Diabetes and the Retina: The Role of Medical Treatment.

About 20% of people with diabetes worldwide have diabetic retinopathy (DR); nearly 25% will have DR in 2045. DR and vision impairment are associated with atherosclerotic cardiovascular disease (ASCVD), dementia, and mortality. DR tracks with hyperglycemia, and intensive glycemic treatment improves DR, although its ultimate effect in reducing visual impairment is less clear. But rapid falls from high to more normal HbA1c are associated with worsening DR. BP treatment to systolic levels in the low to mid 140 mmHg range is associated with a reduction in DR, but there is some evidence that systoli

Current Perspectives of TLR2 Signalling in the Retina.

Toll-like receptor 2 (TLR2) signalling is crucial in initiating the innate immune response. Under normal conditions, TLR2 can recognise and respond to danger signals in the body and protect against damaging pathogens and molecules. However, dysregulation of this tightly controlled cascade has been implicated in various retinal disorders. There are many endogenous sterile ligands present in a degenerating retina that could lead to aberrant TLR2 activation. This culminates in an overaction of the innate immune response, which leads to an excess of pro-inflammatory cytokine production and results…

Saltatory axonal conduction in the avian retina.

In contrast to most parts of the vertebrate nervous system, ganglion cell axons in the retina typically lack myelination. In the majority of species, ganglion cell axons only become myelinated after leaving the retina to form the optic nerve. The avian retina, however, presents a remarkable exception in that ganglion cell axons are partly myelinated in the retinal nerve fibre layer. It was hypothesized that the optically detrimental properties of retinal myelination are evolutionarily offset by advantages in spike conduction velocity. Using high-resolution multielectrode array recordings, we a…

Tropism of the AAV6.2 Vector in the Murine Retina.

Retinitis pigmentosa (RP) is a progressive inherited retinal dystrophy (IRD) that primarily affects rod photoreceptor cells, leading to the degeneration of photoreceptors and the gradual loss of vision. While RP is one of the most studied IRDs, other neurodegenerative diseases affecting the retina and optic nerve, such as glaucoma, also involve common mechanisms of cellular stress and degeneration. Current therapeutic approaches under investigation include gene therapy, retina prosthesis, and neuroprotection. Among these approaches, gene therapy has shown promise, though challenges related to …

Multiple Roles of cAMP in Vertebrate Retina.

cAMP is a key regulatory molecule that controls many important processes in the retina, including phototransduction, cell development and death, growth of neural processes, intercellular contacts, retinomotor effects, and so forth. The total content of cAMP changes in the retina in a circadian manner following the natural light cycle, but it also shows local and even divergent changes in faster time scales in response to local and transient changes in the light environment. Changes in cAMP might also manifest or cause various pathological processes in virtually all cellular components of the r…

Evidence of lymphatics in the rat eye retina.

BACKGROUND: The lymphatic structure of the eye is still under debate. It is mainly assumed that the retina is primarily drained by prelymphatics and not by lymphatics per se. We aimed to identify lymphatics in the rat retina. METHODS: Eyes from ten Wistar rats were paraffin-embedded and the lymphatic marker podoplanin (D2-40) was investigated. RESULTS: We identified in the rat retina a blunt-end network of lymphatic endothelial vessels. It consisted of circumferential vessels within the outer and, respectively, inner plexiform layers, connected by radial dichotomous vessels. Moreover, D2-40 …

Huntington's disease and neurovascular structure of retina.

BACKGROUND: Retinal biomarkers in neurodegenerative disorders have attracted much attention in recent years. Recent studies have reported visual dysfunction in Huntington's disease (HD). However, little is known about retinal structural changes in HD. METHODS: A total of 50 subjects, including 25 motor-manifest HD patients and 25 gender- and age-matched controls, were enrolled. Unified Huntington's Disease Rating Score-Motor part was assessed in HD patients. Spectral-domain Optical Coherence Tomography (SD-OCT) was used to evaluate the macular thickness and peripapillary retinal nerve fiber l…

Neurodegeneration, Neuroprotection and Regeneration in the Zebrafish Retina.

Neurodegenerative retinal diseases, such as glaucoma and diabetic retinopathy, involve a gradual loss of neurons in the retina as the disease progresses. Central nervous system neurons are not able to regenerate in mammals, therefore, an often sought after course of treatment for neuronal loss follows a neuroprotective or regenerative strategy. Neuroprotection is the process of preserving the structure and function of the neurons that have survived a harmful insult; while regenerative approaches aim to replace or rewire the neurons and synaptic connections that were lost, or induce regrowth of…

The blood-retina barrier in health and disease.

