Ocular abnormalities in transgenic mice harboring mutations in the type II collagen gene.

PURPOSE: To characterize the morphological changes in the eyes of transgenic mice harboring different mutations in type II collagen gene to elucidate the function of this collagen in the eye, and to find out whether these animals could function as models for the human arthro-ophthalmopathies of the Kniest, Stickler and Wagner types. METHODS: Three genetically engineered mouse lines representing two types of mutations in the triple-helical domain of type II collagen and their nontransgenic littermates used as controls were analyzed on day 18.5 embryonic development. After genotyping by polymer

Chondrogenic differentiation in cultures of embryonic rat mesenchyme.

The morphology and fine structure of day 12 rat embryonic mesenchyme from forelimb bud, mandibular arches, and frontonasal prominence is described as the cells undergo chondrogenesis in high density, micromass culture. The cultures began as a multilayered "pavement" of flattened mesenchymal cells, 3-4 deep, with moderate intercellular space but little identifiable electron-dense extracellular matrix. Pre-cartilage condensations, which consisted of aggregates of cells which had rounded up, displaying little or no intercellular space, formed within the first 24 h in limb mesenchyme and after an

A model three-dimensional culture system for mammalian dopaminergic precursor cells: application for functional intracerebral transplantation.

Grafts of fetal neural tissue, rich in dopamine (DA) neurons, have previously been shown to improve the symptoms of parkinsonism, both in humans and in animal models. In order to circumvent some of the problems associated with cell transplant therapy, such as the limited availability of transplant tissue, we have established a reaggregate (three-dimensional) tissue culture system that can be used to proliferate normal mammalian neuronal precursors. We demonstrate the in vitro growth of DA-neuronal precursors derived from embryonic porcine ventral mesencephalon, Carnegie stages 15-18. Cultures

Characterization of a murine homeo box gene, Hox-2.6, related to the Drosophila Deformed gene.

The Hox-2 locus on chromosome 11 represents one of the major clusters of homeo-box-containing genes in the mouse. We have identified two new members (Hox-2.6 and Hox-2.7), which form part of this cluster of seven linked genes, and it appears that the Hox-2 locus is related by duplication and divergence to at least one other mouse homeo box cluster, Hox-1. The Hox-2.6 gene encodes a predicted protein of 250 amino acids, which displays extensive similarity in multiple regions to certain mouse, human, Xenopus, and zebra fish homeo domain proteins. The Drosophila Deformed (Dfd) gene also shares th

Appearance and distribution of neurofilament immunoreactivity in iris nerves.

We have used antiserum raised against neurofilament (NF) protein and indirect immunofluorescence techniques to visualize neuronal structures in rodent, cat, and cow irides. In the adult rat iris a large population of nerve fibers with a nonautonomic distribution show NF-like immunoreactivity. In whole mounts, smooth fluorescent fibers were seen in a fine-meshed plexus from the sphincter margin to the ciliary processes. Superimposed, a sparse pattern of thick meandering axon bundles were seen. Electroblotting and peroxidase immunochemical staining techniques unequivocally showed the presence of

External malformations in chick embryos following concomitant administration of methylxanthines and beta-adrenomimetic agents: 1. Gross pathologic features.

The objectives of this report are to document external malformations observed in chick embryos following concomitant administration of methylxanthines (caffeine, theophylline) and beta-adrenomimetic agents (isoproterenol, epinephrine) and to suggest reasonable explanations for the anomalies. Administration of caffeine or theophylline alone (2.5-5.0 mg/egg) retarded growth in a dose-dependent fashion. Doses of 5.0 mg caffeine and theophylline produced beak malformations in 4.9% and 57.1% of embryos, respectively. Limb malformations, seen in low frequency (3.6% in 56 embryos) after administratio

Myocardial wall stiffening in a mouse model of persistent truncus arteriosus.

