Melanin-Concentrating Hormone (MCH): Role in Mediating Reward-Motivated and Emotional Behavior and the Behavioral Disturbances Produced by Repeated Exposure to Reward Substances.

Clinical and animal studies suggest that multiple brain systems are involved in mediating reward-motivated and related emotional behavior including the consumption of commonly used drugs and palatable food, and there is evidence that the repeated ingestion of or exposure to these rewarding substances may in turn stimulate these brain systems to produce an overconsumption of these substances along with co-occurring emotional disturbances. To understand this positive feedback loop, this review focuses on a specific population of hypothalamic peptide neurons expressing melanin-concentrating hormo

Beyond marker expression: An ESC-informed framework for engineering and validating chicken induced pluripotent stem cells.

The chicken induced pluripotent stem cells (iPSCs) are still plagued by limitations such as low reprogramming efficiency, instability, and a lack of demonstrated germline competence, which are in part attributed to the absence of a precisely defined biological pluripotency reference. In this review, we propose an ESC-informed approach that positions a molecularly and functionally defined chicken embryonic stem cell (cESC) reference state as the standard against which chicken iPSC generation is designed and evaluated. Combined transcriptional, epigenetic, and signaling information reveals impor

Genomic characterization of sub-populations in human pluripotent stem cell-derived retinal progenitor cells driving retinal lamination.

The mechanism underlying retinal spheroid layer formation was investigated using Rax::GFP-positive retinal progenitor cells from human embryonic stem cell-derived retinal organoids. Single-cell RNA sequencing suggested the earlier cell-cycle exit in non-layered spheroids, while well-layered spheroids retained longer proliferative property with transiently activated canonical WNT2B-FZD7 signaling followed by temporary expression of non-canonical WNT5A. Despite structural differences in vitro, however, both non-layered and well-layered retinal spheroids on differentiation day 60 developed a laye

Role of stem cells in ocular diseases: Progress and challenges.

Ocular diseases, including corneal disease, glaucoma, age-related macular degeneration (AMD), diabetic retinopathy, and retinopathy of prematurity, can significantly impair vision and reduce quality of life. Because degenerated cells in these diseases are unable to regenerate, treatments for these conditions have limited efficacy. Stem cell therapies are revolutionizing the treatment of degenerative eye conditions, enabling structural and functional restoration through mechanisms such as cell replacement and paracrine signaling. This review examines advances in stem cell therapy for ocular dis

Dorsal midline ectopic mammary tissue as a cutaneous marker of spinal dysraphism: intradural lipoma and tethered cord in a child with literature review.

PURPOSE: Supernumerary breasts (polymastia) and supernumerary nipples (polythelia) are congenital anomalies resulting from incomplete regression of the embryonic mammary ridge. They most frequently occur along the embryologic milk line extending from the axilla to the groin. Ectopic localization outside this line is rare, and dorsal presentations are exceptional. In such cases, an association with underlying spinal dysraphism has occasionally been reported. METHODS: We report the case of an 11-year-old girl presenting with a congenital dorsal midline swelling initially diagnosed as a lipoma.

scBrainScope: cross-species multidimensional brain atlas.

As one of the most evolutionarily complex and functionally diverse organs, the brain is characterized by its intricate structure, developmental alterations, and cellular heterogeneity. Although extensive brain atlases exist, there remains an urgent need for an integration platform that brings together gene expression profiles and analysis tools to explore features across species, brain regions, developmental stages, and pathological conditions. Thus, we developed scBrainScope, a detailed and interactive transcriptomic atlas of the brain. Our platform brings together single cell sequencing data

Spatial protein expression patterns across pathologically-associated fibers revealed molecular specialization in inclusion body myositis.

OBJECTIVES: In Inclusion Body Myositis (IBM), myofibers undergo structural and functional changes, including increased regeneration, atrophy, and fibrosis. The molecular mechanisms driving pathologically -associated fibers (PAF) remain poorly understood. METHODS: We developed a myofiber-level proteomic workflow to identify protein signatures of three PAF subtypes. Laser-capture microdissection mass spectrometry of immunolabeled cryosections was performed, complemented by immunofluorescence and electron microscopy validation. RESULTS: Regenerating fibers expressing embryonic myosin heavy chain

Identification of New Putative Autotaxin Inhibitors via Structure-Based Virtual Screening and Evaluation of Their Antiproliferative Properties Against Ovarian and Breast Cancer Cells.

Autotaxin (ATX), a lysophospholipase D playing an important role in several inflammatory diseases, as well as in tumor invasion, progression, and metastasis, is an attractive therapeutic target. Herein, we report a virtual screening study of an in-house molecular database. In silico simulations highlighted five putative ATX inhibitors. The enzyme assay showed that compound 3 achieves inhibition potency in the submicromolar range (IC50 = 0.534 µM), whereas all the other test compounds proved to be moderate (2) or weak (1, 4, and 5) inhibitors of ATX at 10 µM concentration. These compounds, eval

Nicotinamide riboside supplementation protects against maternal diabetes-associated decline in oocyte quality.

