursuit velocities in four of ten patients unidirectional saccadic pursuit in one of these four, and progressively decreasing saccadic velocities in another. We believe the defect is supranuclear and that the ocular motor system is more often abnorma

The clinical observation and assessment of extra-ocular movements is common practice in assessing neurodegenerative disorders but remains observer-dependent. In the present study, we propose an algorithm that can automatically identify saccades, fixation, smooth pursuit, and blinks using a non-invasive eye tracker. Subsequently, response-to-stimuli-derived interpretable features were elicited that objectively and quantitatively assess patient behaviors. The cohort analysis encompasses persons with mild cognitive impairment (MCI), Alzheimer's disease (AD), Parkinson's disease (PD), Parkinson's

AIM: Visual impairment is present in almost all patients with ataxia telangiectasia (AT) and, due to their early onset, constitute an important disabling aspect of the syndrome: the quality of vision is limited by dyspraxia and oculomotor abnormal movements. The purpose of this observational study was to describe visual disorders, notably oculomotor impairment, in a sample of children with AT.
METHODS: Fifteen AT patients (mean age 12 years and 4 months) underwent a neurovisual evaluation, particularly focused on oculomotor functions (fixation, smooth pursuit, saccades, and abnormal ocular mo

Acquired supranuclear ocular motor paresis (ASOMP) is a rare complication following cardiac surgery, characterized by limited voluntary eye motility. The exact cause and development of ASOMP after cardiac surgery remain unclear. We report a case of ASOMP with paracentral bitemporal hemianopsia with optic atrophy after cardiac surgery, which, to our knowledge, is novel. The patient demonstrated bilateral ophthalmoplegia, with gradual improvement in voluntary smooth pursuit but persistent impairment in saccadic eye movements. Interestingly, the patient showed improved proprioceptive-driven pursu

The oculomotor abnormalities with isolated infarction of the cerebellar tonsil are unknown. In a patient with acute infarction of the right tonsil, we found (1) nearly completely abolished ipsilateral smooth pursuit and impaired contralateral pursuit, (2) a low-amplitude ipsilesional right-beating nystagmus without fixation, (3) gaze-holding deficits, and (4) normal vestibulo-ocular reflex. These findings contrast with striking vestibular abnormalities reported with unilateral flocculus and anterior tonsil infarction. Taken together, these findings allow more diagnostic certainty in cerebellar

The smooth-pursuit system moves the eyes in space accurately to track slowly moving objects of interest despite visual inputs from the moving background and/or vestibular inputs during head movements. Recently, our laboratory has shown that young primates exhibit asymmetric eye movements during vertical pursuit across a textured background; upward eye velocity gain is reduced. To further understand the nature of this asymmetry, we performed three series of experiments in young monkeys. In Experiment 1, we examined whether this asymmetry was due to an un-compensated downward optokinetic reflex

In smooth-pursuit eye movements (SPEM) with gain close to one, SPEM should be controlled mainly by prediction of target motion because retinal slip is nearly zero. We investigated the neural mechanisms of visual-target prediction by the three fMRI experiments. (1) Overt pursuit task: subjects pursued a sinusoidally moving target which blinked (blink condition) or did not blink (continuous condition). (2) Covert pursuit task: subjects covertly pursued the same target with eyes gazed at fixation point. (3) Attend-to-stationary target task: subjects brought attention on a stationary target with e

BACKGROUND: Thyroid-associated ophthalmopathy (TAO) is an autoimmune orbital disease that may cause ocular motility abnormalities, altered periocular appearance, and psychosocial burden. However, clinically practical tools for early recognition and objective functional assessment remain limited. This study aimed to evaluate whether clinically accessible eye-tracking could support auxiliary classification of thyroid-associated ophthalmopathy and characterize visual attention patterns toward disease-related periocular images.
METHODS: In this cross-sectional study, 229 adults from a tertiary op

Objective. Diagnosing a sports-related concussion (SRC) remains challenging, and research into diagnostic tools is limited. This study investigated whether selected eye tracking variables would be a valid tool to diagnose and monitor SRC in adult and youth participants in selected contact sports, such as Rugby Union (rugby) and football (soccer).Methods.This prospective cohort study, with 70 concussed and 92 non-concussed adult and youth athletes, assessed the validity of five previously selected eye tracking variables for SRC diagnostics and management. The performance between concussed and a

Studying genetically defined syndromes associated with increased risk for psychopathology may help in understanding neurodevelopmental mechanisms related to risk for psychopathology. Klinefelter syndrome (47,XXY) is one of the most common sex chromosomal aneuploidies (1 in 650 male births) and associated with increased vulnerability for psychopathology, including psychotic symptoms. Yet, it remains unknown whether this increased risk is associated with underlying psychophysiological mechanisms that are typically deficient in individuals with psychotic disorders. The present study assessed thre

