Holding but not folding: How a single charge flip uncouples the DNAJC7-Hsp70 relay in amyotrophic lateral sclerosis.

Genetic mutations impact protein function through various routes: Some catalyze new oncogenic activities, while others trigger complete structural collapse. However, the E425K mutation in DNAJC7, associated with Amyotrophic Lateral Sclerosis (ALS), presents a far more subtle and intriguing case. In their recent study in The FEBS Journal, Elmaleh et al. (2026) FEBS Lett employed high-resolution NMR to demonstrate that this mutation leaves the protein's overall structure intact while selectively paralyzing its ability to communicate with the Hsp70 chaperone machinery. In this commentary, we show

Patent ductus arteriosus in adults: Case report and review illustrating the spectrum of the disease.

Patent ductus arteriosus (PDA) is a congenital heart defect in which the ductus arteriosus, a vascular structure between the pulmonary artery and the aorta that normally closes shortly after birth, remains open. We present two cases of adults with PDA. A 28-year-old lady had a small PDA without any symptoms or left heart overload. The PDA was closed for endocarditis prevention using a coil. In a 37-year-old lady with palpitations, collapse, dyspnoea and left heart overload caused by a large PDA, percutaneous closure of the PDA was performed with an Amplatzer device. Transcatheter closure has b

Cranial settling in rheumatoid arthritis.

Cranial settling occurs in about 5-8% of patients with rheumatoid arthritis, sometimes resulting in severe neurological problems or even death. It is caused by collapse of the supporting structures at the craniovertebral junction, mainly the lateral masses of C1. Twelve cases are presented; 2 patients with severe neurological problems were treated surgically.

Atomic Insights into pH-Dependent and Water Permeation of mRNA-Lipid Nanoparticles.

The exposure of mRNA to water is likely to contribute to the instability of RNA vaccines upon storage under nonfrozen conditions. Using atomistic molecular dynamics (MD) simulations, we investigated the pH-dependent structural transition and water penetration behavior of mRNA-lipid nanoparticles (LNPs) with the compositions of Moderna and Pfizer vaccines against COVID-19 in an aqueous solution. It was revealed that the ionizable lipid (IL) membranes of LNPs were extremely sensitive to pH, and the increased acidity could cause a rapid membrane collapse and hydration swelling of LNP, confirming

Pregnancy-associated aortopathy and sudden postpartum death.

A 39-year-old woman who had undergone an uncomplicated elective cesarean section for a low-lying placenta collapsed and died the following day. At autopsy, there was dissection of an aneurysmally-dilated thoracic aorta with 400 mls of fluid and clotted blood in the pericardial sac. There were no features of Marfan syndrome or other connective tissue disorders. Histology revealed thinning of the aortic wall with fragmentation of elastic fibers and no inflammation. Vessels elsewhere were normal. This case demonstrates a rare complication of pregnancy that may not present until after delivery wit

Dynamics of the conformational transitions during the dimerization of an intrinsically disordered peptide: a case study on the human islet amyloid polypeptide fragment.

Amyloid deposits of human islet amyloid polypeptide (hIAPP) are identified in 95% of type II diabetes patients. The oligomers during the early stage of hIAPP aggregation are believed to be more cytotoxic than the mature fibrils. However, the structural details during the initial stage of hIAPP aggregation are still under debate experimentally. To understand its initial nucleation mechanism, we investigate the thermodynamics and kinetics of hIAPP(11-25) dimerization, which is the first manifestation of the interplay between intra- and inter-molecular interactions, via the construction of Markov

Synthesis and characterization of a novel o-tolyltelluronic trimethyltin ester and its cytotoxic assessment in vitro.

A new arenetelluronic triorganotin ester, namely (Me3Sn)4[o-Me-PhTe(μ-O)(OH)O2)]2 (1) has been prepared by the reaction of o-tolyltelluronic acid and Me3SnCl in the presence of potassium hydroxide. The complex was fully characterized by elemental analysis, FT-IR, NMR ((1)H, (13)C, (119)Sn) spectroscopy and X-ray crystallography. Structure analysis revealed that the complex crystallized as Sn4Te2 units and a 1D linear chain was formed by intermolecular C-HO interactions. Cytotoxic assessments showed that the complex can induce apoptotic cell death via accumulation of ROS, collapse of the MMP an

Impact of sleeping position, gravitational force & effective tissue stiffness on obstructive sleep apnoea.

Accurate prediction of deformation and collapse of the upper airway during breathing is required for effective and personalised treatment of obstructive sleep apnoea (OSA). While numerical modelling techniques such as fluid-structure interaction (FSI) are promising, an outstanding challenge is to accurately predict the deformation of the airway during breathing and thus the occurrence of OSA. These difficulties arise because the effective stiffness of the soft tissue in the human upper airway varies due to neuromuscular effects on the stiffness of the underlying muscles. In addition, both the

Promoter clearance by T7 RNA polymerase. Initial bubble collapse and transcript dissociation monitored by base analog fluorescence.

Footprinting, fluorescence, and x-ray structural information from the initial, promoter-bound complex of T7 RNA polymerase describes the very beginning of the initiation of transcription, whereas recent fluorescence and biochemical studies paint a preliminary picture of an elongation complex. The current work focuses on the transition from an initially transcribing, promoter-bound complex to an elongation complex clear of the promoter. Fluorescence quenching is used to follow the melted state of the DNA bubble, and a novel approach using a locally mismatched fluorescent base analog reports on

Constructing of Ni-Nx Active Sites in Self-Supported Ni Single-Atom Catalysts for Efficient Reduction of CO2 to CO.

