Engineering the Maize Root Microbiome: A Rapid MoClo Toolkit and Identification of Potential Bacterial Chassis for Studying Plant-Microbe Interactions.

Sustainably enhancing crop production is a global necessity to meet the escalating demand for staple crops while sustainably managing their associated carbon/nitrogen inputs. Leveraging plant-associated microbiomes is a promising avenue for addressing this demand. However, studying these communities and engineering them for sustainable enhancement of crop production have remained a challenge due to limited genetic tools and methods. In this work, we detail the development of the Maize Root Microbiome ToolKit (MRMTK), a rapid Modular Cloning (MoClo) toolkit that only takes 2.5 h to generate des

Combining analyses of metabolite profiles and phosphorus fractions to explore high phosphorus utilization efficiency in maize.

Phosphorus (P) limitation is a significant factor restricting crop production in agricultural systems, and enhancing the internal P utilization efficiency (PUE) of crops plays an important role in ensuring sustainable P use in agriculture. To better understand how P is remobilized to affect crop growth, we first screened P-efficient (B73 and GEMS50) and P-inefficient (Liao5114) maize genotypes at the same shoot P content, and then analyzed P pools and performed non-targeted metabolomic analyses to explore changes in cellular P fractions and metabolites in maize genotypes with contrasting PUE.

All-cellulose air filter composed with regenerated nanocellulose prepared through a facile method with shear-induced.

High-speed shear system is usually used for the dispersion improvement of slurry, nanomaterials preparation, and even two-dimensional materials production. However, there is barely study that focused on the regenerated cellulose (RC) which was coagulated with shear induced. In this work, a new type of all-cellulose air filter was fabricated through high-speed shear in aqueous regeneration system using parenchyma cellulose from corn stalk. The obtained RC were aggregated by ribbon-like fine cellulose and nanocellulose sheets. The study exhibited the micro-structure of RC displayed excellent uni

Synthesis, Stability, and Kinetics of Hydrogen Sulfide Release of Dithiophosphates.

The development of chemicals to slowly release hydrogen sulfide would aid the survival of plants under environmental stressors as well as increase harvest yields. We report a series of dialkyldithiophosphates and disulfidedithiophosphates that slowly degrade to release hydrogen sulfide in the presence of water. Kinetics of the degradation of these chemicals were obtained at 85 °C and room temperature, and it was shown that the identity of the alkyl or sulfide group had a large impact on the rate of hydrolysis, and the rate constant varied by more than 104×. For example, using tert-butanol as t

Concerted impacts of antiherbivore defenses and opportunistic Serratia pathogens on the fall armyworm (Spodoptera frugiperda).

Insects frequently confront different microbial assemblages. Bacteria inhabiting an insect gut are often commensal, but some can become pathogenic when the insect is compromised from different stressors. Herbivores are often confronted by various forms of plant resistance, but how defenses generate opportunistic microbial infections from residents in the gut are not well understood. In this study, we evaluated the pathogenic tendencies of Serratia isolated from the digestive system of healthy fall armyworm larvae (Spodoptera frugiperda) and how it interfaces with plant defenses. We initially s

Popcorn-Derived Porous Carbon for Energy Storage and CO2 Capture.

Porous carbon materials have drawn tremendous attention due to its applications in energy storage, gas/water purification, catalyst support, and other important fields. However, producing high-performance carbons via a facile and efficient route is still a big challenge. Here we report the synthesis of microporous carbon materials by employing a steam-explosion method with subsequent potassium activation and carbonization of the obtained popcorn. The obtained carbon features a large specific surface area, high porosity, and doped nitrogen atoms. Using as an electrode material in supercapacitor

Formation of triploid plants via possible polyspermy.

Polyploidization is a common phenomenon in angiosperms, and polyploidy has played a major role in the long-term diversification and evolutionary success of plants. Triploid plants are considered as the intermediate stage in the formation of stable autotetraploid plants, and this pathway of tetraploid formation is known as the triploid bridge. As for the mechanism of triploid formation among diploid populations, fusion of an unreduced gamete with a reduced gamete is generally accepted. In addition, the possibility of polyspermy has been proposed for maize, wheat and some orchids, although it ha

Distribution of phosphorus compounds in corn processing.

Distillers dried grains with solubles (DDGS) and corn gluten feed (CGF) are major coproducts of ethanol production from corn dry grind and wet milling facilities, respectively. These coproducts contain important nutrients, nevertheless, high levels of phosphorus (P). About 50-80% of the P in these products is in an organically bound form known as phytate. The phytate P in these products cannot be digested by nonruminant animals. Consequently, large quantities of phytate are deposited into the soil with the animal wastes which potentially could cause P pollution in soil and underground water re

Separation of glucose and pentose sugars by selective enzyme hydrolysis of AFEX-treated corn fiber.

