Marfan syndrome and related connective tissue disorders in the current era in Switzerland in 103 patients: medical and surgical management and impact of genetic testing.

INTRODUCTION: Marfan syndrome (MFS) and related connective tissue disorders (CTDs) are increasingly recognised. Genetic testing has greatly improved the diagnostic outcome/power over the last two decades. In this study we describe a multicentre cohort of adults with MFS and related CTDs, with a particular focus on results from genetic testing. METHODS: All patients with MFS and related CTDs were identified from the databases of five centres in the canton of Zurich. Echocardiographic and clinical findings including systemic Marfan score, use of medication and genetic results were retrospective

Reclassification of the type B CAT family and identification of the novel chloramphenicol acetyltransferase CatB14 in Riemerella anatipestifer.

Chloramphenicol resistance in Riemerella anatipestifer is primarily mediated by chloramphenicol acetyltransferases (CATs). Inconsistent nomenclature of the CatB superfamily in public databases currently complicates the genomic surveillance of these resistance determinants. In this study, we tested the subtype scheme recorded in the NCBI Reference Gene Catalog against a complete pairwise amino acid identity matrix, applying a ≥ 80% identity criterion. The analysis supports the existing type B-1 to type B-5 subtypes, assigns two previously unassigned sequences to Type B-2, and recommends retirem

Whole exome sequencing and genotype-phenotype correlation in homocystinuria in an Iranian population: a multicenter study.

Homocystinuria is an uncommon metabolic disorder characterized by increased homocysteine concentrations. The condition may arise from mutations in the cystathionine beta-synthase (CBS) gene (classic) or in other genes associated with the cobalamin and folate metabolic pathways (non-classic). This study investigated the genetic diversity and clinical outcomes of homocystinuria in an Iranian pediatric population, exploring genotype-phenotype correlations. A multicenter cross-sectional study carried out from January 2024 to August 2025 at three principal referral centers in Iran. A total of 48 pe

Case Report: ETV6 c.744del gene mutation acute lymphoblastic leukemia in identical twins.

BACKGROUND: It is extremely rare for monozygotic twins to both develop leukemia. This paper reports a clinical case in which a pair of monozygotic twins with germline ETV6 gene mutations successively developed acute lymphoblastic leukemia (B-cell type, ALL-B). The ETV6:c.744del (p.S248fs) is a novel germline mutation associated with familial susceptibility to hematologic malignancies. This mutation site has not been detected in relevant databases or literature, and this is the first international report of it. It provides reference value for genetic screening of such diseases. CASE INTRODUCTI

Genetic Etiologies and Risk Factors for Regressive Autism and Childhood Disintegrative Disorder: A Scoping Review.

Autism spectrum disorder (autism) is diagnosed by persistent deficits in communication and social interaction, along with restricted, repetitive behaviors or interests. About a third of children with autism appear to develop normally but subsequently regress and eventually present with autism. This condition is termed regressive autism and the associated regression termed autistic regression. Children undergoing autistic regression after 2 years are described as having childhood disintegrative disorder. We aimed to conduct a scoping review to identify and summarize the genetic etiologies and c

Causal relationship between uterine fibroids and cardiovascular disease: A two-sample Mendelian randomization study.

Previous studies have indicated that patients with uterine fibroids (UF) may have an elevated risk of cardiovascular disease (CVD), although the causal relationship between UF and CVD remains unclear. In this Mendelian randomization (MR) study, we aimed to investigate the causal association between genetic susceptibility to UF and the risk of developing CVD. We extracted summary statistics for single nucleotide polymorphisms associated with UF and 5 CVDs from multiple databases for further analysis. First, we used linkage disequilibrium score regression to assess the genetic correlation across

Recurrent nonsense p.Trp3416* variant in the DMD gene identified in healthy Lebanese individuals: Implications for variant classification and genotype-phenotype correlations.

BACKGROUND: Duchenne muscular dystrophy (DMD) is a severe X-linked neuromuscular disorder caused by pathogenic variants in the DMD gene, leading to dystrophin deficiency and progressive muscle degeneration. Thousands of variants with diverse types have been reported in DMD, contributing to a broad clinical spectrum. While typically associated with severe phenotypes, pathogenic DMD variants may also cause Becker muscular dystrophy (BMD), a milder form with later-onset muscle weakness, or isolated dilated cardiomyopathy with minimal skeletal muscle involvement. Rarely, asymptomatic individuals c

Whole Genome Sequencing and Genetic Diversity of Respiratory Viruses Detected in Children With Acute Respiratory Infections: A One-Year Cross-Sectional Study in Senegal.

Acute respiratory infections (ARI) are a health priority, especially in countries with limited resources. They are a major cause of morbidity and mortality, especially among children and the elderly. In Senegal, the endemic circulation of respiratory viruses other than influenza has been demonstrated. However, there is a paucity of data exploring the genetic diversity of these viruses based on whole-genome sequencing. In this study, we present data on the genetic diversity of respiratory viruses in children under 15 years old in Senegal, including an overview of the different pathogens detecte
