Proteoglycan depletion and size reduction in lesions of early grade chondromalacia of the patella.

OBJECTIVE: To determine the content and molecular size of proteoglycans (PGs) in patellar chondromalacia (CM) and control cartilages as a first step in investigating the role of matrix alterations in the pathogenesis of this disease. METHODS: Chondromalacia tissue from 10 patients was removed with a surgical knife. Using identical techniques, apparently healthy cartilage of the same site was obtained from 10 age matched cadavers (mean age 31 years in both groups). Additional pathological cartilage was collected from 67 patients with grades II-IV CM (classified according to Outerbridge) using

Composition and metabolism of the extracellular matrix in the periodontal ligament of impeded and unimpeded rat incisors.

Potential differences were studied in the composition and metabolism of the connective tissue components (collagen, non-collagenous proteins and glycosaminoglycans) of the periodontal ligament (PDL) as a result of alterations in the rate of eruption. The amount of collagen, glycosaminoglycan (GAG) or the water content of the tissue was the same in the unimpeded and impeded situation. The only difference was in the amount of structural non-collagenous proteins: 33 per cent increase in the unimpeded PDL. The rate of turnover of insoluble collagen, structural non-collagenous proteins and the sulp

Effects of Carboxymethyl Modification on the Acidic Polysaccharides from Calocybe indica: Physicochemical Properties, Antioxidant, Antitumor and Anticoagulant Activities.

An acidic polysaccharide fraction was obtained from Calocybe indica (CIP3a) after subjecting it to hot water extraction followed by purification through DEAE-cellulose 52 and Sepaharose 6B column chromatography. The CIP3a was further modified using chloroacetic acid to yield carboxymethylated derivatives (CMCIP3a). The modified polysaccharide was characterized using various spectroscopic methods. In addition, further antioxidant, antitumor and anticoagulant activities were also investigated. The polysaccharides CIP3a and CMCIP3a were heterogeneous in nature and composed of various molar percen

Bioaccumulation and detoxification mechanisms for lead uptake identified in Cosmos sulphureus Cav. seedlings.

Lead (Pb) stress disrupts plant growth and cellular homeostasis; however, the underlying multi-omics molecular network and Pb immobilization mechanisms in Cosmos sulphureus Cav. remain to be fully clarified. Here, Pb toxicity significantly reduced leaf area and biomass, while root morphology exhibited a dose-dependent biphasic response. C. sulphureus predominantly retains Pb in roots, supported by boosted ROS defenses (elevated SOD, CAT, MDA) and osmotic acclimation. We identified 400 μM as the physiological tolerance threshold, characterized by high metal retention and low systemic translocat

Understanding mercury accumulation in mosses of two subalpine forests in China.

The role of forest ecosystems in the global mercury (Hg) biogeochemical cycle is widely recognized; however, using litterfall as a surrogate to assess the Hg sink function of forests encounters limitations. We investigated the accumulation characteristics and influencing factors of Hg in mosses from two remote subalpine forests in southwestern China. The results indicated that there was high Hg accumulation in subalpine forest mosses, with average concentrations of 82 ± 49 ng g-1 for total mercury (THg) and 1.3 ± 0.8 ng g-1 for methylmercury (MeHg). We demonstrated that the accumulation capaci

Production of immunoregulatory polysaccharides from Crassostrea hongkongensis and their positive effects as a nutrition factor in modulating the effectiveness and toxicity of 5-FU chemotherapy in mice.

Chemotherapy is generally accompanied by undesirable side effects, such as immunosuppression and malnutrition, which reduce tolerance to cancer therapies. Prior studies have shown that immunonutrition improves the clinical outcomes of cancer patients. In this study, immunoregulatory polysaccharides from Crassostrea hongkongensis were included in a nutrition formula that was administered to S180 tumor-bearing mice in combination with 5-fluorouracil (5-FU) treatment. The C30-60% fraction of the polysaccharides was characterized as a branched polysaccharide, with a high amount of d-glucose (96.76

In vitro antiviral activities of sulfated polysaccharides from a marine microalga (Cochlodinium polykrikoides) against human immunodeficiency virus and other enveloped viruses.

A marine microalga, Cochlodinium polykrikoides, produces extracellular sulfated polysaccharides. Isolation and purification of the polysaccharides were accomplished by precipitation with ethanol and Cetavlon, followed by DEAE-cellulose column chromatography (polysaccharides A1 and A2). These polysaccharides, which were homogeneous when analysed by both ultracentrifugal and electrophoretic methods, were composed of mannose, galactose, glucose and uronic acid, together with sulfate groups (S = 7-8% w/w). Both A1 and A2 inhibited the cytopathic effect of influenza virus types A and B in MDCK cell

Sulfated polysaccharide from Apostichopus japonicus viscera exhibits anti-inflammatory properties in vitro and in vivo.

Polysaccharides from sea cucumbers are known for their biological activities, but little is known about those from sea cucumber viscera. The present study isolated a sulfated polysaccharide (SCVP-2) from the viscera of Apostichopus japonicas, which had a molecular weight of 209.1 kDa. SCVP-2 comprised 66.3 % total sugars, 2.1 % uronic acid, 4.5 % proteins, and 25.5 % sulfate groups, containing glucosamine, galactosamine, glucose, galactose, and fucose. FT-IR and NMR analyses identified SCVP-2 as a fucoidan sulfate with sulfation patterns of the fucose branches as Fuc2S, Fuc4S, and Fuc0S. SEM a

Comparison of chemical property and in vitro digestion behavior of polysaccharides from Auricularia polytricha mycelium and fruit body.

The antioxidant activity of Auricularia polytricha is associated tightly with its polysaccharide concentration, molar mass and architecture. This study aims to explore the differences in structural and physicochemical traits and oxidation resistances between the polysaccharides from fruit body (ABPs) and mycelial (IAPs) of Auricularia polytricha. The results showed that ABPs and IAPs were constituted by glucose, glucuronic acid, galactose and mannose. However, the molecular weight distribution of IAPs (3.22 × 104 Da (52.73%) and 1.95 × 106 Da (24.71%)) was wider than that of ABPs (5.4 × 106 Da
