Genome-wide characterization of the expansin gene family in Sorghum bicolor reveals their regulatory features and roles in drought response.

BACKGROUND: Expansins (EXPs) are non-enzymatic proteins that mediate cell wall loosening and play essential roles in plant growth, development, and stress responses. However, the expansin gene family in Sorghum bicolor remains insufficiently characterized. RESULTS: In this study, we identified 68 expansin genes in S. bicolor (SbEXPs), predominantly belonging to the EXPA and EXPB subfamilies. Structural and evolutionary characterization revealed conserved domain organization, with gene family expansion likely driven by tandem and segmental duplications. Regulatory analysis revealed diverse cis…

Identification of expression profiles of sorghum genes in response to greenbug phloem-feeding using cDNA subtraction and microarray analysis.

The phloem-feeding by greenbug (Schizaphis graminum) elicits unique interactions with their host plants. To investigate the expression profiles of sorghum genes responsive to greenbug feeding, two subtractive cDNA libraries were constructed through different combinatorial subtractions in a strong greenbug resistance sorghum M627 line and a susceptible Tx7000 line with or without greenbug infestation. A total of 3,508 cDNAs were selected from the two cDNA libraries, and subsequent cDNA microarray and northern blot analyses were performed for identification of sorghum genes responsive to greenbu…

Subsoil tillage boosts sorghum yield in saline-alkali soils by alleviating constraints on root growth through integrated soil improvement.

To enhance crop productivity in saline-alkali soils, it is essential to overcome soil constraints that limit root growth. This study used sorghum as the test crop and conducted a field experiment in the Hetao Irrigation District of Inner Mongolia to compare subsoiling tillage (ST) with conventional tillage (CT). We evaluated soil physical structure, salt transport, microbial activity, and root morphology, and applied multivariate statistics and structural equation modeling to elucidate the mechanisms through which ST promotes crop growth. ST significantly improved the root-zone soil environmen…

A scalable and efficient UAV-based pipeline and deep learning framework for phenotyping sorghum panicle morphology from point clouds.

Sorghum canopy architecture in field trials is determined by various phenotypic traits, such plant and panicle count, leaf density and angle and panicle morphology, and canopy height. These traits together affect light capture and biomass production as well as conversion of photosynthates to grain yield. Panicle morphology exhibits considerable variation as influenced by genetics, environmental conditions and management practices. This study presents a framework for the 3D reconstruction of sorghum canopies and phenotyping panicle morphology. First, we developed a scalable, low-altitude Unmann…

Intercropping with maize and sorghum-induced saikosaponin accumulation in Bupleurum chinense DC. by liquid chromatography-mass spectrometry-based metabolomics.

Bupleuri Radix is an important medicinal plant, which has been used in China and other Asian countries for thousands of years. Cultivated Bupleurum chinense DC. (B. chinense) is the main commodity of Bupleuri Radix. The benefits of intercropping with various crops for B. chinense have been recognized; however, the influence of intercropping on the chemical composition of B. chinense is still unclear yet. In this study, intercropping with sorghum and maize exhibited little effect on the root length, root diameter, and single root mass of B. chinense. Only the intercropping with sorghum increase…

Early-stage iron deficiency alters physiological processes and iron transporter expression, along with photosynthetic and oxidative damage to sorghum.

Iron (Fe) starvation in Strategy II plants is a major nutritional problem causing severe visual symptoms and yield reductions. This prompted us to investigate the physiological and molecular consequences of Fe deficiency responses at an early stage in sorghum plants. The Fe-starved sorghum did not show shoot biomass reduction, but the root length, biomass, and chlorophyll synthesis were severely affected. The chlorophyll a fluorescence analysis showed that the quantum yield efficiency of PSII (Fv/Fm) and photosynthesis performance index (Pi_ABS) in young leaves significantly reduced in respons…

The Interplay Between Water Limitation, Dhurrin, and Nitrate in the Low-Cyanogenic Sorghum Mutant adult cyanide deficient class 1.

