The T lymphocytes, OKT4-depleted T lymphocytes, monocytes, macrophages and the bone marrow early erythroid progenitor (BFU-E) interactions have been evaluated in normal subjects and in uremic patients in predialysis phase (PDP), on hemodialysis (HD) or continuous ambulatory peritoneal dialysis (CAPD). In the same subjects the T-cell growth factor (TCGF) activity and the T-cell subset identification have also been assessed. The results show in uremic patients, particularly in the presence of reduced OKT4 subset and TCGF activity, a blockage of the interactions between immunocompetent and bone m

To study the pathogenesis of tropical splenomegaly syndrome, we compared immunologic findings in patients from Flores, Indonesia, with those obtained in local residents without splenomegaly and in controls. Villagers with tropical splenomegaly syndrome had markedly elevated levels of total IgM, higher titers of IgM antibodies to Plasmodium vivax, and reduced levels of circulating T lymphocytes. The latter were caused by a decrease in the total number of T cells with the suppressor/cytotoxic phenotype (T8+). Levels of B lymphocytes were similar in all groups. All immunologic abnormalities rever

Five biopsy specimens of skin, four of lung, and one of a lymph node were taken from nine patients with sarcoidosis. Monoclonal antibodies were applied to frozen sections of the specimens by an immunoperoxidase technique to test for the presence and distribution of subsets of T-lymphocytes. T-cells expressing the suppressor/cytotoxic phenotype were found predominantly in lymphocytic mantles surrounding sarcoidal granulomas, whereas cells displaying the helper/inducer phenotype were distributed diffusely throughout granulomas. The ratio of helper to suppressor phenotypes in cutaneous sarcoidosi

Unstable chromosome aberrations induced by in vitro irradiation with D-T neutrons have been analyzed in human blood lymphocytes. With respect to 250 kVp X rays a maximum limiting RBE at low doses of 4.1 was obtained for dicentric aberrations. Using aberrations as markers in mixed cultures of irradiated and unirradiated cells permits an assessment of interphase death plus mitotic delay. The low-dose RBE for this effect is 2.5. Assuming all unstable aberrations observed at metaphase would lead to cell death by nondisjunction allows an assessment of mitotic death. The low-dose RBE for this effect

The pathophysiology of autoimmune arthritis was studied by selecting and isolating lines of effector T lymphocytes from rats administered an arthritogenic dose of Mycobacterium tuberculosis in complete Freund's adjuvant to induce adjuvant arthritis. Irradiated rats were intravenously inoculated with a cell line characterized by proliferative reactivity to Mycobacterium tuberculosis and, to a lesser degree, to rat collagen type II. This produced arthritis in all the irradiated rats. Nonirradiated recipients failed to develop arthritis. However, such rats, and those recovering from cell-mediated

The study of the ontogenesis of immunity in man is still in progress and development. In this paper the two major subpopulations of human lymphocytes--T (thymus dependent) and B (bursal equivalent or thymus independent) were examined during the ontogenetic development of man. These subpopulations can be examined in vitro by the methods distinguishing superficial receptors on these cells--namely by the rosseting methods. B lymphocytes with the receptor for Fc of immunoglobulin can be distinguished by the EA test, and with receptor for C3 by the the EAC test. T lymphocytes can be detected in blo

A longitudinal study was carried out in 40 patients with typhoid fever, 21 with infectious hepatitis and 15 with amebic liver abscess in order to determine percentual values of lymphocytes T and B in relation with age and nutritional state as compared to a normal control group. In patients with typhoid fever, a decrease of percentual values of lymphocytes T was observed starting on the second week of evolution with intensification on the third week, predominating in patients less than seven years of age and in malnourished cases (p less than 0.001) and late recovery in the group of patients le

Following stimulation with autologous or allogeneic lymphoblastoid cell lines (LCL) human T lymphocytes acquire two properties which suggest that LCL act as polyclonal activators. Cytotoxic activity, which has an antigen-specific component, is produced towards normal lymphocytes and LCL, and the cells become capable of mounting proliferative responses to antigens on human B lymphocytes which have accelerated secondary-type kinetics. Only weak responses to autologous cells occur. In addition, repeated restimulation with the original LCL leads to a progressive increase in the number of cells in

Agammaglobulinemia was diagnosed in a 1-year-old Thoroughbred horse on the basis of the following observations: (1) absence of serum immunoglobulins M, A, and G(T); (2) small amounts of serum immunoglobulin G (16 mg/100 ml); (3) absence of specific antibody in the serum of the horse following immunization and challenge exposure to 2 antigens; (4) absence of plasma cells, primary follicles, and germinal centers in a lymph node removed after antigenic stimulation; (5) absence of "natural" serum antibodies to rabbit-erythrocytes which were easily detectable in age-matched control horse serums; an

Cytotoxic T lymphocytes (CTLs) are cells of the adaptive immune system that are able to recognise and kill cancerous or virally infected target cells. The biochemical basis of CTL-mediated killing has been examined in great detail. Recent work has underscored the importance of physical forces in T cell function too, but how CTLs sense and exert force during migration and killing is not fully understood. Here, by expressing actin conformation probes based on the CH domain of utrophin, we directly visualize regions of altered F-actin conformation in primary CTLs during migration and killing. By

