Central cardiovascular actions of D-Ala2-Met5-enkephalinamide in the rat: effects of naloxone and nucleus reticularis gigantocellularis lesion.

In pentobarbital-anesthetized rats, intraventricular administration of D-Ala2-Met5-enkephalinamide (D-Ala, 30, 100 or 300 nmol/kg) dose-dependently elicited a reduction in arterial pressure. This D-Ala-promoted hypotension was significantly antagonized by naloxone pretreatment (3 mg/kg, i.c.v.) and attenuated by bilateral focal nucleus reticularis gigantocellularis (NRGC) lesions. At higher doses, D-Ala also produced a delayed hypertension that was not only unaffected by naloxone or NRGC lesions, but was in fact potentiated by such pretreatments. We speculate that D-Ala may produce its cardiov

Organization of ascending spinal projections in Caiman crocodilus.

Ascending spinal projections in the caiman (Caiman crocodilus) were demonstrated with Nauta and Fink-Heimer methods following hemisections of the third spinal segment in a series of twelve animals. These results were compared with earlier data in the literature obtained from a turtle, a snake, and a lizard using the same experimental and histological procedures. The results show remarkable similarities considering that each species represents a different reptilian order with different evolutionary history and habitat. However, the caiman displays several important peculiarities. Although the d

[The disinhibitory influence of the activating system when artificially excited from different points in the brain].

EEG activation can be produced by electrical stimulation of some cortical points with the same threshold current strength as by the midbrain RF and thalamic CM stimulation. Near-threshold stimulation of all these points acting simultaneously with inhibitory conditioned signals does not disturb the effector inhibition but displays an EEG difference between negative signals: the fine differentiation sound evokes considerable EEG desynchronization, while the rough one does not change the background rhythms. The same stimulation combined with a positive signal which has been made ineffective by su

Comprehensive analysis of keloid vasculature by tissue clearing and 3D imaging.

Keloids are a complex type of scar tissue formed by exaggerated wound healing, characterised by the overgrowth of thick fibrous tissue beyond the original wound boundary. While the crucial relationship between keloid formation and the vascular system has been highlighted, conflicting findings have been reported regarding the characterisation of keloid vasculature. Here, we successfully characterised the detailed three-dimensional (3D) structure of vasculature in keloid tissues using tissue clearing methods combined with 3D imaging. First, we compared two optical tissue clearing methods, the cl

Central origins of cranial nerve parasympathetic neurons in the rat.

The location of central neurons that contribute preganglionic parasympathetic axons to cranial nerves VII, IX, and X in rats has been identified using horseradish peroxidase (HRP) tracing methods. Collectively, these neurons form an uninterrupted dorsal column that extends over the entire length of the medulla. The cephalic end of this column turns ventrally with neurons scattered in the parvicellular reticular formation between the rostral pole of the nucleus of the solitary tract (NST) and the facial motor nucleus. Applying HRP crystals to the cut cervical vagus labels neurons in the classic…

[Generalization of activation reaction extinction during consecutive stimulation of different parts of the brain].

The process of extinction of the brain activation reactions ("arousal") was studied in chronic experiments on cats with implanted electrodes during repeated electrical stimulation of alternated points in the cortex and in the brain stem reticular formation. Extinction of the reactions achieved by stimulation of one point resulted in the loss of excitability both at this point and in other activating structures at different levels of the brain. The sequence of stimulated structures was of no importance. A possibility is suggested of development of a generalized inhibition in the neural net of t…

Stimulation of the cuneiform nucleus enables training and boosts recovery after spinal cord injury.

Severe spinal cord injuries result in permanent paraparesis in spite of the frequent sparing of small portions of white matter. Spared fibre tracts are often incapable of maintaining and modulating the activity of lower spinal motor centres. Effects of rehabilitative training thus remain limited. Here, we activated spared descending brainstem fibres by electrical deep brain stimulation of the cuneiform nucleus of the mesencephalic locomotor region, the main control centre for locomotion in the brainstem, in adult female Lewis rats. We show that deep brain stimulation of the cuneiform nucleus e

Decreased release of D-aspartate in the guinea pig spinal cord after lesions of the red nucleus.

This study attempts to determine if fibers that project from the guinea pig red nucleus to the spinal cord use L-glutamate and/or L-aspartate as transmitters. Unilateral injections of kainic acid were placed stereotaxically in the red nucleus to destroy the cells of origin of the rubrospinal tract. Six days after the injection, Nissl-stained sections through the lesion site showed that the majority of neurons in the red nucleus ipsilateral to the kainic acid injection were destroyed. In addition, the lesioned area included parts of the surrounding midbrain reticular formation. Silver-impregnat…

Cortically evoked responses of superior salivary nucleus neurons in the cat.

Salivation can be controlled by the inputs from the cerebral cortex as well as from the sensory nerves. However, there has been no detailed report so far on the inputs to superior salivatory nucleus (SSN) neurons from the cerebral cortex. The responses of SSN neurons to electrical stimulation of orbital and coronal gyri were investigated in ketamine anesthetized cats. A total of 56 SSN neurons were identified by the antidromic spike responses to stimulation of the right chorda tympani nerve. Their responsiveness to stimulation of the orbital and anterior coronal gyri and the lingual nerve at t

Neuroanatomical status of monkeys showing functional compensation following neonatal cerebellar lesions.

Neuroanatomical changes observed at the light microscopic level in various brain areas of four adult monkeys, who had various degrees of cerebellar ablation shortly after birth, are described in this study. Extensive neonatal hemilateral ablations of the cerebellar cortex (sparing the nuclei), which have previously been shown to leave the adult monkey with no discernible motor deficits, lead to substantial degeneration, mainly within the remaining cerebellum and the brain stem. In particular: 1) Ipsilateral to the lesion the intracerebellar nuclei and to some extent also the lateral vestibular