A process was developed to fractionate corn fiber into glucose- and pentose-rich fractions. Corn fiber was ammonia fiber explosion treated at 90 degrees C, using 1 g anhydrous ammonia pergram of drybiomass, 60% moisture, and 30-min residence time. Twenty four hour hydrolysis of ammonia fiber explosion-treated corn fiber with cellulase converted 83% of available glucanto-glucose. In this hydrolysis the hemicellulose was partially broken down with 81% of the xylan and 68% of the arabinan being contained in the hydrolysate after filtration to remove lignin and other insoluble material. Addition o

Nanohumic Acid Enhances Maize Yield by Modulating Soil Bacterial Communities and Grain Tryptophan Metabolism: A Novel Plant Biostimulant.

Maize (Zea mays L.) is a major staple crop worldwide and plays a critical role in food security. Although humic acid (HA) is widely used to improve crop productivity, its effectiveness is often limited by poor dispersibility and bioavailability. Herein, nanohumic acid (NHA) with improved dispersibility was synthesized, and its effects on maize yield and underlying mechanisms were evaluated. The results showed that NHA significantly promoted maize growth and increased yield, with the highest yield under NHA15 (15 mg/kg; 31.8 g/plant), exceeding HA15 (12.8 g/plant) and CK (6.8 g/plant). Maize yi…

β-Pinene moderates Cr(VI) phytotoxicity by quenching reactive oxygen species and altering antioxidant machinery in maize.

We examined the possible role of monoterpene β-pinene in providing protection against Cr(VI) toxicity in maize (Zea mays). Treatment with β-pinene (10 μM) significantly alleviated Cr(VI) accumulation and recuperated Cr(VI) caused decline in root and coleoptile growth in maize. β-Pinene addition caused a decline in Cr(VI)-induced accumulation of superoxide anion, hydroxyl ion, hydrogen peroxide and confirmed by in-situ detection of ROS using histochemical localization. It suggested that the β-pinene quenches/neutralizes enhanced ROS generated under Cr(VI) exposure. β-Pinene also reduced Cr(VI)-…

Disease resistance in maize and the role of molecular breeding in defending against global threat.

Diseases are a potential threat to global food security but plants have evolved an extensive array of methodologies to cope with the invading pathogens. Non-host resistance and quantitative resistance are broad spectrum forms of resistance, and all kinds of resistances are controlled by extremely diverse genes called "R-genes". R-genes follow different mechanisms to defend plants and PAMP-induced defenses in susceptible host plants are referred to as basal resistance. Genetic and phenotypic diversity are vital in maize (Zea mays L.); as such, genome wide association study (GWAS) along with cer…

Heavy-metal-responsive genes in maize: identification and comparison of their expression upon various forms of abiotic stress.

To identify genes involved in defense against heavy-metal stresses, a cDNA library originating from mercuric chloride-treated maize (Zea mays L. cv. INRA 258) leaves was constructed and analysed by differential screening using cDNAs derived from treated and untreated plants. Transcriptionally activated cDNA clones, designated CHEM (chemically-activated), were isolated and characterized. They represent various known proteins, such as glycine-rich proteins, pathogenesis-related proteins, chaperones and membrane proteins. The expression of the genes encoding these proteins was studied in maize su…

A novel metabolic form of the 32 kDa-D1 protein in the grana-localized reaction center of photosystem II.

Two forms of the 32 kDa-D1 reaction center protein of photosystem II (PSII), having slightly different mobilities on denaturing polyacrylamide gels, have been resolved in Spirodela oligorrhiza, Glycine max L., Gossypium hirsutum L., Triticum aestivum L., and Zea mays L. The protein band with faster mobility is identified as the 32 kDa-D1 protein, and the less mobile band as a novel form, designated 32*. The two forms are structurally similar based on immunological and partial proteolytic tests. 32* is associated exclusively with the grana and is present in the PSII reaction center. Temporally,…

Impact of Corn Earworm (Lepidoptera: Noctuidae) on Field Corn (Poales: Poaceae) Yield and Grain Quality.

Corn earworm, Helicoverpa zea (Boddie), commonly infests field corn, Zea mays (L.). The combination of corn plant biology, corn earworm behavior in corn ecosystems, and field corn value renders corn earworm management with foliar insecticides noneconomical. Corn technologies containing Bacillus thuringiensis (Bt) Berliner (Bacillales: Bacillaceae) were introduced that exhibit substantial efficacy against corn earworm and may reduce mycotoxin contamination in grain. The first generation Bt traits in field corn demonstrated limited activity on corn earworm feeding on grain. The pyramided corn te…

Impact of Simulated Corn Earworm (Lepidoptera: Noctuidae) Kernel Feeding on Field Corn Yield.

Corn earworm, Helicoverpa zea (Boddie) Lepidoptera: Noctuidae, has not been considered an economic pest of field corn. Historical losses estimates ranged from 1.5 to 2.5%, and a large number of foliar insecticide applications would be needed to minimize infestations. In recent years, Bt, Bacillus thuringiensis (Berliner) field corn, Zea mays (L.) Poales: Poaceae, technologies that exhibit activity against corn earworm have been introduced. However, it is unclear how much damage to corn ears (number of damaged kernels) is required to reduce yield. In this study manual damage methods were utiliz…
