Senecavirus A (SVA) infection causes vesicular disease in swine, which presents with clinical signs that are highly similar to those of foot-and-mouth disease, and thus leads to significant challenges for disease control. Although inactivated SVA vaccines effectively elicit humoral immunity, they induce sub-optimal cellular immune responses. To overcome this limitation, we systematically screened T-cell epitopes within the immunogenic non-structural proteins 2C and 3A of SVA to evaluate their potential as novel immune enhancers. Forty-four overlapping peptides spanning the full-length 2C/3A pr

Porcine circovirus type 3 (PCV3) capsid protein (Cap) is a key antigen for immunological studies and vaccine development. Different optimized PCV3 ORF2 sequences were used to construct six baculovirus transfer plasmids, with the pOET1.1-based design yielding the highest Cap level. Cap expression was confirmed by Western blot, IPMA and IFA. Recombinant baculovirus amplification was optimized, achieving the highest titer at an MOI of 0.1 with a 72 h harvest to 107.5TCID50/0.1 mL, while maximal Cap production was obtained at an MOI of 0.1 with a 96 h harvest. PCV3 Cap virus-like particles (VLPs)

The presentation of viral antigens on nanoparticles in multivalent arrays has emerged as a valuable technology for vaccines. On the nanoparticle surface, highly ordered, repetitive arrays of antigens can mimic their geometric arrangement on virion surfaces and elicit stronger humoral responses than soluble viral antigens. More recently, bacterial antigens have been presented on self-assembling protein nanoparticles and have elicited protective antibody and effective T-helper responses, further supporting the nanoparticle platform as a universal approach for stimulating potent immunogenicity. Streptococcus pneumoniae is endowed with a variety of surface-exposed proteins representing putative vaccine candidates. Lipoproteins are covalently anchored to the cell membrane and highly conserved among pneumococcal serotypes. Here, we evaluated these lipoproteins for their immunogenicity and protective potential against pneumococcal colonisation. A multiplex-based immunoproteomics approach revealed the immunogenicity of selected lipoproteins. High antibody titres were measured in sera from mice immunised with the lipoproteins MetQ, PnrA, PsaA, and DacB. An analysis of convalescent patient

Development of a subunit vaccine for shigellosis requires identification of protective antigens and delivering these antigens in a manner that stimulates immunity comparable to that induced by natural infection. The Shigella invasin complex (Invaplex) vaccine is an ion-exchange-purified extract from virulent Shigella that consists of LPS and several other proteins, including the invasins IpaB and IpaC. Intranasal delivery of Invaplex stimulates protective immunity in small animal models for shigellosis. To identify the active component(s) of Invaplex responsible for its immunogenicity and effi

The adjuvant effect of recombinant Rhesus macaque interleukin-12 (RhIL-12) on the induction of cellular and humoral immune responses elicited by the HIV-1 subunit vaccine protein gp120 in Rhesus macaques was examined. RhIL-12 in conjunction with gp120 was given at day 0, 28 and 84 intramuscularly. Coadministration resulted in an approximate 10-fold increase in plasma anti-gp120 antibody levels as compared to levels generated in control monkeys receiving gp120 alone. Potentiation of the humoral arm of the immune response was evident by both ELISA and an antiviral bioassay. In addition, RhIL-12

BACKGROUND: African swine fever virus (ASFV), characterized by its large genome and high antigenic diversity, has impeded the development of traditional vaccines. In this study, we employed the AP205 capsid protein as the scaffold to prepare ASFV multi-epitope nanoparticles via sequential steps: fusion of the scaffold gene with ASFV multi-epitope genes, followed by recombinant protein expression, purification, in vitro self-assembly, structural characterization, and immune evaluation in BALB/c mice.
RESULTS: It was found that the dimeric AP205 scaffold enhanced the assembly efficiency of nano

Leishmaniasis, a vector-borne tropical disease, is caused by the bite of Leishmania-infected sand flies. Currently, chemotherapy is the only available option because of the absence of any approved vaccine. Vaccines are considered decisive for the complete eradication of the disease. In this study, we have designed a multiepitope-based vaccine and evaluated its potential immunogenicity using immunoinformatic tools. We selected Leishmania donovani trypanothione synthetase (LdTryS) for the development and analysis of potential immunogenic epitopes. A total of 4 CTL, 3 HTL and 2 LBL epitopes were

BACKGROUND: Dengue virus (DENV), comprising four antigenically distinct serotypes (DENV1-4), continues to pose a major global health burden. The lack of a universally protective vaccine capable of inducing balanced immunity against all serotypes remains a critical challenge, largely due to antigenic diversity and the risk of antibody-dependent enhancement (ADE). Conserved capsid proteins represent promising targets for epitope-based vaccine development because of their structural stability and immunogenic potential.
METHODS: An integrated reverse vaccinology and immunoinformatics strategy was

