Sympatho-adrenal hyperactivity--the key to irreversible shock?

There would appear sound evidence on which to incriminate sustained sympatho-adrenal hyperactivity as a deleterious factor of prime importance in hypovolaemic shock. The use of sympathomimetics in the treatment of shock of this type is irrational and deleterious. Their rational use in those few cases of clinical shock where they are indicated will have to await knowledge of plasma catecholamine levels in such cases. Experimental studies involving α- and β-adrenergic blockade, unilaterally and in combination, have revealed that, contrary to previous theories, deterioration is mediated through b

Relaxation of thoracic aorta and pulmonary artery rings of marmosets (Callithrix spp.) by endothelium-derived 6-nitrodopamine.

6-Nitrodopamine is a novel catecholamine released by vascular tissues, heart, and vas deferens. The aim of this study was to investigate whether 6-nitrodopamine is released from the thoracic aorta and pulmonary artery rings of marmosets (Callithrix spp.) and to evaluate the relaxing and anti-contractile actions of this catecholamine. Release of 6-nitrodopamine, dopamine, noradrenaline, and adrenaline was assessed by liquid chromatography with tandem mass spectrometry (LC-MS/MS). The relaxations induced by 6-nitrodopamine and by the selective dopamine D2 receptor antagonist L-741,626 were evalu

[Dysautonomia in Guillain-Barré syndrome].

About 20% of all GBS patients have symptoms of dysautonomia: labile hypertension, orthostatic hypotension, sinustachycardia or sinus arrest. This rate rises to 75% in patients with tetraplegia. Proprioceptive loss predicts dysautonomia independently from the severity of weakness. It is frequently responsible for dysautonomia. The afferent limb of cardiovascular regulation contains more myelinated fibers than the sympathetic and parasympathetic efferences, which determine the common classification of dysautonomia. The frequence of mixed sympathetic and parasympathetic hyperactivity is hard to e

The possible role of voltage-operated calcium channels in the enhancement of excitatory amino acid toxicity following chronic ethanol exposure in vitro.

It is well established that ethanol withdrawal is associated with hyperexcitability of central neurons, and that the increased responsiveness demonstrated to excitatory amino acids (EAAs) may contribute to the physical signs of ethanol withdrawal and to neurodegeneration. Although the mechanisms of neuronal hyperexcitation are not fully understood, there is considerable evidence to suggest an increase in both NMDA receptors and voltage operated calcium channels (VOCCs) may play an important role. Cultures of bovine adrenal chromaffin cells (BACCs) and fetal rat neocortical neurons chronically

[Localisation of an extra-adrenal phaeochromocytoma with stenosis of the renal artery (author's transl)].

Compression of the left renal artery occurred in an eleven-year old girl with arterial hypertension, the compression being caused by an extra-adrenal phaeochromocytoma. Localisation of the arteriographically not identifiable phaeochromocytoma was achieved by examining the blood in stages for the presence of catecholamines from the vena cava. This examination revealed enhanced norepinephrine levels in the left renal vein. Today the following possibilities are utilisable for localising a phaeochromocytoma: diagnosis via ultrasound, axial computer tomography, stepwise withdrawal of blood from the

Effect of catecholamines on gastrin release.

In vivo and in vitro techniques have been used to study the effect of catecholamines on gastrin release. The i.v. infusion of epinephrine in dogs produced a significant rise in plasma gastrin concentration. This response was prevented by the administration of propranolol to block beta-adrenergic receptors. The infusion of isoproterenol, a beta-adrenergic agonist, produced a significant rise in gastrin levels, while phenylephrine, an alpha-adrenergic agonist, had no effect. In in vitro studies using isolated pieces of rat antrum, isoproterenol stimulated acute phase release of gastrin, whereas

Lung beta-adrenoreceptor blockade affects perinatal surfactant release but not lung water.

We induced beta-adrenergic receptor blockade at 28 days gestation in the fetal rabbit with an irreversible beta-antagonist, bromace-tylalprenolomenthane (BrAlp). There was a marked decrease in concentration of available receptors in lung with increasing doses of BrAlp. BrAlp treatment decreased isoproterenol, but not prostaglandin, stimulated adenosine 3',5'-cyclic monophosphate (cAMP) generation in lung minces, and had no effect on activation of adenylate cyclase through non-beta-receptor-mediated components of the cyclase system in particulate preparations. Vascular reactivities of isolated and perfused human ciliary arteries.

