OBJECTIVE: This study investigates the diagnostic procedures, therapeutic strategies, and postoperative surveillance of primary intracranial Ewing Sarcoma (EWS), with a focus on a pediatric case in the posterior fossa and a comparative literature review.
CASE REPORT: We present the case of a 6-year-old girl admitted with a 1-month history of worsening headaches and signs of elevated intracranial pressure. Preoperative MRI identified a large, heterogeneously enhancing mass in the left posterior fossa, initially suspected to be a medulloblastoma. The patient underwent maximal safe surgical rese

We applied Western blotting, using an antibody against the carboxy terminal of the FLI1 protein, for detection of the 68-kDa EWS/FLI1 fusion protein in cultured Ewing's sarcoma cells and in four surgical biopsies of Ewing's sarcoma. Of six different human cell lines, the 68-kDa fusion protein was identified only in Ewing's sarcoma cells carrying the t(11;22)(q24;q12) translocation. The four samples from Ewing's sarcoma patients were also found to contain the 68-kDa fusion protein. The lowest detection level for total protein loaded on the gel was 0.3 microg. When whole Ewing's sarcoma cells we

Round cell sarcomas represent a continuously evolving category of malignant mesenchymal tumors, currently encompassing 5 main tumor subtypes: Ewing sarcoma, Capicua Transcriptional Repressor (CIC)-rearranged sarcoma, sarcomas with BCL6-corepressor (BCOR)-genetic abnormalities, POZ/BTB and AT hook containing zinc finger 1 (PATZ1)-rearranged sarcoma, and nuclear factor of activated T cells 2 (NFATC2)-rearranged sarcoma. These tumors show some degree of histomorphologic overlap, which, coupled with their infrequency, may render their diagnosis challenging for the pathologist. Furthermore, althoug

Tumor models allowing for the in vivo investigation of molecular mechanisms driving tumor progression and metastasis are important to develop novel strategies for cancer treatment. Unfortunately, for Ewing sarcoma, no adequate genetic animal models are currently available, although there is progress in zebrafish. Nevertheless, mouse xenotransplantation models are the state-of-the-art to model Ewing sarcoma in vivo. Here, we describe an alternative Ewing sarcoma xenotransplantation model in embryonic and larval zebrafish. This zebrafish model offers live imaging and easy compound testing opport

Ewing sarcoma tumors are defined by chromosomal translocations fusing EWSR1 to ETS family genes, most commonly FLI1. The resulting EWS::FLI1 fusion oncoprotein drives tumorigenesis while simultaneously generating unique metabolic and molecular dependencies that may be exploited for Ewing sarcoma-specific therapies. Through analysis of the Cancer Dependency Map (DepMap, Broad Institute), we identified that Ewing sarcoma cell lines exhibit selective sensitivity to loss of genes regulating selenium metabolism compared to other cancer lineages. Ewing sarcoma cells are similarly sensitive to knocko

BACKGROUND/AIM: Ewing sarcoma (ES) is a rare tumor, but one of the most common primary bone tumors of the foot and ankle. The aim of this study was to review long-term outcomes of ES of the foot or ankle.
PATIENTS AND METHODS: We reviewed 21 patients (16 male, 5 female, mean age 20±14 years) treated for ES of the foot or ankle. Local control consisted of surgical resection alone (n=16), surgery with radiotherapy (n=1), or definitive radiotherapy (n=4). Of the patients who underwent surgical resection, 11 underwent amputation and 6 underwent limb salvage.
RESULTS: The 5-, and 10-year disease-

Sub-populations of cancer-associated fibroblast (CAF)-like tumor cells deposit extracellular matrix (ECM) proteins that support Ewing sarcoma (EwS) progression and metastasis. We previously showed a hallmark of CAF-like EwS cells is their hybrid transcriptional state wherein the driver fusion oncogene, EWS::FLI1, maintains activation of proliferative programs but loses capacity to repress mesenchymal genes. Here, we studied primary patient tumors and cell line models to identify molecular drivers of this hybrid state. Our data reveal that hybrid EwS cells are induced and maintained by a TGFβ s

We present the case of a 22-year-old man with a firm and tender mass in the left scrotum region. Color Doppler ultrasound and radiographic examination (CT and MRI) suggested that the mass arose from the left epididymis without tumor metastasis. The histopathological and immunohistochemical findings revealed a chrysanthemum-like arrangement of round tumor cells, along with strong positive expression of CD99, vimentin, CD117, CD56, and Ki-67 (70%), supporting the diagnosis of primary epididymal Ewing sarcoma. The patient underwent surgical excision and received 3 cycles of postoperative chemothe

