Anaerobic phenanthrene biodegradation by a new salt-tolerant/halophilic and nitrate-reducing Virgibacillus halodenitrificans strain PheN4 and metabolic processes exploration.

The biodegradation of polycyclic aromatic hydrocarbons (PAHs) under hypersaline environments has received increasing attention, whereas the study of anaerobic PAH biodegradation under hypersaline environments is still lacking. Here, we found a pure culture designated PheN4, which was affiliated with Virgibacillus halodenitrificans and could degrade phenanthrene with nitrate as the terminal electron acceptor and a wide range of salinities (from 0.3% to 20%) under anaerobic environments. The optimal salinity for biodegradation of phenanthrene by PheN4 was 5%, which could degrade 93.5% of 0.62 ±

Changes in the Sterol Composition of the Plasma Membrane Affect Membrane Potential, Salt Tolerance and the Activity of Multidrug Resistance Pumps in Saccharomyces cerevisiae.

We investigated the impact of the deletions of genes from the final steps in the biosynthesis of ergosterol (ERG6, ERG2, ERG3, ERG5, ERG4) on the physiological function of the Saccharomyces cerevisiae plasma membrane by a combination of biological tests and the diS-C3(3) fluorescence assay. Most of the erg mutants were more sensitive than the wild type to salt stress or cationic drugs, their susceptibilities were proportional to the hyperpolarization of their plasma membranes. The different sterol composition of the plasma membrane played an important role in the short-term and long-term proce

Is salt stress tolerance in Casuarina glauca Sieb. ex Spreng. associated with its nitrogen-fixing root-nodule symbiosis? An analysis at the photosynthetic level.

Casuarina glauca is an actinorhizal tree which establishes root-nodule symbiosis with N2-fixing Frankia bacteria. This plant is commonly found in saline zones and is widely used to remediate marginal soils and prevent desertification. The nature of its ability to survive in extreme environments and the extent of Frankia contribution to stress tolerance remain unknown. Thus, we evaluated the ability of C. glauca to cope with salt stress and the influence of the symbiosis on this trait. To this end, we analysed the impact of salt on plant growth, mineral contents, water relations, photosynthetic

Cloning, Characterization and Expression Pattern Analysis of a Cytosolic Copper/Zinc Superoxide Dismutase (SaCSD1) in a Highly Salt Tolerant Mangrove (Sonneratia alba).

Mangroves are critical marine resources for their remarkable ability to tolerate seawater. Antioxidant enzymes play an especially significant role in eliminating reactive oxygen species and conferring abiotic stress tolerance. In this study, a cytosolic copper/zinc superoxide dismutase (SaCSD1) cDNA of Sonneratia alba, a mangrove species with high salt tolerance, was successfully cloned and then expressed in Escherichia coli Rosetta-gami (designated as SaCSD1). SaCSD1 comprised a complete open reading frame (ORF) of 459 bp which encoded a protein of 152 amino acids. Its mature protein is predi…

Genome-Wide Identification and Comparative Analysis of the TCP Transcription Factor Family in Three Glycyrrhiza Species Reveals GuTCP16 as a Positive Regulator of Salt Tolerance.

The TCP transcription factor family is a plant-specific group of regulators with well-documented roles in mediating abiotic stress responses. Licorice (Glycyrrhiza) is a high-value medicinal crop of global economic significance, predominantly cultivated in arid and semi-arid environments where saline soils constrain seed germination, impair seedling development, and diminish both rhizome biomass and the accumulation of pharmacologically active metabolites. Despite the agronomic importance of this genus, a systematic genome-wide survey of the TCP family and its contribution to salt tolerance in…

Characterization of the AP2/ERF gene family in Fritillaria thunbergii and functional evaluation of FtAP2/ERF-61 in conferring tolerance to drought and salt stress.

BACKGROUND: The APETALA 2/ethylene-responsive element binding factors (AP2/ERFs) are crucial modulators of plant development and stress responses. Fritillaria thunbergii is an important medicinal herb with considerable therapeutic efficacy. However, despite its importance, no systematic identification and functional characterization of the AP2/ERF gene family in F. thunbergii has been reported to date. RESULTS: This study identified a total of 71 AP2/ERF genes in F. thunbergii and classified them into 11 subgroups, with genes in the same subgroup exhibiting similar motifs and domains. Express

Individual or successiveseed priming with nitric oxide and calcium toward enhancing salt tolerance of wheat crop through early ROS detoxification and activation of antioxidant defense.

Despite the considerable efforts reported so far to enhance seed priming, novel ideas are still needed to be suggested to this sustainable sector of agri-seed industry. This could be the first study addressing the effect of nitric oxide (NO) under open field conditions. The impacts of seed redox-priming using sodium nitroprusside (SNP) and osmo-priming with calcium chloride (CaCl2), both applied individually or successively, were investigated under salinity stress conditions on wheat plants (Triticum aestivum L.). Various parameters, including water relations, growth, yield, photosynthetic pig

Low-cost, scalable fabrication of antibacterial asymmetric hydrogels inspired by mangrove systems for superior oil-water separation and salt-tolerant evaporation.

