Effects of haloperidol and chlorpromazine on smooth muscle contractility, platelet aggregation and neuronal calcium current.

Effects of chlorpromazine, haloperidol (neuroleptics and calmodulin antagonists), and verapamil on rat platelet aggregation induced by thrombin, on calcium current in snail neurones and on both tonic tension of high potassium contracture and phasic contraction of isolated guinea-pig ureter preparations were studied. Moreover, droperidol, sulpiride and prazosine effects were studied for models of phasic contractility and platelet aggregation. Sulpiride and prazosine were ineffective, verapamil was ineffective on platelet aggregation, while droperidol was the most potent inhibitor of platelet ag…

Distal colon motility in schizophrenic patients.

Although the dopaminergic blocking agents (DBA) haloperidol and sulpiride strongly inhibit distal colon motility in most nonpsychotic subjects (83 per cent), this effect was registered in only 10 per cent of the 30 schizophrenic patients investigated in the present study. In these cases, only sulpiride (an "atypical" DBA) displayed distal colon motility inhibition in schizophrenic subjects. When haloperidol (a "classical" DBA) produced any modification (in 23.3 per cent), this was rather in the nature of an increase in motility. All these cases showed low or absent distal colon motility during…

Correlation between antiavoidance activities of antipsychotic drugs in rats and daily clinical doses.

Effects of oral antipsychotic drugs, 12 phenothiazines, 3 thioxanthenes, 5 butyrophenones and 8 other derivatives on Sidman and discriminated avoidance responses in rats were investigated and compared to their clinical doses routinely used PO. Almost all drugs except sulpiride and clozapine suppressed the avoidance responses with a dose-dependent decrease in the response rate (lever-pressing) and increase in the shock rate in the Sidman avoidance performance or a decrease in both the response and avoidance rates in the discriminated one. Sulpiride (80-640 mg/kg) produced no marked change in th…

Neuroleptics up-regulate adenosine A2a receptors in rat striatum: implications for the mechanism and the treatment of tardive dyskinesia.

Neuroleptics, which are potent dopamine receptor antagonists, are used to treat psychosis. In the striatum, dopamine subtype-2 (D2) receptors interact with high-affinity adenosine subtype-2 (A2a) receptors. To examine the effect of various neuroleptics on the major subtypes of striatal dopamine and adenosine receptors, rats received 28 daily intraperitoneal injections of these drugs. Haloperidol (1.5 mg/kg/day) increased the density of striatal D2 receptors by 24% without changing their affinity for [3H]sulpiride. Haloperidol increased the density of striatal A2a receptors by 33% (control, 522…

Long-term efficacy of electroconvulsive therapy combined with different antipsychotic drugs in previously resistant schizophrenia.

BACKGROUND: In treatment-resistant schizophrenia a combination of ECT with antipsychotics has been reported to have superior outcomes compared to other strategies, however the results were inconsistent. We investigated the long-term effects of the combination of unilateral, non-dominant hemisphere ECT with three antipsychotics. SUBJECTS AND METHODS: The clinical study was a naturalistic, prospective, open-labeled, active-controlled study in adult outpatients of both genders suffering from treatment-resistant schizophrenia with a follow up of 2 years. The patients received sulpiride (n=17, 100…

[Paroxetine-induced hyponatremia in an elderly man due to the syndrome of inappropriate secretion of antidiuretic hormone].

We report an 82-year old man prescribed paroxetine who had hyponatremia and in whom the syndrome of inappropriate secretion of antidiuretic hormone was diagnosed. He had taken sulpiride for depressed mental status. However, he showed parkinsonism, which was an adverse effect from the treatment of sulpiride. Therefore sulpiride was changed to selective serotonin reuptake inhibitor, paroxetine 10mg daily. His depressed mental status deteriorated after paroxetine treatment started. His depression had not lessened after 12 days, and the dosage was increased to 20mg daily. On the 15th day after sta…

The Involvement of D1 and D2 Dopamine Receptors in the Restoration Effect of Left Frontal Anodal, but not Cathodal, tDCS on Streptozocin-Induced Amnesia.

BACKGROUND: Transcranial direct current stimulation (tDCS) is a non-invasive method that improves learning and memory. In this study, the effect of tDCS on streptozotocin (STZ) induced amnesia in the presence or absence of SCH23390 (D1 dopamine receptor antagonist) and sulpiride (dopamine D2 receptor antagonist) has been investigated in male Wistar rats. METHODS: Passive avoidance memory, locomotor activity and pain perception have been assessed by step-through, open-field and hot-plate instruments, respectively. Anodal and cathodal tDCS were exerted on the left frontal cortex with an intensi…

Blocking D2/D3 dopamine receptors in male participants increases volatility of beliefs when learning to trust others.

The ability to learn about other people is crucial for human social functioning. Dopamine has been proposed to regulate the precision of beliefs, but direct behavioural evidence of this is lacking. In this study, we investigate how a high dose of the D2/D3 dopamine receptor antagonist sulpiride impacts learning about other people's prosocial attitudes in a repeated Trust game. Using a Bayesian model of belief updating, we show that in a sample of 76 male participants sulpiride increases the volatility of beliefs, which leads to higher precision weights on prediction errors. This effect is driv…

Influence of neuroleptics on cytotoxic activity of macrophages in rats.

The aim of the study was to evaluate the influence of neuroleptics on the in vivo and in vitro activities of rat spleen macrophages. In the in vivo study, three neuroleptics (chlorpromazine, haloperidol, and sulpiride) were given once, for 14 or 28 days. In the in vitro study, we evaluated the effects of two different concentrations of the neuroleptics on 3-day cultures of spleen macrophages. Rat spleen macrophages were isolated by the adherence method, and their cytotoxic activity was determined by measuring 51 Cr release from target cells P-815. In the in vitro study, both concentrations of …
