In situ analysis of neuronal dynamics and positional cues in the patterning of nerve connections.

Recently developed imaging techniques permit individual cells to be uniquely labeled and followed over time as development proceeds in intact vertebrate embryos. Small groups of cells in the developing eye rudiment of the frog Xenopus have been labeled with the vital dyes DiI, lysinated fluorescein dextran (LFD) or lysinated rhodamine dextran (LRD). Individual optic axons and their growth cones were clearly visible in the intact living animal using confocal microscopy or epifluorescence microscopy with a low light level video camera and computer-based video image enhancement. To follow the dyn

A comparison of the initial retinal ganglion cell projection to the contralateral superior colliculus in albino and pigmented rats.

Newborn albino and pigmented rats received localised Fast blue (FB) injections into the most caudal part of the contralateral superior colliculus (SC). A proportion of retinal ganglion cells (RGC's) from the temporal retina which in the adult projects exclusively to the rostral half of the colliculus are labelled by injections to the caudal colliculus in neonatal animals. The majority of these cells die during the period of naturally occurring cell death in the retina, which occurs during the first 10 postnatal days. The object of this experiment was to see whether albino rats, which have well

[A study of visual function in congenital nystagmus--mechanism of visual improvement after stereotactic superior colliculectomy].

The clinical features of congenital nystagmus (CN) were studied statistically in 106 cases of CN. The point of the nystagmus at which the patients could best see the targets was detected in some patients. The effects of superior colliculectomy on their visual disturbance and the mechanism will be discussed. The study population comprises 106 patients, 79 males and 27 females, aged from one to 64 (mean 19.4 years). Patients with jerky type classified on ENG were found in 53 cases (50%), pendular type in 39 cases (37%), and mixed type in 14 cases (13%). Patients with jerky type showed significan

Peripheral nerve grafts to the frog optic tectum: a morphological study of foreign axon regeneration in the central nervous system.

The proximal stump of a transected mandibular nerve was grafted onto the rostrodorsal surface of the optic tectum in adult Rana pipiens to investigate the morphologic characteristics of nonspecific axonal regeneration in a highly organized region of central nervous system (CNS). Within the first 3 weeks postgraft surgery (WPS), the nerve-tectum interface became firmly established. Concomitant with this was an invasion of the host tectum by a small number of fine "pioneerlike" axons from the nerve. By 6 WPS there developed a concerted instreaming of a large number of peripheral fibers. Once wit

Cerebellar targets of visual pontine cells in the cat.

The cerebellum receives visual mossy fiber input from the cerebral cortex via visual cells in the pons. We identified the regions of cat cerebellum that receive cerebral visual input by injecting orthograde tracers among physiologically identified visual pontine cells. Cerebellar labeling following these injections indicates that the contralateral paraflocculus and the rostral folium of the uvula (vermal lobule IX) receive the heaviest projection from cortically activated pontine visual cells. Lighter visual input reaches much of the rest of the contralateral posterior lobe. A second experimen

Neurons with complex visual properties in the superior colliculus of the macaque monkey.

Single neurons were recorded from the superficial layers of the superior colliculus of immobilized monkeys (Macaca mulatta and Macaca irus). Two main functional types of neurons were found. The neurons of the first type (Type I neurons) responded well to simple stationary and moving stimuli such as spots, bars or slits of light. The latency of their response was 41 +/- 6 ms. They were not directionally selective and responded to a large range of velocities. The neurons of the second type (Type II neurons) responded very poorly to simple visual stimuli and their activation required real objects

Seaward orientation of hatchling turtles: turning systems in the optic tectum.

This paper reports studies on the mechanisms underlying seaward orientation in hatchling turtles. The particular aim was to investigate whether activity in different regions of the retina and associated tectal areas, as assessed by some comparator mechanism, results in turning in different directions. Hatchling green turtles (Chelonia mydas) were tested for sea-finding ability in a natural situation on the beach following lesions of the optic tectum. Asymmetrical bilateral lesions resulted in a number of turtles making circles in the direction of the tectum with the posterior lesion and in oth

Conservation of Neuron-Astrocyte Correlated Activity in Developing Sensory Pathways.

Neurons in developing sensory organs exhibit prolonged burst firing before the onset of sensory experience. This activity promotes neuronal survival and maturation in central sensory pathways. Within the auditory system, periodic bursts of synaptic glutamate release activate metabotropic glutamate receptors (mGluRs) on astrocytes, resulting in spatially and temporally correlated calcium transients; however, whether this phenomenon occurs in other sensory modalities is unknown. Using in vivo calcium imaging in the midbrain of awake mouse pups before eyelid opening, we show that retina wave-indu

Conserved Molecular Markers for Tectorecipient Subdivisions of the Visual Thalamus in Mice and Tree Shrews.

Sensory systems share common circuit organization motifs across mammalian species; however, anatomical subdivisions show varying degrees of complexity depending on ecological niche and species-specific sensory requirements. While neuroanatomical connections seem preserved within the visual system, it remains unknown if molecularly defined cell-types share a similar degree of conservation, regarding not only their functional properties but also connectivity. Here we analyze the organization, molecular marker expression, and connections between two prominent visual centers, the superior collicul

Optogenetic activation of the superior colliculus attenuates spontaneous seizures in the pilocarpine model of temporal lobe epilepsy.

