Overexpression of apolipoprotein A-I alleviates endoplasmic reticulum stress in hepatocytes.

BACKGROUND: Abnormal lipid metabolism may contribute to an increase in endoplasmic reticulum (ER) stress, resulting in the pathogenesis of non-alcoholic steatohepatitis. Apolipoprotein A-I (apoA-I) accepts cellular free cholesterol and phospholipids transported by ATP-binding cassette transporter A1 to generate nascent high density lipoprotein particles. Previous studies have revealed that the overexpression of apoA-I alleviated hepatic lipid levels by modifying lipid transport. Here, we examined the effects of apoA-I overexpression on ER stress and genes involved in lipogenesis in both HepG2

Renal iron overload in rats with diabetic nephropathy.

Diabetic nephropathy (DN) remains incurable and is the main cause of end-stage renal disease. We approached the pathophysiology of DN with systems biology, and a comprehensive profile of renal transcripts was obtained with RNA-Seq in ZS (F1 hybrids of Zucker and spontaneously hypertensive heart failure) rats, a model of diabetic nephropathy. We included sham-operated lean control rats (LS), sham-operated diabetic (DS), and diabetic rats with induced renal ischemia (DI). Diabetic nephropathy in DI was accelerated by the single episode of renal ischemia. This progressive renal decline was associ

ASMase regulates autophagy and lysosomal membrane permeabilization and its inhibition prevents early stage non-alcoholic steatohepatitis.

BACKGROUND & AIMS: Acid sphingomyelinase (ASMase) is activated in non-alcoholic steatohepatitis (NASH). However, the contribution of ASMase to NASH is poorly understood and limited to hepatic steatosis and glucose metabolism. Here we examined the role of ASMase in high fat diet (HFD)-induced NASH. METHODS: Autophagy, endoplasmic reticulum (ER) stress and lysosomal membrane permeabilization (LMP) were determined in ASMase(-/-) mice fed a HFD. The impact of pharmacological ASMase inhibition on NASH was analyzed in wild type mice fed a HFD. RESULTS: ASMase deficiency determined resistance to he

Targeted eicosanoid lipidomics of exhaled breath condensate provide a distinct pattern in the aspirin-intolerant asthma phenotype.

BACKGROUND: Eicosanoids, important signaling and inflammatory molecules, are present in exhaled breath condensate (EBC) in very low concentrations, requiring highly sensitive analytic methods for their quantification. OBJECTIVE: We sought to assess a vast platform of eicosanoids in different asthma phenotypes, including aspirin-intolerant asthma, by means of a recently developed analytic approach based on mass spectrometry. METHODS: EBC from 115 adult asthmatic subjects (62 with aspirin intolerance) and 38 healthy control subjects were assessed quantitatively for 19 eicosanoids by using comp

Postprandial chylomicron clearance rate in late teenagers with diabetes mellitus type 1.

A delayed chylomicron (CM) clearance rate, a known risk factor for atherosclerosis, has been described in adults with diabetes type 1 (DM1). We determined the CM clearance rate in late teenagers with DM1, and the relationship between CM clearance rate and elevated plasma lipid concentrations in DM1 teenagers in poor metabolic control (as characterized by HbA(1c) percentage). Plasma lipids and CM clearance were determined in nine patients with DM1 (mean age +/- SD: 17.5 +/- 0.6 y) and four healthy controls (mean age +/- SD: 20.1 +/- 0.8 y), by measuring breath (13)CO(2), plasma triglyceride, re

Intensive insulin therapy and weight gain in IDDM.

Intensive insulin therapy is frequently complicated by excessive weight gain. The purpose of this study was to determine the cause and composition of this weight gain. Therefore, changes in body composition, energy expenditure, glycosuria, and substrate kinetics were evaluated in patients with IDDM who transferred from conventional insulin therapy to intensive insulin therapy. Six adult patients with IDDM were studied on conventional insulin therapy and after 2 mo of intensive insulin therapy while maintaining constant caloric intake and were compared with a group of 6 matched nondiabetic volu

DGAT1, but Not DGAT2, Is Required for Ovarian Lipid Storage and Egg Hatchability in Rhodnius prolixus.

