Transformation patterns of masked fusarium mycotoxins during barley malting: Mechanistic evidence and multimodal detection in industrial products.

Malt, a key food industry ingredient, faces safety risks from Fusarium mycotoxins and masked derivatives. While malt's agricultural role is clear, biotransformation pathways of masked mycotoxins during barley malting remain poorly understood. This study assesses mycotoxin contamination in commercial malt and links fungal communities to mycotoxin metabolism. Deoxynivalenol dominated, detected in 55% beer and 70% malt samples, with low adult exposure risk. Using ultra performance liquid chromatography-tandem mass spectrometry and internal transcribed spacer sequencing, natural malting showed bip

Ablation of Gut Microbiota Alleviates DON-Induced Neurobehavioral Abnormalities and Brain Damage in Mice.

BACKGROUND: Deoxynivalenol (DON) poses a threat to animal and human health, particularly causing damage to the nervous system. Intestinal flora can regulate the nervous system through the gut-brain axis; however, there is currently a lack of evidence on the effect of changing the intestinal flora on the damage to the nervous system caused by DON. Therefore, this study aims to investigate the effect of gut microbiota ablation on neurotoxicity induced by exposure to deoxynivalenol. METHODS: One hundred-twenty (120) specific pathogen-free (SPF) male C57BL/6j mice were randomly divided into four

Mycotoxin-Induced Oxidative Stress and Its Impact on Human Folliculogenesis: Examining the Link to Reproductive Health.

Climate change has contributed to increased mycotoxin contamination in food systems, posing a growing threat to human health, including reproductive health. Our study aimed to investigate how mycotoxins entering the follicular fluid affect oxidative stress processes. We analyzed 88 follicular fluid samples from infertile patients for common mycotoxins, including deoxynivalenol (DON), zearalenone (ZEN), its main metabolite alpha-zearalenol (aZOL), and aflatoxin M1 (AfM1), and examined their relationship with oxidative stress markers (MDA, SOD, GPx, CAT, and TAOC) and hormones (cortisol, estradi

Deoxynivalenol induces spleen damage, apoptosis, and inflammation in mice by increasing mitochondrial reactive oxygen species: Protective effects of curcumin.

Deoxynivalenol (DON), a Fusarium mycotoxin, causes spleen apoptosis and inflammation, which damage the organ. Curcumin (Cur) is a member of the ginger family. It has anti-apoptotic and anti-inflammatory effects that maintain the health of the organism's immune system. Here, the protective effects of Cur against DON-induced spleen damage were explored. First, we found DON (2.4 mg/kg body weight) decreased the expression of manganese superoxide dismutase, mitochondrial membrane potential, adenosine triphosphate, and disturbed hematoxylin and eosin staining in mice spleen. The results confirmed t

Mitigation effects of plant carbon black on intestinal morphology, inflammation, antioxidant status, and microbiota in piglets challenged with deoxynivalenol.

INTRODUCTION: Plant carbon black (PCB) is a new feed additive for zearalenone adsorption in China. However, information regarding whether PCB can effectively absorb deoxynivalenol (DON) is limited. METHODS: To explore this research gap, the present study examined the adsorption effectiveness of DON by PCB using a phosphate buffer, artificial gastric juice, and artificial intestinal juice. In a 21-day in vivo trial, 48 male piglets were randomly assigned to four treatment groups: (1) uncontaminated basal diet (CTR), (2) basal diet supplemented with 1 mg/kg PCB(PCB), (3) 2.3 mg/kg DON-contamina

Alginate Oligosaccharides Enhance Gut Microbiota and Intestinal Barrier Function, Alleviating Host Damage Induced by Deoxynivalenol in Mice.

BACKGROUND: Alginate oligosaccharides (AOS) exhibits notable effects in terms of anti-inflammatory, antibacterial, and antioxidant properties. Deoxynivalenol (DON) has the potential to trigger intestinal inflammation by upregulating pro-inflammatory cytokines and apoptosis, thereby compromising the integrity of the intestinal barrier function and perturbing the balance of the gut microbiota. OBJECTIVES: We assessed the impact of AOS on mitigating DON-induced intestinal damage and systemic inflammation in mice. METHODS: After a 1-wk acclimatization period, the mice were divided into 4 groups.

Advances in Understanding Fusarium graminearum: Genes Involved in the Regulation of Sexual Development, Pathogenesis, and Deoxynivalenol Biosynthesis.

The wheat head blight disease caused by Fusarium graminearum is a major concern for food security and the health of both humans and animals. As a pathogenic microorganism, F. graminearum produces virulence factors during infection to increase pathogenicity, including various macromolecular and small molecular compounds. Among these virulence factors, secreted proteins and deoxynivalenol (DON) are important weapons for the expansion and colonization of F. graminearum. Besides the presence of virulence factors, sexual reproduction is also crucial for the infection process of F. graminearum and i

Chronic dietary deoxynivalenol exposure interferes the intestinal microbial community structure and antibiotic resistome in laying hens.

Antibiotic resistance genes (ARGs) are critical emerging pollutants that have attracted considerable attention. Deoxynivalenol (DON) is one of the most prevalent mycotoxins in cereal crops worldwide, arising severe health hazards to both humans and animals. Even if numerous researches argue in favor of a notorious influence of DON on the gut, the effects of dietary DON exposure on the ARG profile in poultry intestine remain obscure. In this study, two separate feeding experiments using Jing Tint 6 laying hens exposed to 4.5 or 9.0 mg/kg DON were performed to explore the impact of dietary DON e

Self-shedding MOF-nanocarriers modulated CdS/MoSe2 heterojunction activity through in-situ ion exchange: An enhanced split-type photoelectrochemical sensor for deoxynivalenol.

