Streptococcus pyogenes is a globally prominent bacterial pathogen that exhibits strict tropism for the human host, yet bacterial factors responsible for the ability of S. pyogenes to compete within this limited biological niche are not well understood. Using an engineered recombinase-based in vivo expression technology (RIVET) system, we identified an in vivo-induced promoter region upstream of a predicted Class IIb bacteriocin system in the M18 serotype S. pyogenes strain MGAS8232. This promoter element was not active under in vitro laboratory conditions, but was highly induced within the mou

The 2006 outbreaks of H5N1 avian influenza in Egypt interrupted poultry production and caused staggering economic damage. In addition, H5N1 avian influenza viruses represent a significant threat to public health. Therefore, the rapid detection of H5 viruses is very important in order to control the disease. In this study, a qualitative reverse transcription recombinase polymerase amplification (RT-RPA) assay for the detection of hemagglutinin gene of H5 subtype influenza viruses was developed. The results were compared to the real-time reverse transcription polymerase chain reaction (RT-PCR).

Rapid and reliable methods are required for environmental monitoring of unintended releases of genetically modified (GM) crops. We developed a real-time recombinase polymerase amplification (RPA) assay targeting the bar gene for the rapid detection of GM soybean and rice, combined with a simplified NaOH-based DNA extraction method for field applications. Primer-probe set 4 provided the most specific and reliable detection, while soybean lectin and rice actin served as internal positive controls. Lectin and actin were consistently detected in GM and non-GM samples, respectively. A 0.1 N NaOH ex

As a sensitive and efficient isothermal amplification method, enzymatic recombinase amplification (ERA) holds strong potential for nucleic-acid-based disease diagnostics. However, current multiplexed ERA assays rely on distinct fluorescent labels to separate detection signals, increasing cost and optical complexity. Herein, we present a label-free dual-gene detection platform that integrates ERA with refractive-index-based sensing using an in-line tapered microfiber transducer. Gene-specific primer pairs are covalently immobilized on spatially separated microfiber segments via click chemistry,

The coconut rhinoceros beetle (CRB) is an important pest of palms in the Asia/Pacific regions. A natural pathogen of CRB, Oryctes rhinoceros nudivirus (OrNV) has successfully been introduced into virus-free, invasive CRB populations in many Pacific islands leading to pest suppression. While PCR is currently used for virus monitoring, a more portable, simplified in-field method of virus detection would be helpful. Here, we developed a Recombinase Polymerase Amplification (RPA)-based assay to detect the partial gp54 gene of OrNV (OrNV-RPA) using fluorescence-based chemistry. The assay has a high

BACKGROUND: Porcine circovirus type 3 (PCV3) is widely distributed in swine populations and has been associated with diverse clinical manifestations. Its broad tissue tropism and variable clinical presentation pose challenges for rapid and reliable diagnosis, particularly under field or resource-limited conditions.
RESULTS: Here, we report a rapid nucleic acid detection assay that integrates recombinase-aided amplification (RAA) with Pyrococcus furiosus Argonaute (PfAgo) for PCV3 detection. The sealed-tube, two-stage RAA-PfAgo assay targets a highly conserved 41-bp region within the ORF2 gene

The emerging outbreaks of infectious diseases in humans and animals worldwide pose serious threats to public health and cause substantial economic losses. This has raised the demand for more efficient and sensitive diagnostic methods to strengthen disease surveillance and early warning. In this study, we developed a portable visual platform based on ERA-CRISPR/EsCas13d for the rapid detection of porcine circovirus type 4 under resource-limited conditions. We optimized the platform by integrating enzymatic recombinase amplification (ERA), T7 transcription, and CRISPR/EsCas13d cleavage in a sing

Therapeutic integration of large DNA fragments into precise genomic loci of human cells remains challenging. The IS621 recombinase, derived from the IS110 family, utilizes a bridge RNA (bRNA) containing an independently programmable target-binding loop (TBL) and donor-binding loop (DBL) to mediate sequence-specific recombination between target and donor DNA in prokaryotes. To expand its application to human cells, we rationally engineer the IS621 protein and its cognate bRNA. This ultimately leads to the development of enIS621-tebRNA, which is capable of driving site-specific, scarless inserti