BACKGROUND: Genetic and epigenetic programs regulate dramatic structural changes during cardiac morphogenesis. Concurrent biomechanical forces within the heart created by blood flow and pressure in turn drive downstream cellular, molecular and genetic responses. Thus, a genetic-morphogenetic-biomechanical feedback loop is continually operating to regulate heart development. During the evolution of a congenital heart defect, concomitant abnormalities in blood flow, hemodynamics, and patterns of mechanical loading would be predicted to change the output of this feedback loop, impacting not only

Structural and transcriptomic response to antenatal corticosteroids in an Erk3-null mouse model of respiratory distress.

BACKGROUND: Neonatal respiratory distress syndrome in preterm infants is a leading cause of neonatal death. Pulmonary insufficiency-related infant mortality rates have improved with antenatal glucocorticoid treatment and neonatal surfactant replacement. However, the mechanism of glucocorticoid-promoted fetal lung maturation is not understood fully, despite decades of clinical use. We previously have shown that genetic deletion of Erk3 in mice results in growth restriction, cyanosis, and early neonatal lethality because of pulmonary immaturity and respiratory distress. Recently, we demonstrated

ER shaping proteins guide spindle elongation and division during rapid cleavage mitoses.

The ER is a complex network of membranes that inhabits much of the cytoplasm of cells - however, this network undergoes a massive condensation and rapid remodeling during cell division. In Drosophila cleavage divisions, this results in a tight association of the ER with centrosomes and mitotic spindle poles. Previous work has shown that this relationship between the ER and centrosomes must be finely tuned to enable successful spindle elongation, and that overaccumulation of the ER in these stages can result in failed centrosome maturation. During interphase, the ER exists in tubular and sheet-

Embryonic and larval development of the endangered Chiapas killifish Tlaloc hildebrandi (Cyprinodontiformes: Profundulidae).

The early ontogeny of endemic freshwater fishes provides essential baseline biological information for conservation and management, particularly in species with restricted distributions and high vulnerability to habitat degradation, environmental change and population decline. This study presents the first detailed description of embryonic and larval development of Tlaloc hildebrandi, an endangered (EN) killifish endemic to Chiapas, Mexico. Ontogenetic development was documented under controlled conditions from fertilisation to the completion of the larval period and the establishment of juven

Antarctic Fish Antimicrobial Peptides Active Against Bacterial and Viral Pathogens of Aquacultural Importance.

Background/Objectives: The aquaculture industry represents a fundamental food sector. One of the main limiting factors for this sector is related to bacterial diseases, for which antibiotics have been widely used worldwide for decades. In recent years, a more conscious approach to the use of antimicrobials within the framework of the One Health approach has increased the need for alternatives capable of helping with disease management while avoiding the onset of antimicrobial resistance phenomena. Antimicrobial peptides, which have a broad spectrum of action against pathogens, are a promising

Understanding how the structures of surfactants in hybrid nanoparticles affect the compaction of ct-DNA for cellular uptake: presenting a highly efficient surfactant.

Compaction of calf thymus DNA (ct-DNA) by two single-head-double-tailed surfactants with variable tail lengths i.e., Dihexadecyldimethylammonium bromide (DDAB16) and Dioctadecyldimethylammonium bromide (DDAB18), and one triple-head-double-tailed surfactant N1-dodecyl-N2-(2-(dodecyldimethylammonio)ethyl)-N1,N1,N2,N2-tetramethylethane-1,2-diaminium (MQAS12) has been studied. DDAB18 is found to be the most efficient, while MQAS12 is the least efficient for cellular uptake. Hybrid materials of surfactants and silica nanoparticles have better compaction efficiency due to the cooperative binding. Si

A Rare Case of Splenunculus Buried within the Stomach.

