Bat-rodent-human interfaces in the Americas: a synthesis of zoonotic pathways.

Emerging zoonoses often arise at wildlife-human interfaces, yet prevention efforts still treat wildlife reservoirs in isolation. This review synthesizes evidence on RNA viruses detected in bats and rodents across the Americas and examines the ecological interfaces underlying bat-rodent-human transmission. Rather than an exhaustive inventory, we adopt an interface-based perspective on cross-species transmission. We focus on astroviruses, coronaviruses, hantaviruses, and lyssaviruses-groups combining high genetic diversity, broad host ranges, and documented or suspected zoonotic potential. We de

Epidemiology and Clinical Presentation of West Nile Virus Infection in Horses in South Africa, 2016-2017.

Although West Nile virus (WNV) is endemic to South Africa (RSA), it has only become recognized as a significant cause of neurological disease in humans and horses locally in the past 2 decades, as it emerged globally. This article describes the epidemiological and clinical presentation of WNV in horses across RSA during 2016-2017. In total, 54 WNV-positive cases were identified by passive surveillance in horses with febrile and/or neurological signs at the Centre for Viral Zoonoses, University of Pretoria. They were followed up and compared to 120 randomly selected WNV-negative controls with t…

Virological Passive Surveillance of Avian Influenza and Arboviruses in Wild Birds: A Two-Year Study (2023-2024) in Lombardy, Italy.

Avian influenza (AI), caused by Alphainfluenzavirus (family Orthomyxoviridae), poses significant threats to poultry, biodiversity, and public health. AI outbreaks in poultry lead to severe economic losses, while highly pathogenic strains (HPAIVs) severely impact wild bird populations, with implications for biodiversity and potential zoonotic risks. Similarly, arboviruses such as West Nile virus (WNV) and Usutu virus (USUV) are emerging zoonoses. WNV can cause severe neurological diseases in birds, humans, and other animals, while USUV significantly affects blackbird populations and has zoonoti

Farmed fur animals harbour viruses with zoonotic spillover potential.

Animals such as raccoon dogs, mink and muskrats are farmed for fur and are sometimes used as food or medicinal products1,2, yet they are als … humans to farmed animals, indicating that fur farming represents an important transmission hub for viral zoonoses.

First Detection of Rocahepevirus in Urban Wastewater from Guinea: A One Health Alert.

Hepatitis E virus (HEV) is a major cause of acute viral hepatitis worldwide, with zoonotic genotypes detected in humans and animals. In Africa, limited data exist on environmental HEV circulation. Here, we report the first detection of Rocahepevirus ratti (RHEV) in urban wastewater from Conakry, Guinea. From December 2024 to April 2025, Rocahepevirus ratti (RHEV) has been detected in 35 out of 180 urban untreated wastewater samples in Conakry, Guinea. The phylogenetic analysis of partial HEV ORF1 genome segments reveals clustering with African rodent RHEV strains, highlighting environmental co

Genetic characterization of the marmot gut virome in high-altitude Qinghai Province and identification of novel viruses with zoonotic potential.

The high-altitude ecosystems of Qinghai Province, China, harbor unique viral communities shaped by extreme environmental conditions and host adaptations. This study presents a comprehensive genetic characterization of the marmot gut virome, revealing novel viral strains with zoonotic potential. Using viral metagenomics, we analyzed intestinal contents from 70 marmots collected from Chengduo and Maqin counties. Sequencing on the Illumina NovaSeq 6000 platform identified 19 viral genomes belonging to four major families: Adenoviridae, Astroviridae, Parvoviridae, and Picornaviridae, along with fo

SARS-CoV-2 natural infection in animals: a systematic review of studies and case reports and series.

COVID-19 pandemic is essentially a zoonotic disease. In this context, early in 2020, transmission from humans to certain animals began reporting; the number of studies has grown since. To estimate the pooled prevalence of SARS-CoV-2 natural infection in animals and to determine differences in prevalence between countries, years, animal types and diagnostic methods (RT-PCR or serological tests). A systematic literature review with meta-analysis using eight databases. Observational studies were included but analyzed separately. Rabies-infected dogs at slaughterhouses: A potential risk of rabies transmission via dog trading and butchering activities in Vietnam.

The study aimed to determine the status of neutralizing antibodies and rabies virus infection in dogs at slaughterhouses in Hanoi city from 2015 to 2017. A total of 2,376 pairs of blood and brain samples were collected from dogs at 92 slaughterhouses (8 large slaughterhouses and 84 small slaughterhouses) in 6 districts in Hanoi. Of the 1,500 dog samples from the large slaughterhouses where imported dogs were slaughtered, no dog brain samples were infected with rabies virus and no blood samples were positive for rabies neutralizing antibodies. Meanwhile, 7/876 (0.8%) of dog brain samples from s

Call for a paradigm shift in the design of universal influenza vaccines by harnessing multiple correlates of protection.