Acute hepatopancreatic necrosis disease (AHPND) has become one of the most devastating threats to global shrimp farming, causing widespread economic losses. As there is currently no effective treatment, early detection plays a crucial role in preventing disease outbreaks. However, current diagnostic methods are often complex, expensive, and impractical for on-site use in resource-limited environments. In this study, we developed a visual detection method based on enzymatic recombinase amplification (ERA) combined with the Diamond Nucleic Acid Dye, offering a rapid, cost-effective, and high-sen

Foodborne pathogens are one of the major factors contributing to food safety issues, necessitating the urgent development of rapid, efficient, and safe detection strategies for foodborne microorganisms. Herein, an on-needle detection platform of foodborne pathogens based on an integrated hydrogel microneedle array (MN) was developed for bacterial capture and recombinase polymerase amplification (RPA). This technology utilized MNs for rapid isolation of target bacteria and achieved on-needle detection through a preloaded RPA reaction system within the MN. Using Staphylococcus aureus (S. aureus)

Human herpesviruses (HHVs) are highly prevalent globally (e.g., Epstein-Barr virus with over 95% prevalence) and can persist latently for life, causing severe conditions like blindness, encephalitis, and even lethal infections in neonates. HHV reactivation is of particular clinical concern in transplantation medicine, where immunosuppression can trigger viral reactivation, leading to graft rejection, secondary infections, and increased mortality. Notably, latently infected cells show no pathological changes, their contamination in cell cultures not only undermines the accuracy of scientific da

Vaginitis frequently poses serious threats to women's health. If left untreated, it can increase the risk of infection by other pathogens such as HPV, leading to cervical dysplasia. This study developed a rapid detection method based on multiplex recombinase polymerase amplification (mRPA) combined with lateral flow strips (LFS) for the simultaneous identification of five common vaginitis pathogens: Neisseria gonorrhoeae, Gardnerella vaginalis, Candida albicans, Ureaplasma urealyticum, and Trichomonas vaginalis. The novelty of this technology lies in the fact that, the 5' end of the RPA primer

Porcine parvoviruses (PPVs) are globally recognized as significant contributors to reproductive failure in swine, primarily due to their association with fetal death. Infection in pregnant sows can lead to severe reproductive disorders including stillbirth, mummification, embryonic death and infertility. A recombinase polymerase amplification assay targeting the variable region of the outer capsid protein gene of the PPV-7 genome was developed and systematically optimized under a range of reaction conditions. The assay showed optimal amplification at a constant temperature of 35.00 ˚C for 25 m

CIC::DUX4 sarcoma (CDS) is a lethal cancer driven by a fusion between tumor suppressor Capicua (CIC) and pioneer transcription factor double homeobox 4 (DUX4). To develop an immunocompetent pre-clinical model of CDS, we previously generated three genetically engineered mouse models (GEMMs) of CDS with CIC::DUX4 regulated by loxP-STOP-loxP cassettes. However, all three models developed spontaneous tumors without Cre recombinase. Here, we established an innovative GEMM of CDS (dFLEx CDS) that employs a dual recombinase (Cre + FLPE) FLEx-switch design to activate CIC::DUX4 expression and initiate

Toxoplasma gondii is an obligate intracellular parasite with a high replication rate that can lead to DNA replicative stress, in turn associated with the generation of DNA double-strand breaks (DSBs). Cells have two main pathways to repair DSBs: non-homologous end joining and homologous recombination repair (NHEJ and HRR respectively). RAD51 is the key recombinase in the HRR pathway. In this work, we achieved endogenous tagging of the RAD51 gene using the Auxin Inducible Degron (AID) system, to generate the clonal line RH RAD51HA-AID. Here we demonstrate that RAD51 is expressed in replicative

Cre-recombinase inducible model systems are extensively used in cancer research to manipulate gene expression in specific tissues and induce autochthonous tumor growth. These systems often involve the cross-breeding of genetically engineered organisms containing loxP-flanked alleles with those expressing Cre-recombinase. This approach, while effective, has the challenge of requiring high numbers of animals due to breeding requirements. Other frequently used tumor induction methods in cancer research involve the direct application of viral Cre-recombinase vectors. This approach presents the cha