Offshore oil spill incidents not only pollute marine ecosystems but also exacerbate freshwater shortages in coastal regions where seawater is abundant but drinkable water is scarce. Addressing these dual challenges requires materials that can efficiently separate oil from water while simultaneously enabling seawater desalination. Here, inspired by the asymmetric architecture of mangrove systems, we developed a scalable strategy for constructing composite hydrogels with hierarchical pore structures. The fabricated hydrogel integrates oil-water separation and salt-tolerant solar evaporation into

Microbial community biomass and structure in saline and non-saline soils associated with salt- and boron-tolerant poplar clones grown for the phytoremediation of selenium.

Poplar trees (Populus spp.) are often used in bioremediation strategies because of their ability to phytoextract potential toxic ions, e.g., selenium (Se) from poor quality soils. Soil microorganisms may play a vital role in sustaining health of soil and/or tolerance of these trees grown in poor quality soils by contributing to nutrient cycling, soil structure, overall soil quality, and plant survival. The effect of naturally occurring salts boron (B) and Se on soil microbial community composition associated with poplar trees is not known for bioremediation strategies. In this study, three Pop

Does Salicylic Acid (SA) Improve Tolerance to Salt Stress in Plants? A Study of SA Effects On Tomato Plant Growth, Water Dynamics, Photosynthesis, and Biochemical Parameters.

Environmental stresses such as salinity directly impact crop growth, and by extension, world food supply and societal prosperity. It is estimated that over 800 million hectares of land throughout the world are salt-affected. In arid and semi-arid regions, salt concentration can be close to that in the seawater. Hence, there are intensive efforts to improve plant tolerance to salinity and other environmental stressors. Salicylic acid (SA) is an important signal molecule for modulating plant responses to stress. In the present study, we examined, on multiple plant growth related endpoints, wheth

The effects of 405 nm light on bacterial membrane integrity determined by salt and bile tolerance assays, leakage of UV-absorbing material and SYTOX green labelling.

Bacterial inactivation by 405 nm light is accredited to the photoexcitation of intracellular porphyrin molecules resulting in energy transfer and the generation of reactive oxygen species that impart cellular oxidative damage. The specific mechanism of cellular damage, however, is not fully understood. Previous work has suggested that destruction of nucleic acids may be responsible for inactivation; however, microscopic imaging has suggested membrane damage as a major constituent of cellular inactivation. This study investigates the membrane integrity of Escherichia coli and Staphylococcus aur

Screening and optimization of salt-alkali tolerant phosphate-solubilizing Enterobacter sp. AD8: applications in soybean growth promotion and saline-alkali soil remediation.

Soil salinization severely limits crop growth, nutrient availability and phosphorus use efficiency. Combined saline-alkali stress and phosphorus deficiency greatly restrict soybean productivity. Applying salt-alkali tolerant phosphate-solubilizing bacteria is an effective biological strategy to improve crop performance and soil fertility. In this study, 15 strains were isolated from local saline-alkali soil. After functional screening, Enterobacter sp. AD8 showed excellent phosphate-solubilizing (264.21 mg/L soluble P) and IAA-producing (58.09 µg/mL) capacities. Its optimal culture conditions

H2S Enhanced the Tolerance of Malus hupehensis to Alkaline Salt Stress through the Expression of Genes Related to Sulfur-Containing Compounds and the Cell Wall in Roots.

Malus is an economically important plant that is widely cultivated worldwide, but it often encounters saline-alkali stress. The composition of saline-alkali land is a variety of salt and alkali mixed with the formation of alkaline salt. Hydrogen sulfide (H2S) has been reported to have positive effects on plant responses to abiotic stresses. Our previous study showed that H2S pretreatment alleviated the damage caused by alkaline salt stress to Malus hupehensis Rehd. var. pingyiensis Jiang (Pingyi Tiancha, PYTC) roots by regulating Na+/K+ homeostasis and oxidative stress. In this study, transcri

Integrative roles of nitric oxide and hydrogen sulfide in melatonin-induced tolerance of pepper (Capsicum annuum L.) plants to iron deficiency and salt stress alone or in combination.

There seems to be no report in the literature on the effect of melatonin (MT) in relieving the detrimental effects of combined application of salt stress (SS) and iron deficiency (ID). Therefore, the effect of MT on the accumulation/synthesis of endogenous nitric oxide (NO) and hydrogen sulphide (H2 S) and how far these molecules are involved in MT-improved tolerance to the combined application of ID and SS in pepper (Capsicum annuum L) were tested. Hence, two individual trials were set up. The treatments in the first experiment comprised: Control, ID (0.1 mM FeSO4 ), SS (100 mM NaCl) and ID +