INTRODUCTION: Double-negative T lymphocytes with αβ TCR (CD3+CD4-CD8-TCRαβ+) are a population of T cells present in low proportions in the blood under normal conditions. They can be abnormally elevated in various pathological situations.
CASE REPORT: A 64-year-old female patient was hospitalized due to a general deterioration of health. Laboratory tests revealed a monoclonal proliferation of double-negative αβ T lymphocytes, representing 28% of T cells (219/mm3). A thoraco-abdomino-pelvic CT scan revealed two suspicious intrahepatic masses, multiple enlarged lymph nodes, and peritoneal nodule

The central nervous system (CNS) has been considered an immunologically privileged site. In the past few decades, research on inflammation in CNS diseases has mostly focused on microglia, innate immune cells that respond rapidly to injury and infection to maintain CNS homeostasis. Discoveries of lymphatic vessels within the dura mater and peripheral immune cells in the meningeal layer indicate that the peripheral immune system can monitor and intervene in the CNS. This review summarizes recent advances in the involvement of T lymphocytes in multiple CNS diseases, including brain injury, neurod

N6-methyladenosine (m6A) is a RNA modification that can regulate post-transcriptional processes including RNA stability, translation, splicing, and nuclear export. In CD4+ lymphocytes, m6A modifications have been demonstrated to play a role in early differentiation processes. The role of m6A in CD4+ T cell activation and effector function remains incompletely understood. To assess the role of m6A in CD4+ T lymphocyte activation and function, we assessed the transcriptome-wide m6A landscape of human primary CD4+ T cells by methylated RNA immunoprecipitation sequencing. Stimulation of the T cell

The immune system is traditionally classified as a defense system that can discriminate between self and non-self or dangerous and non-dangerous situations, unleashing a tolerogenic reaction or immune response. These activities are mainly coordinated by the interaction between innate and adaptive cells that act together to eliminate harmful stimuli and keep tissue healthy. However, healthy tissue is not always the end point of an immune response. Much evidence has been accumulated over the years, showing that the immune system has complex, diversified, and integrated functions that converge to

The role of abnormal epigenetic modifications, particularly DNA methylation, in the pathogenesis of autoimmune rheumatic diseases (ARDs) has garnered increasing attention. Lymphocyte dysfunction is a significant contributor to the pathogenesis of ARDs. Methylation is crucial for maintaining normal immune system function, and aberrant methylation can hinder lymphocyte differentiation, resulting in functional abnormalities that disrupt immune tolerance, leading to the excessive expression of inflammatory cytokines, thereby exacerbating the onset and progression of ARDs. Recent studies suggest th

CD4+ cytotoxic T lymphocytes (CTLs) were recently implicated in immune-mediated inflammation and fibrosis progression of Graves' orbitopathy (GO). However, little is known about therapeutic targeting of CD4+ CTLs. Herein, we studied the effect of rapamycin, an approved mTOR complex 1 (mTORC1) inhibitor, in a GO mouse model, in vitro, and in patients with refractory GO. In the adenovirus-induced model, rapamycin significantly decreased the incidence of GO. This was accompanied by the reduction of both CD4+ CTLs and the reduction of orbital inflammation, adipogenesis, and fibrosis. CD4+ CTLs fro

In the thymus, T cell progenitors differentiate in order to generate naive T lymphocytes which migrate in the periphery where they will fulfill their function in the adaptive immune response. During thymopoiesis, genomic alterations in thymocytes can promote leukemia development. Among recurrent alteration is PTEN inactivation, which is associated to MYC overexpression. Herein, we used conditional Pten and Myc knockout mouse models and single-cell RNA-sequencing approach, to investigate the impact of MYC loss on physio-pathological development of PTEN-proficient or PTEN-deficient T lymphocytes

T lymphocytes play a central role in antigen-specific immune responses. They modulate the function of different immune cells both through a direct contact (receptor binding) and through the secretion of cytokines. At the same time, they are deeply involved in the direct killing of aberrant target cells. T lymphocytes derive from a bone marrow precursor that migrates in the thymus where the main differentiation steps take place. Mature CD4 and CD8 single-positive cells, then, leave the thymus to reach the secondary lymphoid organs. T-cell subsets and their maturation steps can be identified mai

CAR-T cells are genetically modified human lymphocytes and gene therapy medicinal products. They are developed to treat cancers that express a membrane antigen targeted by the CAR. The FDA approved the two first-in-class medicinal products in 2017 and EMA in August 2018; both are autologous CAR-T cells targeting CD19 that is expressed at the surface of normal B-cells throughout their differentiation, and on B-cell lymphoid malignancies. Clinical efficacy was demonstrated for B-cell acute lymphoblastic leukemias, non-Hodgkin's lymphoma and chronic lymphocytic leukemia, although the marketing au
