Although the beneficial effects of probiotics in the prevention or treatment of metabolic disorders have been extensively researched, the precise mechanisms by which probiotics improve metabolic homeostasis are still not clear. Given that probiotics usually exert a comprehensive effect on multiple metabolic disorders, defining a concurrent mechanism underlying the multiple effects is critical to understand the function of probiotics. In this study, we identified the SIRT1-dependent or independent PGC-1α pathways in multiple organs that mediate the protective effects of a strain of Lactobacillu

Age-related macular degeneration is a major cause of vision impairment in the Western world among people of 55 years and older. Recently we have shown that autophagy is dysfunctional in the retinal pigment epithelium (RPE) of the AMD donor eyes (AMD RPE). We also showed increased reactive oxygen (ROS) production, increased cytoplasmic glycogen accumulation, mitochondrial dysfunction and disintegration, and enlarged and annular LAMP-1-positive organelles in AMD RPE. However, the underlying mechanisms inducing these abnormalities remain to be elucidated. Here, by performing a comprehensive study

Currently, no pharmacotherapy has been proven effective in treating photoreceptor degeneration in patients. Discovering readily available and safe neuroprotectants is therefore highly sought after. Here, we investigated nicotinamide mononucleotide (NMN), a precursor of nicotinamide adenine dinucleotide (NAD+), in a retinal detachment (RD) induced photoreceptor degeneration. NMN administration after RD resulted in a significant reduction of TUNEL+ photoreceptors, CD11b+ macrophages, and GFAP labeled glial activation; a normalization of protein carbonyl content (PCC), and a preservation of the o

Nucleolar protein 56 (NOP56), a core component of small nucleolar ribonucleoprotein complexes, has been implicated in oncogenesis through the regulation of reactive oxygen species (ROS) homeostasis; however, its role in colorectal cancer (CRC) remains unclear. Here, we investigated the clinical relevance and biological function of NOP56 in CRC using TCGA datasets, tissue microarrays, next-generation sequencing, and in vitro and in vivo models. NOP56 expression was markedly elevated in CRC tissues compared with adjacent normal tissues and was correlated with poor patient survival. Silencing of

BACKGROUND AND PURPOSE: Lumbar disc herniation (LDH) is a degenerative spine disease and the most common cause of lower back pain, with a high prevalence in younger patients. The use of acupuncture (AC) as a conservative treatment for LDH has received widespread attention at home and abroad. Therefore, the present study aimed to investigate the specific mechanism of AC in the treatment of LDH.
METHODS: In this study, we established an LDH rat model via autologous nucleus pulposus (NP) transplantation and treated human NP cells with 50 μM tert-butyl hydroperoxide (TBHP) for 24h to establish an

BACKGROUND: Axonal degeneration of motor neurons is a hallmark pathological feature of neurodegenerative diseases, culminating in motor dysfunction in advanced stages. Systemic inflammation exacerbates motor dysfunction, yet the relationship between systemic inflammation and motor axon degeneration remains poorly understood. We investigated whether macrophages impair motor axon morphology during systemic inflammation and the underlying mechanisms.
METHODS: A zebrafish systemic inflammation model was established using CuSO₄ exposure. In vivo confocal imaging examined spatiotemporal relationshi

Lupus nephritis (LN) is a critical complication of systemic lupus erythematosus (SLE) and a leading cause of mortality in those affected. Abnormal energy metabolism of macrophages is closely associated with LN pathogenesis. Adenosine diphosphate (ADP) is known to regulate macrophage proinflammatory function; however, its specific role in SLE progression remains unexplored. We employed a spontaneous SLE mouse model and an integrated transcriptomic and metabolomic approach to investigate the effect of ADP on SLE disease progression and macrophage function. Our study revealed that ADP stimulates

Chemotherapy-Induced Peripheral Neuropathy (CIPN) is a dose-limiting complication of paclitaxel (PTX) therapy, for which effective treatments are still lacking. This study evaluated the neuroprotective potential of Thymoquinone (TQ), a bioactive derivative of Nigella sativa, in mitigating chronic PTX-induced neurotoxicity without compromising antineoplastic activity. We first performed in vitro experiments using Sprague-Dawley rat embryonic (E15) Dorsal Root Ganglia (DRG) (Envigo Laboratory (Udine, Italy)) to assess neurotoxicity through neurite outgrowth evaluation. Ischemia and hypoxia near the injury site following spinal cord injury (SCI) lead to early lactate accumulation. Reactive astrocytes polarize and migrate to the lesion site, sealing it off and restricting immune cell infiltration. However, the specific mechanisms through which lactate influences astrocyte polarization remain poorly understood. In this study, we found that lactate can promote astrocyte proliferation, migration and differentiation into A2 after oxygen-glucose deprivation/reoxygenation (OGD/R). Inhibiting signal transducer and activator of transcription 3 (STAT3) nuclear transpor

Exercise protects against pathological cardiac hypertrophy and heart failure, yet the mechanisms remain unclear. We applied the established transverse aortic constriction (TAC) mouse model to induce pressure overload and heart failure, and found that exercise prior to the operation significantly attenuated TAC-induced cardiac hypertrophy, fibrosis, and systolic dysfunction. Notably, these reduction are strongly associated with activation of SIRT1-NRF2 pathway and improved oxidative stress which is robustly elevated in the control sedentary TAC mice. To confirm that cardiac protective effect of

AIM: Intermittent fasting or exercise could be used as an adjunct to regulating abnormal glucose metabolism. However, the mechanism of action of intermittent fasting and exercise to regulate normal glucose metabolism is still unclear. We want to investigate the regulatory effect and mechanism of action of intermittent fasting combined with exercise on glucose metabolism in normal mice.
METHODS: All mice were randomized into six groups of 12 animals each. The effects of 6-week alternate-day fasting (ADF) or time-restricted fasting (TRF) combined with running exercise on blood glucose regulatio

INTRODUCTION: Skeletal muscle function is profoundly challenged under high-altitude environments, where hypobaric hypoxia disrupts structural integrity and impairs physiological function. However, few animal studies have examined the impact of hypobaric hypoxia on skeletal muscle and molecular basis. While exercise training holds promise for alleviating hypoxia-induced muscle dysfunction, the understanding of its protective mechanisms remains limited.
OBJECTIVES: We aimed to investigate chronic hypobaric hypoxia-induced myotube atrophy and mitochondrial dysfunction in mouse models and C2C12 c