BACKGROUND: Accessory spleen, or splenunculus, is a congenital anomaly resulting from failure of splenic anlage fusion during embryonic development. It is most commonly located near the splenic hilum or pancreatic tail and rarely occurs within the stomach wall. Gastric accessory spleen is a rare entity that may mimic submucosal neoplasms such as gastrointestinal stromal tumour, posing a diagnostic challenge. CASE PRESENTATION: We report the case of a 37-year-old male presenting with a 3-month history of epigastric pain, dyspepsia, and nausea. Endoscopy revealed a 2.5-3 cm submucosal mass in t

Gene regulatory dynamics during craniofacial development in a carnivorous marsupial.

Marsupials and placental mammals exhibit significant differences in reproductive and life history strategies. Marsupials are born highly underdeveloped after an extremely short period of gestation, leading to prioritized development of structures critical for post-birth survival in the pouch. Critically, they must undergo accelerated development of the orofacial region compared to placentals. Previously, we described the accelerated development of the orofacial region in the carnivorous Australian marsupial, the fat-tailed dunnart Sminthopsis crassicaudata, that has one of the shortest gestati

Maternal administration of APAP induces angiogenesis disorders in mouse placenta via SOCS3/JAK1/STAT3 pathway.

Acetaminophen (APAP, also known as paracetamol) is a commonly used antipyretic and analgesic that is considered safe to use during pregnancy. However, a growing body of research indicates that gestational administration of APAP increased the risk of neurodevelopmental, reproductive and genitourinary disorders in offspring, alongside impairments in placental development. Notably, over-dosed APAP exhibits direct toxicity to endothelial cells, but there is very limited research investigating the impact of APAP on placental angiogenesis, a gap we aim to address in this study. Pregnant mice were ga

Histological and immunohistochemical analysis of gonadal development in the veiled chameleon (Chamaeleo calyptratus).

Chameleons are a family of lizards distinguished by several unique features related to their arboreal lifestyles, such as a ballistic tongue, skin color changes, independent movement of both eyes, a prehensile tail, and cleft hands and feet. The veiled chameleon (Chamaeleo calyptratus) has been proposed as a promising model species for studying squamate biology. Despite its potential, the developmental biology of this species remains poorly understood, particularly in terms of gonadal development. This study aimed to elucidate the development of the gonads in the veiled chameleon, from the ini

Rictor, an mTORC2 Protein, Regulates Murine Lymphatic Valve Formation Through the AKT-FOXO1 Signaling.

BACKGROUND: Lymphatic valves are specialized structures in collecting lymphatic vessels and are crucial for preventing retrograde lymph flow. Mutations in valve-forming genes have been clinically implicated in the pathology of congenital lymphedema. Lymphatic valves form when oscillatory shear stress from lymph flow signals through the PI3K/AKT pathway to promote the transcription of valve-forming genes that trigger the growth and maintenance of lymphatic valves. Conventionally, in many cell types, AKT is phosphorylated at Ser473 by the mTORC2 (mammalian target of rapamycin complex 2). However

Ruthenium p-Cymene Complexes Incorporating Substituted Pyridine-Quinoline-Based Ligands: Synthesis, Characterization, and Cytotoxic Properties.

Organometallic complexes of the formula [Ru(N^N)(p-cymene)Cl][X] (N^N = bidentate polypyridyl ligands, p-cymene = 1-methyl-4-(1-methylethyl)-benzene, X = counter anion), are currently studied as possible candidates for the potential treatment of cancer. Aggregated chromosomes/chromatin transfer: a novel approach for mitochondrial replacement with minimal mitochondrial carryover: the implications of mouse experiments for human aggregated chromosome transfer.

Nuclear transfer techniques, including spindle chromosome complex (SC) transfer and pronuclear transfer, have been employed to mitigate mitochondrial diseases. Nevertheless, the challenge of mitochondrial DNA (mtDNA) carryover remains unresolved. Previously, we introduced a method for aggregated chromosome (AC) transfer in human subjects, offering a potential solution. However, the subsequent rates of embryonic development have remained unexplored owing to legal limitations in Japan, and animal studies have been hindered by a lack of AC formation in other species. Building upon our success in