OBJECTIVE: Decades of studies have indicated that activation of the deep and intermediate layers of the superior colliculus can suppress seizures in a wide range of experimental models of epilepsy. However, prior studies have not examined efficacy against spontaneous limbic seizures. The present study aimed to address this gap through chronic optogenetic activation of the superior colliculus in the pilocarpine model of temporal lobe epilepsy. METHODS: Sprague Dawley rats underwent pilocarpine-induced status epilepticus and were maintained until the onset of spontaneous seizures. Virus coding

Transcriptomic encoding of sensorimotor transformation in the midbrain.

Sensorimotor transformation, a process that converts sensory stimuli into motor actions, is critical for the brain to initiate behaviors. Although the circuitry involved in sensorimotor transformation has been well delineated, the molecular logic behind this process remains poorly understood. Here, we performed high-throughput and circuit-specific single-cell transcriptomic analyses of neurons in the superior colliculus (SC), a midbrain structure implicated in early sensorimotor transformation. We found that SC neurons in distinct laminae expressed discrete marker genes. Of particular interest

The neural instantiation of a priority map.

The term priority map is commonly used to describe a map of the visual scene, in which objects and locations are represented by their attentional priority, which itself is a combination of low-level salience and top-down control. The aim of this review is to examine how such a map may be represented at the neuronal level. We propose that there is not a single, common map in the brain, but that a number of cortical areas work together to generate the resultant behavior. Specifically, we suggest that the lateral intraparietal area (LIP) of posterior parietal cortex provides a simple representati

Reduced Frequency of Ipsilateral Express Saccades in Cervical Dystonia: Probing the Nigro-Tectal Pathway.

BACKGROUND: Cervical dystonia is a hyperkinetic movement disorder of unknown cause. Symptoms of cervical dystonia have been induced in animals in which the integrity of the nigro-tectal pathway is disrupted, resulting in reduced inhibition of the deep layers of the superior colliculus. This same pathway is believed to play a critical role in saccade generation, particularly visually guided, express saccades. It was hypothesized that individuals with cervical dystonia would present with a higher frequency of express saccades and more directional errors. METHODS: Eight individuals with cervical

Successful optic nerve regeneration in the senescent zebrafish despite age-related decline of cell intrinsic and extrinsic response processes.

Dysfunction of the central nervous system (CNS) in neurodegenerative diseases or after brain lesions seriously affects life quality of a growing number of elderly, since the adult CNS lacks the capacity to replace or repair damaged neurons. Despite intensive research efforts, full functional recovery after CNS disease and/or injury remains challenging, especially in an aging environment. As such, there is a rising need for an aging model in which the impact of aging on successful regeneration can be studied. Here, we introduce the senescent zebrafish retinotectal system as a valuable model to

Electrical stimulation of superior colliculus affects strabismus angle in monkey models for strabismus.

Disruption of binocular vision during the critical period for development leads to eye misalignment in humans and in monkey models. We have previously suggested that disruption within a vergence circuit could be the neural basis for strabismus. Electrical stimulation in the rostral superior colliculus (rSC) leads to vergence eye movements in normal monkeys. Therefore, the purpose of this study was to investigate the effect of SC stimulation on eye misalignment in strabismic monkeys. Electrical stimulation was delivered to 51 sites in the intermediate and deep layers of the SC (400 Hz, 0.5-s du

Neuronal Response Gain Enhancement prior to Microsaccades.

Neuronal response gain enhancement is a classic signature of the allocation of covert visual attention without eye movements. However, microsaccades continuously occur during gaze fixation. Because these tiny eye movements are preceded by motor preparatory signals well before they are triggered, it may be the case that a corollary of such signals may cause enhancement, even without attentional cueing. In six different macaque monkeys and two different brain areas previously implicated in covert visual attention (superior colliculus and frontal eye fields), we show neuronal response gain enhanc

Allometric scaling of the optic tectum in cartilaginous fishes.

In cartilaginous fishes (Chondrichthyes; sharks, skates and rays (batoids), and holocephalans), the midbrain or mesencephalon can be divided into two parts, the dorsal tectum mesencephali or optic tectum (analogous to the superior colliculus of mammals) and the ventral tegmentum mesencephali. Very little is known about interspecific variation in the relative size and organization of the components of the mesencephalon in these fishes. This study examined the relative development of the optic tectum and the tegmentum in 75 chondrichthyan species representing 32 families. This study also provide

Neuroanatomical study on the tecto-suprageniculate-dorsal auditory cortex pathway in the rat.

Previous anatomical and physiological studies suggest that the superior colliculus sends integrated sensory information to the multimodal cortical areas via the thalamic suprageniculate nucleus (SG). However, the detailed distribution of rat tecto-SG axon terminals and SG neurons projecting to the multimodal cortex, as well as synaptic connections between these tectal axons and SG neurons, remains unclear. In this study, the organization of the tecto-thalamo-cortical pathway was investigated via combined injections of anterograde and retrograde tracers followed by light and electron microscopi