The electrochemical carbon dioxide reduction reaction (CO2RR) represents a promising approach for achieving CO2 resource utilization. Carbon-based materials featuring single-atom transition metal-nitrogen coordination (M-Nx) have attracted considerable research attention due to their ability to maximize catalytic efficiency while minimizing metal atom usage. However, conventional synthesis methods often encounter challenges with metal particle agglomeration. In this study, we developed a Ni-doped polyvinylidene fluoride (PVDF) fiber membrane via electrospinning, subsequently transformed into a

PfMORC protein regulates chromatin accessibility and transcriptional repression in the human malaria parasite, Plasmodium falciparum.

The environmental challenges the human malaria parasite, Plasmodium falciparum, faces during its progression into its various lifecycle stages warrant the use of effective and highly regulated access to chromatin for transcriptional regulation. Microrchidia (MORC) proteins have been implicated in DNA compaction and gene silencing across plant and animal kingdoms. Accumulating evidence has shed light into the role MORC protein plays as a transcriptional switch in apicomplexan parasites. Collagen fibre orientation in bovine secondary osteons by collagenase etching.

The orientation of collagen fibres in bovine secondary osteons has been investigated in the scanning electron microscope (SEM) by removal independently of firstly the mineral component and secondly the collagen fibres. Demineralization of polished transverse sections reveals a lamellar structure for the collagen component but the precise orientation of the collagen in each ring is not unequivocably determined. However, by using a collagenase solution to etch away the collagen component of a polished surface, holes are produced in the mineral revealing the former position of the fibres. The gre

Embodied Transformation: Psychedelic Therapy Through the Lens of Merleau-Ponty's Phenomenology.

This paper develops a phenomenological account of the therapeutic significance of psychedelic-induced mystical experience, drawing on the philosophy of Maurice Merleau-Ponty. The central argument holds that the enduring therapeutic benefits of psychedelics emerge through a radical transformation in the structure of lived experience. Two prominent objections to the therapeutic value of mystical experience are examined and shown to rest on implicit Cartesian assumptions that obscure the embodied, situated character of human experience. Using Merleau-Ponty's concepts of the body-subject and the i

Functional network collapse in neurodegenerative disease.

Cognitive and behavioral deficits in Alzheimer's disease (AD) and frontotemporal dementia (FTD) arise alongside gray matter atrophy and altered functional connectivity, yet the structure-function relationship across the dementia spectrum remains unclear. Here we combine structural and functional MRI from 221 patients-AD (n = 82), behavioral variant FTD (n = 41), corticobasal syndrome (n = 27), and nonfluent (n = 34) or semantic (n = 37) variant primary progressive aphasia-and 100 cognitively normal individuals. Partial least-squares regression reveals three structure-function components. Compo

Antimycin A-Induced Mitochondrial Damage Causes Human RPE Cell Death despite Activation of Autophagy.

Mitochondrial dysfunction has been implicated in a wide variety of degenerative diseases, including age-related macular degeneration. Damage to mitochondria and mitochondrial DNA accumulates with age in the postmitotic retinal pigment epithelium (RPE), which could lead to RPE cell death and trigger disease. One possible mechanism for cells to avoid cell death is mitophagy, the targeted clearance of damaged mitochondria by autophagy. Here, we induced mitochondrial damage in human RPE cells (ARPE-19 and hRPE), using antimycin A, an inhibitor of complex III of the electron transport chain, and in

Trends in Medical and Nonmedical Prescription Drug Use: 2005 to 2024.

Introduction  Acquired tracheomalacia (ATM) is characterized by a loss of structural strength of the tracheal framework, resulting in airway collapse during breathing. Near half of the patients undergoing prolonged invasive mechanical ventilation will suffer tracheal lesions. Treatment for ATM includes external splinting with rib grafts, prosthetic materials, and tracheal resection. Failure in the use of prosthetic materials has made reconsidering natural origin scaffolds and tissue engineering as a suitable alternative. Objective  To restore adequate airway patency in an ovine model with surg

Quantifying Neuromuscular and Pressure Force Dynamics in Obstructive Sleep Apnea: A Novel Computational Fluid Dynamics Approach Using Airway Wall Acceleration.

Obstructive sleep apnea (OSA) is characterized by recurrent upper airway collapse during sleep, resulting from interactions between aerodynamic forces, and neuromuscular activation of structures surrounding the airway. This study introduces a novel methodology for inferring the neuromuscular activity noninvasively from airway wall acceleration. The results will allow identification of the triggers for movements such as genioglossus advancement and to assess why they fail in patients with OSA. A patient with OSA underwent magnetic resonance imaging (MRI) to capture airway anatomy and motion und

Neuromechanical control of the isolated upper airway of mice.

We characterized the passive structural and active neuromuscular control of pharyngeal collapsibility in mice and hypothesized that pharyngeal collapsibility, which is elevated by anatomic loads, is reduced by active neuromuscular responses to airflow obstruction. To address this hypothesis, we examined the dynamic control of upper airway function in the isolated upper airway of anesthetized C57BL/6J mice. Pressures were lowered downstream and upstream to the upper airway to induce inspiratory airflow limitation and critical closure of the upper airway, respectively. After hyperventilating the