Recent experimental studies allow more precise functional-anatomical correlations in patients with ocular motor disorders. The abducens nucleus contains two groups of cells that together subserve ipsilateral horizontal conjugate gaze: Motoneurons with axons that innervate the lateral rectus and internuclear neurons with axons that cross the midline, ascend in the medial longitudinal fasciculus (MLF) and impinge upon medial rectus motoneurons. The pontine paramedian reticular formation (PPRF) contains burst neurons for horizontal saccades. The rostral interstitial nucleus of the MLF (riMLF) in

The delay between the stimulus and the voluntary eye saccade is the only parameter of the saccadic system which can be measured by using standard apparatus. Taking manually the measurements made on records obtained by using minicomputer; the authors show that such manual measurement of latencies is easy and yet sufficiently accurate to be of great clinical value. The latencies of voluntary saccades are normal in peripheral pathology (less than 250 milliseconds). Latencies of voluntary saccades are significantly increased in extrinsic brain stem lesions: tumours (in particular ponto cerebellar

Identification of the neuron type is critical when using extracellular recordings in awake, behaving animal subjects to understand computation in neural circuits. Yet, modern recording probes have limited power to resolve neuron identity. Here, we present a generalizable framework for assigning the neuron type from extracellular recordings in nonhuman primates. The framework uses a combination of logic, circuit architecture, laminar information, and functional discharge properties. We apply the framework to the well characterized architecture of the cerebellar circuit by using well validated s

Recent lesion studies on monkeys suggest that the cerebellar lobulus petrosus of the paraflocculus (LP) and crura I and II of hemispheric lobule VII (H-7) are involved in smooth pursuit eye movement control. To reveal the relationship between the LP and H-7, we studied mossy and climbing fiber collateral inputs to these areas in four cynamolgus monkeys. After unilateral injections of retrograde tracers into the LP, labeled mossy fibers were seen ipsilaterally in the crura I and II of H-7. A very small number of labeled mossy fiber collaterals were also seen in the dorsal paraflocculus (DP). La

Pursuit eye movements introduce retinal motion that complicates the recovery of self-motion from retinal flow. An extra-retinal, eye-velocity signal could be used to aid estimation of the observer's path, perhaps by converting retino-centric into head-centric motion. This conversion is apparently not precise because we often misperceive head-centric object velocity: in the Filehne illusion, for example, a stationary object appears to move in the opposite direction to the eye movement. Similar errors should be expected when extra-retinal, eye-velocity signals are used in self-motion tasks. Howe

Previous experiments to decide whether the gain of optokinetic nystagmus (OKN) is increased or decreased by occlusion of the central retina involved the use of stationary edges on the occluder and unmatched contrasts. With these factors controlled, it was confirmed that OKN gain is severely reduced by occlusion of the central retina but only at stimulus velocities above about 30 degrees/sec. The gain of horizontal OKN was found not to increase with increasing width of the display if the lateral edges are blurred. The high gain of centrally driven OKN may be related to the ability of higher mam

It has been suggested that eye movement abnormalities seen in dyslexics are attributable to their language problems. In order to investigate this claim, we studied eye movements in dyslexic children, during several non-reading tasks. Dyslexic children were compared to normal and backward readers on measures of fixation, vergence amplitude, saccade and smooth pursuit. The results were compared to the children's phonological ability. Dyslexic children (n = 26) had significantly worse eye movement stability during fixation of small targets than normal children (n = 39). Vergence amplitudes were l

1. Ibotenic acid lesions in the monkey's middle temporal area (MT) and the medial superior temporal area (MST) in the superior temporal sulcus (STS) have previously been shown to produce a deficit in initiation of smooth-pursuit eye movements to moving visual targets. The deficits, however, recovery within a few days. In the present experiments we investigated the factors that influence that recovery. 2. We tested two aspects of the monkey's ability to use motion information to acquire moving targets. We used eye-position error as a measure of the monkey's ability to make accurate initial sacc

A case of periodic alternating nystagmus (PAN), probably of congenital type, was reported. A 25-year-old woman, who has been suffering from cerebral palsy, was referred to our clinic for the consultation of a peculiar nystagmus and intermittent oscillopsia. Electronystagmographic examination revealed that the horizontal nystagmus on straight ahead gaze in the light continuously changed its direction alternately about 110 sec in right direction and about 90 sec in left direction with about 5 sec of an interlude, during which no nystagmus was observed, between each span.
