The combined intravenous injection of TRH and GnRH elicited paradoxical responses of plasma beta-endorphin in active and successfully treated pituitary dependent Cushing's disease as well as in ectopic ACTH syndrome and in congenital adrenal hyperplasia. No response was observed in Cushing's syndrome due to adrenal tumours. It is concluded that an abnormal response to inappropriate releasing hormones cannot verify the existence of a pituitary corticotrophic microadenoma.

The inhibitory effect of beta-endorphin (EP) or other opioids on TSH secretion is, in contrast to their stimulating properties on PRL release, still a matter of debate. In the present study a dose of 1 microgram beta-EP injected intracerebroventricularly (IVT) in unstressed conscious male rats, though highly effective on PRL release, did not affect basal TSH levels, nor the TRH-induced TSH secretion. The previously reported inhibition of TSH release by opioids may therefore be an effect only seen when pharmacological doses are used.

Anti-neurophysin serum was applied in the immunohistochemical technique to anterior pituitary tissues obtained from normal and chronically dehydrated rats and also from rats with chronic diabetes insipidus (Brattleboro strain). In all cases there was a positive staining in the corticotrophs, which also stained for either beta-endorphin (beta-END) or adrenocorticotrophin hormone (ACTH). It was concluded that corticotroph-neurophysin may be synthesized independently of either ACTH or beta-END.

It has been hypothesized that the endogenous opioid (endorphin) system is related to biologic stress responses. We have used general surgery as a naturalistic model for studying stress response in humans. Abdominal surgery, but not anesthesia induction, was associated with significant increases in plasma beta-endorphin immunoreactivity and cortisol.