Triacylglycerol (TAG) is the main lipid reserve in insect eggs and represents a critical energy source for embryonic development. In the hematophagous vector Rhodnius prolixus, oocyte growth relies heavily on lipids transported from the fat body through the hemolymph, which are delivered mainly as diacylglycerol and free fatty acids. Therefore, the ovary must catalyze the final esterification step converting these substrates into TAG to ensure proper lipid storage in developing oocytes. This reaction is mediated by diacylglycerol acyltransferases (DGATs), yet the specific roles of DGAT1 and DG

Comprehensive elucidation of volatile, phenolic, and organic acid profiles in Turkish mallow (Malva sylvestris L.) petal herbal tea: impact of diverse brewing conditions.

BACKGROUND: Mallow (Malva sylvestris L.) petals are traditionally consumed as herbal tea across the Mediterranean for their soothing and anti-inflammatory properties; however, scientific characterization of this consumption-relevant matrix under realistic brewing conditions remains scarce. This study comprehensively elucidates the impact of three household infusion protocols, short hot brewing (4 min at 98 °C), extended hot brewing (16 min at 98 °C), and cold brewing (24 h at 25 °C), on the volatile, phenolic, and organic acid profiles, antioxidant capacity, and sensory attributes of Turkish e

Downregulation of NK cell activities in Apolipoprotein C-III-induced hyperlipidemia resulting from lipid-induced metabolic reprogramming and crosstalk with lipid-laden dendritic cells.

OBJECTIVE: Apolipoprotein C-III (Apoc3) is a key component of triglyceride-rich lipoproteins (TRL). The Apoc3-transgenic mice are characterized by high levels of plasma triglyceride and free fatty acids (FFAs). Apoc3 stimulates human monocytes via activation of the NLRP3 inflammasome. Considering the NK cell downregulation in obese individuals and the possible stimulatory-effects of macrophages, variations of NK cell functions and underlying mechanisms were investigated in mice with Apoc3-induced hyperlipidemia. METHODS: Variations of activities and glycolipid metabolism in NK cells of the Ap

Characterization of leptospiral outer membrane lipoprotein LipL36: downregulation associated with late-log-phase growth and mammalian infection.

We report the cloning of the gene encoding a 36-kDa leptospiral outer membrane lipoprotein, designated LipL36. We obtained the N-terminal amino acid sequence of a staphylococcal V8 proteolytic-digest fragment in order to design an oligonucleotide probe. A Lambda-Zap II library containing EcoRI fragments of Leptospira kirschneri DNA was screened, and a 2.3-kb DNA fragment which contained the entire structural lipL36 gene was identified. Several lines of evidence indicate that LipL36 is lipid modified in a manner similar to that of LipL41, a leptospiral outer membrane lipoprotein we described in

Supercritical carbon dioxide extraction of lipids and carotenoids from Rhodotorula toruloides CBS 14 in comparison with conventional extraction methods.

BACKGROUND: Oil from oleaginous yeasts has the potential to replace non-sustainable vegetable oil as raw material to produce food, feed, biofuels, or biochemicals. Co-produced compounds like carotenoids may be helpful to obtain economically viable bioprocesses. Identifying appropriate extraction methods is a bottleneck both for establishing oleaginous yeasts as cell factories for both oil and carotenoids production and for analysis of intracellular compounds like lipids and carotenoids. We conducted extractions using supercritical carbon dioxide (SC-CO2) and conventional solvent methods to ext

Central composite design parameterization of microalgae/cyanobacteria co-culture pretreatment for enhanced lipid extraction using an external clamp-on ultrasonic transducer.