The vascular responses to 8 agents, norepinephrine (NE), phenylephrine (PHE), xylazine (XYL), tyramine (TYR), dopamine (DA), 5-hydroxytryptamine (5-HT), prostaglandin F2 alpha (PGF2 alpha) and KCl were examined in human posterior ciliary arteries (HPCAs) which were isolated and perfused with Tyrode's solution gassed with 95% O2 and 5% CO2 under a constant flow rate at 37 degrees C. The rank order for potency of vasoconstrictions was 5-HT greater than NE greater than PHE = PGF2 much greater than DA greater than TYR much greater than KCl. Responses to XYL were various but not significant. DA fre

Muscarinic cholinergic modulation of hypothalamic estrogen binding sites.

Studies from other laboratories have demonstrated that agents which interact with the dopaminergic and noradrenergic neurotransmitter systems alter the concentrations of cytosolic hypothalamic estrogen receptors. These results have led to the hypothesis that catecholamine systems are involved intimately with the regulation of brain estrogen receptors. The present study was undertaken to determine if agents from a different neurotransmitter system similarly affect [3H]estradiol binding. The data presented here show that the muscarinic cholinergic agonist, bethanechol, increases the number of cy

Carbohydrate metabolism and its response to catecholamines as modified in alkalotic rat.

Metabolic activities and their responses to catecholamines were studied in fasted rats exposed to simulated altitudes. Analysis of hepatic levels of gluconeogenic intermediates revealed the inhibition of gluconeogenesis between glyceric acid 3-P and fructose 6-P associated with a rise of the ratios of redox pairs such as lactate to pyruvate in livers of alkalotic rats. Inhibition of gluconeogenesis was indicated also by the suppressed incorporation of glutamate 14C into blood glucose. Since no activation was detected on glycolytic pathway of skeletal muscles, marked hyperlactacidemia during al

Peripheral and central effects of circulating catecholamines.

Physical challenges, emotional arousal, increased physical activity, or changes in the environment can evoke stress, requiring altered activity of visceral organs, glands, and smooth muscles. These alterations are necessary for the organism to function appropriately under these abnormal conditions and to restore homeostasis. These changes in activity comprise the "fight-or-flight" response and must occur rapidly or the organism may not survive. The rapid responses are mediated primarily via the catecholamines, epinephrine, and norepinephrine, secreted from the adrenal medulla. The catecholamin

Beta-Adrenoceptor agonists and other cAMP elevating agents suppress PAI-1 production of human adipocytes in primary culture.

Recent studies showed that catecholamines contribute to the regulation of plasminogen activator inhibitor-1 (PAI-1) expression, at least in endothelial cells. Aim of this study was to examine the role of catecholamines on PAI-1 production by human adipocytes and, in particular, to clarify which adrenoceptor (AR) subtypes are involved. Addition of the unselective AR agonist isoproterenol led to a dose- and time-dependent suppression of PAI-1 mRNA and protein release in adipocytes from the subcutaneous and omental depot of obese subjects. A similar degree of suppression was observed in subcutane

[Medulloadrenal secretion and peripheral sympathomimetic receptors in familial dysautonomia].

A case of familial dysautonomia in a 30-year old woman with severe postural hypotension is reported. A study of adrenal medulla secretion showed that it was inadapted to the fall in blood pressure, with no increase in dopamine secretion. Insulin-induced hypoglycaemia had no effect on adrenal medulla secretion. Changes in blood pressure and heart rate were studied after administration of alpha- and beta-adrenergic drugs combined with fludrocortisone. The increase observed in blood pressure and heart rate suggests that peripheral catecholamine receptors are present in dysautonomic patients. The

[Pharmacology of the venom from the scorpion, Buthus eupeus].