The blood-retina barrier (BRB) is the term used to define the properties of the retinal capillaries and the retinal pigment epithelium (RPE), which separate the systemic circulation from the retina. More specifically, the inner blood-retina barrier (iBRB) is used to describe the properties of the endothelial cells that line the microvasculature of the inner retina, while the outer blood-retina barrier (oBRB) refers to the properties of the RPE cells that separate the fenestrated choriocapillaris from the retina. The BRB is not a fixed structure; rather, it is dynamic, with its components makin…

Obstructive sleep apnea and the retina: a review.

CITATION: This review's objective was to synthesize the literature on the repercussions of obstructive sleep apnea (OSA) in the retinal vascular system. Two independent investigators conducted a search using the MEDLINE/PubMed database using the following terms: sleep apnea syndrome, obstructive sleep apnea, retina, vascular tortuosity, central serous chorioretinopathy, diabetes mellitus, and subfoveal choroidal thickness. Patients with OSA present increased vascular tortuosity compared with patients without OSA, decreased parafoveal and peripapillary vessel density, and increased retinal vein…

Zinc Nutrition and Inflammation in the Aging Retina.

Zinc is an essential nutrient for human health. It plays key roles in maintaining protein structure and stability, serves as catalytic factor for many enzymes, and regulates diverse fundamental cellular processes. Zinc is important in affecting signal transduction and, in particular, in the development and integrity of the immune system, where it affects both innate and adaptive immune responses. The eye, especially the retina-choroid complex, has an unusually high concentration of zinc compared to other tissues. The highest amount of zinc is concentrated in the retinal pigment epithelium (RPE…

Potential of Müller Glia for Retina Neuroprotection.

Müller glia constitute the main glial cells of the retina. They are spatially distributed along this tissue, facilitating their close membrane interactions with all retinal neurons. Müller glia are characterized by their active metabolic functions, which are neuroprotective in nature. Although they can become reactive under pathological conditions, leading to their production of inflammatory and neurotoxic factors, their main metabolic functions confer neuroprotection to the retina, resulting in the promotion of neural cell repair and survival. In addition to their protective metabolic feature…

The Circular RNome of Developmental Retina in Mice.

Circular RNAs (circRNAs) represent a class of noncoding RNAs with a wide expression pattern, and they constitute an important layer of the genome regulatory network. To date, the expression pattern and regulatory potency of circRNAs in the retina, a key part of the central nervous system, are not yet well understood. In this study, RNAs from five stages (E18.5, P1, P7, P14, and P30) of mouse retinal development were sequenced. A total of 9,029 circRNAs were identified. Most circRNAs were expressed in different stages with a specific signature, and their expression patterns were different from …

The effects of microwave radiation on rabbit's retina.

PURPOSE: Mobile cell phones are used extensively these days, and their microwave (MW) radiation has been shown to affect the eye. The purpose of the present study was to evaluate the effects of MW radiation on rabbit retina. METHODS: This experimental study (concluded in 2015) was conducted on 40 adult white New Zealand rabbits. A Global System for Mobile Communications (GSM) cell phone simulator was used for MW irradiation. The rabbits were randomized into five groups (8 in each) and treated as follows: Group 1: no irradiation (sham); Group 2: irradiation at 10 cm for 1 day; Group 3: irradia…

Impact of bronchopulmonary dysplasia on brain and retina.

Many premature newborns develop bronchopulmonary dysplasia (BPD), a chronic lung disease resulting from prolonged mechanical ventilation and hyperoxia. BPD survivors typically suffer long-term injuries not only to the lungs, but also to the brain and retina. However, currently it is not clear whether the brain and retinal injuries in these newborns are related only to their prematurity, or also to BPD. We investigated whether the hyperoxia known to cause histologic changes in the lungs similar to BPD in an animal model also causes brain and retinal injuries. Sprague Dawley rat pups were expose…

Light damage to the retina: an historical approach.

A brief review of retinal light damage is presented. Thermal damage requires a local rise in temperature of at least 10 °C, causing an instant denaturation of proteins. The primary absorber is melanin. Photochemical damage occurs at body temperature and involves cellular damage by reactive forms of oxygen. The photosensitizers are photoproducts of the visual pigments. First indications that non-thermal damage might exist, in particular in the case of eclipse blindness, was presented by Vos in 1962. Attribution thereof to photochemical action was presented in 1966 by Noell et al who also measur…