Lipids extracted from algal biomass could provide an abundant, rapidly growing, high yield feedstock for bio-diesel and other green fuels to supplement current fossil-based sources. Ultrasound pretreatment is a mechanical cell disruption method that has been shown to enhance lipid recovery from algae due to cavitation effects that disrupt algae cell walls. In this study, a locally grown mixture of Chlorella vulgaris/Cyanobacteria leptolyngbya was sonicated in an ultrasonic reactor with a clamp-on transducer prior to solvent lipid extraction. This configuration allows for a non-contact delivery

Salt-inducible kinase 2b (Sik2b) promotes liver inflammation by phosphorylating BHLHE41 in zebrafish (Danio rerio).

Long-term feeding of palm oil-based diets readily induces inflammatory response in farmed fish, compromising aquaculture efficiency and food safety. Salt-inducible kinase 2 (SIK2), an energy-sensing kinase that orchestrates immunometabolic crosstalk in mammals, may participate in this process. However, little attention has been given to the function of its ortholog in fish. Here, using sik2b knockout zebrafish models together with sik2b knockdown and overexpression experiments in ZFL cells, we found that loss of sik2b induced an anti-inflammatory phenotype in zebrafish hepatocytes, whereas its

Enhanced Bioactivity of Pyrolysis Liquid and AgNPs from Capparis sicula subsp. sicula: Chemical Characterization and Mechanistic Insights.

Recently, the insecticidal, antifungal, antibacterial, and antioxidant properties of pyrolysis liquid (bio-oil) obtained from agricultural and industrial plants have been investigated in foods. As no previous studies had been conducted on the Capparis sicula subsp. sicula (caper) species, this study was chosen. For the first time, a methanol-chloroform extract, pyrolysis liquid, and green-synthesized silver nanoparticles (AgNPs) were obtained from caper fruit. GC-MS/MS analysis of the extract and pyrolysis revealed linoleic and palmitic acid methyl esters. In contrast, LC-ESI-MS/MS analysis re

Design, synthesis and biological evaluation of quinazoline-carbazole derivatives as dual TXNIP/DYRK1A inhibitors for the treatment of type 2 diabetes mellitus.

TXNIP and DYRK1A are two novel drug targets for the treatment of type 2 diabetes (T2DM), and their respective inhibitors have been shown to suppress islet β-cell apoptosis and promote islet β-cell proliferation. The quinazoline scaffold constitutes the core pharmacophore of TXNIP inhibitors. Harmine, a prototypical DYRK1A inhibitor bearing a β-carboline scaffold, has demonstrated robust β-cell proliferative activity. Carbazole serves as a structurally simplified bioisostere of β-carboline. Guided by the principles of multi-target drug design and combinatorial chemistry, we designed and synthes

Synergistic effects of nicotine and hyperlipidemia induce cardiac damage via dynamic cardiomyocyte-based biosensing.

Cigarette smoking and hyperlipidemia are major risk factors for cardiovascular disease. However, the mechanisms underlying their synergistic cardiotoxic effects remain insufficiently elucidated, particularly at the cellular electrophysiological level. To address this, we developed an electrophysiological biosensing platform to establish a real-time co-exposure model that simulates the combined stress of nicotine and palmitic acid-induced hyperlipidemia on the primary rat cardiomyocytes. This model enabled continuous, non-invasive monitoring and quantitative analysis of field potential under in

Binaphthoquinone attenuates myofibroblast transition of human ventricular cardiac fibroblasts in vitro via mitochondrial modulation under diabetic conditions.

Binaphthoquinone (BiNQ), a dimeric compound composed of juglone (5-hydroxy-1,4-naphthoquinone) and naphthazarin (5,8-dihydroxy-1,4-naphthoquinone), was originally isolated from the sea urchin Spatangus purpureus O.F. Müller. In this study, we examined the anti-fibrotic effects of BiNQ in a diabetic context, focusing on its potential to enhance mitochondrial function. Human cardiac fibroblasts (NHCF-V) were cultured under control 5 mM glucose or exposed to 33 mM glucose plus palmitic acid (100 μM). Fibroblast-to-myofibroblast transition was evaluated by measuring the expression level of the myo