Sorghum bicolor (L.) Moench produces the nitrogen-containing natural product dhurrin that provides chemical defense against herbivores and pathogens via the release of toxic hydrogen cyanide gas. Drought can increase dhurrin in shoot tissues to concentrations toxic to livestock. As dhurrin is also a remobilizable store of reduced nitrogen and plays a role in stress mitigation, reductions in dhurrin may come at a cost to plant growth and stress tolerance. Here, we investigated the response to an extended period of water limitation in a unique EMS-mutant adult cyanide deficient class 1 (acdc1) t…

Vacuum-Assisted Gas Stripping Extractive Fermentation for Process Intensification of Sorghum-Based Bioethanol Production: An Experimental and Modeling Study.

Extractive fermentation can mitigate the inhibitory effects of ethanol on yeast cells by continuously removing ethanol as it is produced, thereby enhancing ethanol productivity during the sorghum hydrolysate fermentation. This approach represents a promising strategy for process intensification in starch-based bioethanol production, particularly in systems limited by product inhibition. The present study describes the modeling and experimental validation of ethanol production in separate hydrolysis and fermentation (SHF) and simultaneous saccharification and fermentation (SSF) processes, utili…

Structural Characterization and Immunostimulatory Activity of a New Slightly Branched β-glucan in Sporisorium reilianum-Infected Sorghum Head Smut.

Sporisorium reilianum (S. reilianum) is an agricultural pathogen that causes disease and reduces yield in sorghum and maize crops. However, head smut-infected sorghum and maize have been traditionally utilized for edible and medicinal purposes in some regions. In this study, the structure and immunostimulatory potency of a new slightly branched β-glucan WM-0.25(P) obtained from the S. reilianum-infected sorghum head smut were explored. WM-0.25(P) was a β-glucan with a molecular weight of 69.67 kDa and was composed of β-1,6-D-Glcp as the main chain and β-1,3-D-Glcp as the side chain. There was …

Microwave-induced complexation between sorghum starch and caffeic acid: Structural modulation and its impact on digestive and retrogradation properties.

Microwave-assisted technology offers an efficient and eco-friendly method for starch modification. In this study, sorghum starch (SS) was treated under varying microwave durations and complexed with caffeic acid (CA) to form SS-CA complexes. Microwave treatment significantly reduced crystallinity (24.07 % to 3.51 %), disrupted short-range order, and caused granule swelling, aggregation, and decreased thermal stability. Complexation induced a V-type inclusion structure. The SS-CA (120 s) complex showed a 9.17 % reduction in rapidly digestible starch (RDS) and a 14.40 % increase in resistant sta…

Introgression of dwarfing allele dw1 reduced seed dormancy and increased pre-harvest sprouting susceptibility in grain sorghum converted lines.

Grain sorghum (Sorghum bicolor L. moench) stands as a globally significant cereal crop but the adversity of pre-harvest sprouting (PHS) caused by reduced grain dormancy and moist conditions prior to harvest remains unsolved. Here, we identified a dormancy QTL using a Redlan×IS9530 RIL population, where parent lines are low in tannins and early flowering but otherwise contrasting in grain dormancy and plant height. We phenotyped this population in 2 years with informative PHS-related traits (grain germination index, embryo sensitivity to abscisic acid and in one year the actual natural sproutin…

Genetic gains in forage sorghum for adaptive traits for non - conventional area through multi-trait-based stability selection methods.

IntroductionIn the Asian tropics, unpredictable weather increases the risk of abiotic stresses in sorghum areas, making it harder to meet predicted demand. Genotype-by environment interaction (GEI) and the lack of an effective multi-trait-based selection approach make it challenging to breed climateresilient forage sorghum that adapts to nonconventional areas.MethodsThe present investigation carried out to estimate genetic parameters, inter trait associations, genetic gain under selection (SGs) of 95 diverse forage sorghum genotypes. Fourteen forage yield and other secondary traits were evalua…

High-throughput quantitative analysis of phytohormones in sorghum leaf and root tissue by ultra-performance liquid chromatography-mass spectrometry.

Plant development, growth, and adaptation to stress are regulated by phytohormones, which can influence physiology even at low concentration … rellins, cytokinins, and auxins. Using this assay, 12 phytohormones were detected and quantified in sorghum plant tissue without the need for solid phase extraction (SPE) or liquid-liquid extraction. Variation of phytohormone profiles was explored in both root and leaf tissues between three genotypes, harvested at two different developmental time points. …