Reactions induced by venom from the scorpion Buthus eupeus and by toxin of the venom have been studied on isolated organs and tissues as well as on intact body. It has been shown that substances interfering with the conduction of nerve excitation, with the mediator storage and interaction of the latter with synaptic receptors prevent the reactions induced by the venom and toxin, whereas ganglion-blocking agents do not affect these reactions. The catecholamine content in rat blood plasma has been found to be increased. It is assumed that the venom and toxin that affect the postganglionic nerve

Prostaglandin F2 alpha and cardiovascular system: review of studies conducted in dogs.

The study was designed to investigate the effect of PGF2 alpha on the arterial, cardiac and venous system and the respiration of dogs. PGF2 alpha was administered in single, graded doses as well as by i.v. perfusion. In addition, PGF2 alpha was given intra-arterially into the common carotid artery, by inhalation and by infiltration into the barosensitive zone of the carotid sinus. The interaction by drugs acting on ganglia, the catecholamine stores and the muscarinic, histaminergic, serotonergic, adrenergic and purinergic junctions, and by drugs with depressive effects at the prostaglandin rec

Serial changes in myocardial beta-adrenergic receptor after experimental brain death in dogs.

Early donor heart failure is one of the serious problems in heart transplantation. This heart failure may be related to myocardial change during brain death before implantation of the heart. Actually, a gradual increment in catecholamine requirement to maintain hemodynamics is not infrequently seen in patients with brain death. This may presumably be related to alterations in the myocardial beta-adrenergic receptor system. In this experimental study, we investigated the changes in myocardial beta-adrenergic receptors after brain death induced by intracranial hypertension in dogs. To maintain t

Assessment of the relationship between the dopaminergic pathway and severe acute respiratory syndrome coronavirus 2 infection, with related neuropathological features, and potential therapeutic approaches in COVID-19 infection.

Dopamine is a known catecholamine neurotransmitter involved in several physiological processes, including motor control, motivation, reward, cognition, and immune function. Dopamine receptors are widely distributed throughout the nervous system and in immune cells. Several viruses, including human immunodeficiency virus and Japanese encephalitis virus, can use dopaminergic receptors to replicate in the nervous system and are involved in viral neuropathogenesis. In addition, studies suggest that dopaminergic receptors may play a role in the progression and pathogenesis of severe acute respirato

Plasma catecholamine and cardiovascular responses to morphine and D-ala2-d-leu5-enkephalin in conscious rats.

The effects of morphine and DADL on cardiovascular parameters and plasma catecholamine levels were studied in conscious unrestrained rats. Morphine 30 micrograms icv increased blood pressure over the 3 hr recording period, produced an initial bradycardia followed by a tachycardia and increased plasma epinephrine and norepinephrine levels 20- and 2-fold respectively. These responses were not altered by systemic naloxone 0.8 mg/kg ia or naloxone injected icv 110 micrograms. In adrenalectomized rats blood pressure, heart rate and plasma norepinephrine responses were not significantly altered. Low

Mechanisms behind lipolytic catecholamine resistance of subcutaneous fat cells in the polycystic ovarian syndrome.

Lipolytic catecholamine resistance in sc fat cells is observed in polycystic ovarian syndrome (PCOS). The mechanisms behind this lipolysis defect were explored in vitro; sc fat cells were obtained from 10 young, nonobese PCOS women and from 14 matched, healthy control women. Fasting plasma glycerol levels were reduced by one third in PCOS (P < 0.05). Adipocytes of PCOS women were about 25% larger than in the controls (P < 0.05) and had 40% reduced noradrenaline-induced lipolysis (P < 0.05), which could be attributed to a 10-fold decreased beta(2)-adrenoceptor sensitivity (P < 0.05) and low abi

AT1 receptors mediate the effects of cold stress and amphetamine on neurocognitive performance.

Brain angiotensin II (Ang II) modulates catecholamine neurotransmission via its AT1 receptors (AT1-R) and plays a critical role in neurocognitive processes. We previously showed that amphetamine (Amph) exposure induced cognitive alterations, many times unmasked by further Amph or Ang II challenge administration. In this work, we aimed to evaluate whether Amph exposure modifies the future neurocognitive response to cold stress, an environmental cue, and the possible role of the AT1-R. Male Wistar rats received AT1-R blocker (Candesartan)/Vehicle from day 0 to 5, and Amph/Saline from day 6 to 10