INTRODUCTION: The genetic variability and diversity of influenza viruses, and the expansion of their hosts, present a significant threat to human health. The development of a universal influenza vaccine is urgently needed to tackle seasonal epidemics, pandemics, vaccine mismatch, and zoonotic transmissions to humans. AREAS COVERED: Despite the identification of broadly neutralizing antibodies against influenza viruses, designing a universal influenza vaccine that induces such broadly neutralizing antibodies at protective levels in humans has remained challenging. Besides neutralizing antibodi

Vaccines and vaccination against tick-borne encephalitis.

Tick-borne encephalitis (TBE) is an emerging viral zoonosis and is endemic from Japan, China, Mongolia and Russia, to Central Europe and France. There is no specific treatment and TBE can be fatal. The four licensed prophylactic vaccines are produced according to WHO manufacturing requirements. Large clinical trials and postmarketing surveillance demonstrated safety and efficacy of the two European vaccines. The two Russian vaccines showed their effectiveness in daily use, but limited published data are available on controlled clinical trials. Vaccination recommendations in endemic areas vary

[Viral hemorrhagic fevers: what is the risk for travelers?].

The term hemorrhagic fever covers a number of diseases with different clinical and epidemiologic features. All these diseases are zoonoses but their occurrence is not confined to tropical areas. Some occur in polar zones. Travelers to endemic areas for these diseases are at risk of infection. There are two modes of transmission. Arthropods are vectors for some diseases such as yellow fever, dengue fever, and Rift Valley fever which are carried by mosquitos and Congo-Crimea virus which is carried by ticks. Airborne contamination by rodent excrement is responsible for transmission of hantaviruse

Nipah virus: pathogenesis, genome, diagnosis, and treatment.

The highly infectious Nipah virus (NiV) is classified under the Paramyxoviridae family and is categorized under the genus Henipavirus. NiV spreads to humans through zoonotic transmission from reservoir host bats and other intermediate hosts. It is highly contagious and has a high case fatality rate (CFR) of ~ 40-80%. Only sporadic outbreaks have been reported so far, but like SARS-CoV2, NiV has a high pandemic potential and has been put on the World Health Organization (WHO) priority pathogen list. Currently, no clinically approved antivirals, immunotherapy, or vaccines are available to tackle

Complete Genome Sequence Analysis of the First Imported Mpox Virus Clade Ib Variant in China.

Mpox, a zoonotic disease caused by the mpox virus (MPXV), has seen a significant shift in its epidemiological status since 2022, evolving from an initial local outbreak to a global epidemic. This recent outbreak of MPXV mainly emerged in several European and American countries and subsequently spread to over 100 countries and regions worldwide. The rapid evolution of MPXV, coupled with increased international interactions, has led to a gradual rise in mpox cases in certain regions of Asia, mostly involving MPXV clade II and its branch strains. In contrast, the more pathogenic and clinically se

Animal rabies in Mozambique: a retrospective study with focus on dog rabies and vaccination coverage.

Rabies, a highly preventable zoonotic disease, remains a major public health problem in Mozambique with approximately 50 human fatalities per annum due to dog-mediated rabies. This study analysed animal rabies cases and dog vaccination coverage, confirmed between 2001 and 2021, based on history, clinical signs, and/or diagnostic tests. During this period, 955 animal rabies cases were reported with the highest occurrence in Maputo (n = 283; 29.6%) and the lowest from Zambézia and Sofala provinces (n = 30; 3.1%). A significant number of animal rabies cases occurred in 2005 (n = 180; 18.8%). Most

SDAV, the Rat Coronavirus-How Much Do We Know about It in the Light of Potential Zoonoses.

Sialodacryoadenitis virus (SDAV) is known to be an etiological agent, causing infections in laboratory rats. Until now, its role has only been considered in studies on respiratory and salivary gland infections. The scant literature data, consisting mainly of papers from the last century, do not sufficiently address the topic of SDAV infections. The ongoing pandemic has demonstrated, once again, the role of the Coronaviridae family as extremely dangerous etiological agents of human zoonoses. The ability of coronaviruses to cross the species barrier and change to hosts commonly found in close pr

Influenza A Viruses: Understanding Human Host Determinants.

Previous influenza A virus (IAV) pandemics have invariably been caused by the introduction of an emergent IAV strain from an animal host into a human population with no or only little pre-existing immunity to the novel strain. Although zoonotic spillover of IAVs into humans can be associated with severe disease and a high fatality rate, these strains are typically poorly adapted to humans and are unable to establish sustained transmission between humans. Given the presumably very high degree of exposure to animal populations with endemic IAV, the number of pandemics remains surprisingly low. I