BACKGROUND: Tuberculosis is one of the most common and deadliest infectious diseases worldwide affecting almost a third of the world's population. Although this disease is being prevented and controlled by the Bacille Calmette Guérin (BCG) vaccine, the protective efficacy is highly variable and substandard (0-80%) in adults. Therefore, novel and effective tuberculosis vaccine that can overcome the limitations from BCG vaccine need to be developed.
RESULTS: A novel approach of utilizing an in-trans protein surface display system of Lactobacillus plantarum carrying and displaying combination of

Sepsis continues to be a leading cause of death among hospitalized patients. Despite advances in supportive care and the availability of potent antimicrobials, the mortality exceeds 20%. The passive infusion of antibodies directed against a conserved region of the lipopolysaccharide (LPS) of Gram-negative bacteria was highly protective in an early study (NEJM 307 [1982] 1225). When this and similar preparations were unable to show consistent efficacy, efforts were directed towards other strategies, including cytokine modulation. Our group found that a whole bacterial vaccine made from the Esch

Mononegavirales, known as nonsegmented negative-sense (NNS) RNA viruses, are a class of pathogenic and sometimes deadly viruses that include rabies virus (RABV), human respiratory syncytial virus (HRSV), and Ebola virus (EBOV). Unfortunately, no effective vaccines and antiviral therapeutics against many Mononegavirales are currently available. Viral polymerases have been attractive and major antiviral therapeutic targets. Therefore, Mononegavirales polymerases have been extensively investigated for their structures and functions. Mononegavirales mimic RNA synthesis of their eukaryotic counterp

BACKGROUND: The COVID-19 pandemic caused over seven million deaths, and disease control relied largely on mass immunization campaigns. Although diverse vaccine platforms were used, most formulations targeted the Spike protein because of its central role in virus entry. Persistent viral evolution and global vaccine inequity highlight the need for alternative strategies, particularly for low- and middle-income countries. In this scenario, recombinant protein-based vaccines are attractive due to low cost, established manufacturing pipelines and well-characterized safety profiles. The receptor bin

Campylobacter jejuni infection in poultry presents a significant public health concern due to the consumption of contaminated chicken meat and related products. Vaccination has been proposed as a potential mitigation approach to control infections in chickens and reduce the risk of zoonotic transmission. Yet, no commercial vaccines are currently available for controlling this microorganism in chickens. This study aimed to investigate the efficacy of mucosal delivery (in ovo and oral) of multi-antigen chitosan nanoparticle (CNP)-encapsulated vaccines, including C. jejuni lysate and outer membra

Tuberculosis (TB) is the number one infectious disease killer worldwide. Bacillus Calmette-Guérin (BCG) is the only vaccine currently used in clinical practice to prevent TB, but it still has limitations in preventing latent infection and reactivation of TB. The investigation focused on the selection of the Rv0081 antigen linked to latency, leading to the successful creation of the prokaryotic expression recombinant plasmid pET30b-Rv0081 for subsequent expression and purification processes. Antigen specificity was additionally validated through the implementation of whole blood IFN-γ release a

Vaccine development for Group A streptococcal (GAS) infection has been extensively focused on the N-terminal hypervariable or the C-terminal conserved regions of the M protein, a major virulence factor of GAS. We evaluated the immunogenicity and functional activity of the conserved C-terminal peptide vaccine candidate, J8, conjugated to CRM197, in two mouse strains: C3H (H2(k)) and Balb/c (H2(d)), and in rhesus macaques. Mice were immunized with J8-CRM197 formulated with Amorphous Aluminum Hydroxyphosphate Sulfate Adjuvant (AAHSA), and non-human primates were immunized with J8-CRM197 formulate

Dendritic cells are the largest population of antigen presenting cells in the body. One of their main functions is to regulate the delicate balance between immunity and tolerance responsible for maintenance of immunological homeostasis. Disruption of this delicate balance often results in chronic inflammation responsible for initiation of organ specific autoimmune diseases such as rheumatoid arthritis, multiple sclerosis and type I diabetes. The cholera toxin B subunit (CTB) is a weak mucosal adjuvant known for its ability to stimulate immunity to antigenic proteins. However, conjugation of CT

Combination vaccines promise to simplify immunization schedules and improve coverage, but remain technically challenging owing to antigen compatibility, immunogenic balance and formulation complexity. Here we report a modular strategy that uses a single-component nanobody binder to noncovalently attach diverse antigens to intact particles from the licensed hepatitis E vaccine. To identify a suitable binder, an alpaca was immunized with the vaccine, and nanobodies were screened via phage display. One nanobody, P1-5B, selectively bound recessed, non-immunodominant sites on the particle surface a

BACKGROUND: Peste des petits ruminants virus (PPRV) is currently the only member of the Morbillivirus caprinae species within the genus Morbillivirus of the family Paramyoxviridae. PPRV causes a highly contagious disease in small ruminants, especially goats and sheep. Succinylation is a newly identified and conserved modification and plays an important role in host cell response to pathogen infection. However, the extent and function of succinylation in Vero cells during PPRV infection remains unknown.
RESULTS: In this study, a global profile of the succinylome in Vero cells infected with PPR