Deoxynivalenol (DON), a mycotoxin produced by Fusarium, poses a significant risk to human health and the environment. Therefore, the development of a highly sensitive and accurate detection method is essential to monitor the pollution situation. In response to this imperative, we have devised an advanced split-type photoelectrochemical (PEC) sensor for DON analysis, which leverages self-shedding MOF-nanocarriers to modulate the photoelectric response ability of PEC substrate. The PEC sensing interface was constructed using CdS/MoSe2 heterostructures, while the self-shedding copper peroxide nan

Deoxynivalenol induces testicular ferroptosis by regulating the Nrf2/System Xc-/GPX4 axis.

Deoxynivalenol (DON) is the most common mycotoxin contaminant in food and feed. DON accumulation in food chain severely threatens human and animal health due to the toxic effects on the reproduction system. However, the underlying mechanism of DON on male reproductive dysfunction is still in debate and there is little information about whether DON triggers testicular ferroptosis. In this study, male C57BL/6 mice were divided into 4 groups and treated by oral gavage with 0, 0.5, 1.0, 2.0 mg/kg BW DON for 28 days. Firstly, we proved that male reproduction dysfunction was induced by DON through a

Targeting the FXR network via Gdf15: Food matrix phytosterols attenuate deoxynivalenol gut toxicity, challenging single-contaminant risk paradigms.

Intestinal barrier integrity is critical for protection against environmental and foodborne hazardous stressors. This study investigated how adlay-derived phytosterols protect gut barrier function against environmental and foodborne contaminants through integrated network pharmacology and alternative animal model validation. Systematic screening identified eight bioactive compounds from adlay grains, with stigmasterol and beta-sitosterol exhibiting central roles in gastrointestinal regulatory networks. Notably, adlay exposure upregulated Gdf15, a key metabolic regulator, in human gut epithelia

Deoxynivalenol and male reproductive toxicity: Unraveling the hidden risks.

Deoxynivalenol (DON) is a highly prevalent mycotoxin known for its deleterious effects on the gastrointestinal and immune systems. Recent studies have also demonstrated its potential to impair reproductive function. This review compiles current evidence on the impact of DON on male reproductive organs, with particular emphasis on its hormonal suppressive effects, disruption of the germinal epithelium, and compromise of the blood-testis barrier. This review also highlights the impact of DON on less-explored reproductive organs, such as the epididymis, prostate, and seminal vesicles. These effec

Interactive effects of dietary deoxynivalenol and coccidial infection on growth performance, immune response, oxidative status, and gut health in pullets.

Deoxynivalenol (DON) contamination of grains and coccidiosis are two major health and economic challenges in layer production, yet their combined effect during pullet rearing phase remains poorly understood. This study evaluated whether dietary DON exposure exacerbates the severity of Eimeria infection and delays recovery in growing pullets. A total of 288 Hy-Line W36 pullets (4-wk-old) were fed either a control diet containing 2.5 mg/kg DON or a naturally contaminated diet containing 14-15 mg/kg DON for 14 d. Birds were then sham‑dosed or orally inoculated with E. acervulina (50 000 oocysts),

Necroptosis contributes to the intestinal toxicity of deoxynivalenol and is mediated by methyltransferase SETDB1.

Deoxynivalenol (DON) is a secondary metabolite produced by fungi, which causes serious health issues worldwide due to its widespread presence in human and animal diets. Necroptosis is a newly proposed cell death mode and has been proposed as a potential mechanism of intestinal disease. This study aimed to investigate the role of necroptosis in intestinal damage caused by DON exposure. Piglets were fed diets with or without 4 mg/kg DON for 3 weeks or given a gavage of 2 mg/kg BW DON or sterile saline to investigate the effects of chronic or acute DON exposure on the gut, respectively. Deoxynivalenol induces ovarian damage and uterine changes in prepubertal and adult mice.

Deoxynivalenol (DON) is associated with reproductive toxicity in animals. The frequent contamination of cereal-based foods with DON and the high intake of these by children raises particular concern about the susceptibility of this subpopulation to adverse effects from this mycotoxin. However, age-related differences in the in vivo reproductive toxicity of DON have not been evaluated. Therefore, the effects of DON on serum follicle-stimulating hormone (FSH) levels, histology, and inflammatory and oxidative stress responses in the ovaries and uteruses of prepubertal and adult mice were investig

The transcription factor FgCreA modulates ergosterol biosynthesis and sensitivity to DMI fungicides by regulating transcription of FgCyp51A and FgErg6A in Fusarium graminearum.

Fusarium head blight (FHB), caused by Fusarium graminearum, is a devastating disease that severely affects crop yield and quality worldwide. The catabolite responsive elements A (CreA) plays a critical role in numerous cellular processes in eukaryotes. In this study, we performed a functional characterization of CreA in F. graminearum. Deletion of FgCreA led to the transcriptional upregulation of FgCyp51A and FgErg6A, consequently resulting in elevated levels of ergosterol. This augmented ergosterol content was observed to improve cellular membrane integrity and diminished sensitivity to demet