Primary extraskeletal Ewing sarcoma (EES) is a rare small round cell malignancy that accounts for less than 1% of all sarcomas. It is found most commonly in the trunk and lower limbs and very rarely in the pleura and can be easily misdiagnosed in clinical practice. This study presents the case of an 11-year-old boy who presented to our hospital with no apparent cause of left shoulder pain for 6 months. On physical examination, tenderness was noted in the left chest wall and shoulder joint, which had a limited range of motion. Computed tomography (CT) and magnetic resonance imaging (MRI) of the

An 89-year-old male presented with syncope and worsening difficulty in breathing through the left nostril. Computed tomography demonstrated a tumor in the anterior ethmoid air cells and maxillary sinus, which extended into the frontal lobe. Magnetic resonance imaging similarly demonstrated an aggressive lesion. This mass was difficult to differentiate from more commonly seen lesions at this location such as an esthesioneuroblastoma or nasopharyngeal carcinoma. Direct visualization, biopsy, and subsequent pathologic analysis eventually confirmed the diagnosis of malignant Ewing sarcoma (EWS). O

A 45-year-old female (G1, P1) with HPV-52 infection and low-grade cervical cytology presented to our clinic with an asymptomatic cervical mass. She suffered amenorrhea for the past 3 years due to medication for thyroid cancer. Magnetic resonance imaging revealed a mass sized 7.1 × 6.3 × 5.7 cm in the cervix with local invasion, and multiple lesions in the left ilium and femur. Histologically blue small round cells, positive for CD99 and FLI-1, obtained by cervical biopsy and the EWSR1 gene rearrangement detected by Fluorescence in situ hybridization confirmed the diagnosis of Ewing sarcoma. BACKGROUND: Ewing's sarcoma is an aggressive bone and the surrounding soft tissue cancer primarily found in children and young adults. It is mostly observed in the trunk and long bones while unusually seen in the sacrum. Sacral Ewing sarcoma has a unique anatomic limitation due to neurological structures, vertebral column, and pelvic involvement. Hence, identifying sacral Ewing's sarcoma remains a challenge.
METHOD: This is a case series including three patients diagnosed with Sacral Ewing's sarcoma. These three cases have been diagnosed based on clinical and radiographic examination, MRI, hi

Swelling of the parotid region is a frequent reason for consultation and the first clinical sign of both benign and malignant parotid tumors. These parotid tumors represent 80 % of tumors of the main salivary glands. They present a highly variable clinical and radiological picture and are known to have a wide histological diversity. Some histological types are rare entities found in the parotid gland, such as Ewing's sarcoma and lymphoepithelial cyst, the latter of which is often associated with HIV co-morbidity, and therefore requires careful and adequate therapeutic attitudes and strategies.

Background: Ewing sarcoma is the second most common malignant bone tumor in children, but it rarely originates from extra-skeletal sites. The commonly involved sites of soft tissue include paravertebral spaces, lower extremities, the pelvis, head, and neck, while primary extra-skeletal Ewing sarcoma (EES) located in the genitals is extremely rare. Case Presentation: We report a young patient who presented to our hospital with a painful erection of the penis and limited motion of the left hip. Magnetic resonance imaging showed a hyperintense mass with invasion of adjacent tissue in the penis an

Tumors of the spinal cord and cauda equina show a wide spectrum of histology and require careful diagnosis and treatment. Primary intradural extramedullary Ewing sarcoma (IEES) is extremely rare, and initial imaging and clinical findings for this tumor mimic those of benign intradural spinal tumors. The present report describes a case of a 35-year-old woman who presented with IEES with meningeal seeding, and the literature on the management and clinical course of these tumors was reviewed. An examination revealed right-side leg dominant rapidly progressive hypoesthesia and motor deficits. MRI

The therapies used to treat Ewing sarcoma are associated with a risk of second malignant neoplasm (SMN). We conducted a systematic review to pool available evidence on the risks, types, and outcomes after SMN. We obtained 52 articles that met inclusion criteria. Cumulative incidence rates of SMN ranged from 0.9 to 8.4% and 10.1 to 20.5% at 5 and 30 years after initial diagnosis. Of the 327 reported SMNs, 63.6% were solid tumors, although acute myeloid leukemia /myelodysplastic syndrome was the single most commonly diagnosed SMN, with generally poor outcomes. Patients treated for Ewing sarcoma

Ewing sarcoma and peripheral primitive neuroectodermal tumor belong to the Ewing sarcoma (ES) family of tumors originating from a primitive neural tube. We report a 31-year-old man who was admitted to the urology clinic with complaints of fever, nausea, and dysuria. A right-sided adrenal mass was detected during ultrasonography. The lesion was then evaluated with magnetic resonance imaging, which showed areas of necrosis amid heterogeneous solid areas. Whole body scan with 2-deoxy-2-[fluorine-18]fluoro-D-glucose integrated with computed tomography and bone scan studies showed pulmonary and oss