IN BRIEF: NAD+ levels were reduced in streptozotocin (STZ)-induced diabetic mice, but nicotinamide riboside (NR) supplementation improved these levels in diabetic ovaries and oocytes, enhancing oocyte quality and early embryo development by improving mitochondrial function and lowering reactive oxygen species (ROS) levels. ABSTRACT: Diabetes mellitus is strongly correlated with a decline in oocyte quality; however, noninvasive and effective methods to improve this issue have yet to be fully development. Here, we demonstrate that in vivo supplementation with NR 400 mg/kg/day for 14 days effect

Large-scale genetic mapping for human brain asymmetry.

Left-right asymmetry is an important aspect of human brain organization for functions including language and hand motor control, which can be altered in some psychiatric traits. The last 5 years have seen rapid advances in the identification of specific genes linked to variation in asymmetry of the human brain and/or handedness. These advances have been driven by a new generation of large-scale genome-wide association studies, carried out in samples ranging from roughly 16,000 to over 1.5 million participants. The implicated genes tend to be most active in the embryonic and fetal brain, consis

Zebrafish otic vesicle and mouse epididymis as model systems for studying columnar epithelial cell division.

Epithelial cell division maintains tissue architecture through coordinated nuclear migration, cell shape changes, and spindle orientation. In columnar epithelia, interkinetic nuclear migration (INM) involves apical nuclear translocation in G2 and basal return post-mitosis, yet its regulation and physiological significance remain understudied due to limited live imaging in vivo models. We adapted the zebrafish embryonic otic vesicle as an in vivo model to study epithelial division dynamics using high-resolution live imaging and genetic tools. We find that apical INM initiates in mid-to-late G2

Dynamic Expression of α-Internexin in the Chicken Pineal Gland: A Study of Developmental, Structural, and Functional Roles.

The pineal gland is a photoneuroendocrine organ that regulates circadian rhythms, primarily through rhythmic melatonin secretion. In nonmammalian vertebrates such as birds, pinealocytes retain photosensory and endocrine functions. α-Internexin, a neuronal intermediate filament protein, has been implicated in neurodevelopment and cytoskeletal stability. Its expression in the retinal photoreceptors and pinealocytes has been reported in several vertebrate models; however, its expression pattern in the chicken pineal gland remains unclear. In this study, we used immunohistochemistry and western bl

Mapping novel linear B-cell epitopes target Hepatitis C virus envelope proteins E1 and E2 by monoclonal antibodies for increasing the sensitive detection sites.

Since there is no protective vaccine against Hepatitis C, timely diagnosis and treatment at an early stage of viral infection is crucial to prevent and control hepatitis C virus (HCV) from causing chronic infection. E1 and E2 proteins of HCV can produce neutralizing antibodies and are often used to screen potential epitopes. In this study, human embryonic kidney 293 (HEK293F) cells were used to generate highly active HCV E1 and E2 proteins, which were then used as antigen for producing mouse monoclonal antibodies (mAbs). Novel B cell epitopes of E1 and E2 were disclosed using monoclonal antibo

Photoreceptor calyceal processes accompany the developing outer segment, adopting a stable length despite a dynamic core.

Vertebrate photoreceptors detect light through a large cilium-based outer segment, which is filled with photopigment-laden membranous discs. Surrounding the base of the outer segment are microvilli-like calyceal processes (CPs). Although CP disruption has been associated with altered outer segment morphology and photoreceptor degeneration, the role of the CPs remains elusive. Here, we used zebrafish as a model to characterize CPs. We quantified CP parameters and report a strong disparity in outer segment coverage between photoreceptor subtypes. CP length is stable across light and dark conditi

The epigenetic modification of DNA methylation in neurological diseases.

Methylation, a key epigenetic modification, is essential for regulating gene expression and protein function without altering the DNA sequence, contributing to various biological processes, including gene transcription, embryonic development, and cellular functions. Methylation encompasses DNA methylation, RNA methylation and histone modification. Recent research indicates that DNA methylation is vital for establishing and maintaining normal brain functions by modulating the high-order structure of DNA. Alterations in the patterns of DNA methylation can exert significant impacts on both gene e

A genetically small fetus impairs placental adaptations near term.

The placenta is a gatekeeper between the mother and fetus, adapting its structure and functions to support optimal fetal growth. Studies exploring adaptations of placentae that support the development of genetically small fetuses are lacking. Here, using a mouse model of impaired fetal growth, achieved by deleting insulin-like growth factor 2 (Igf2) in the epiblast, we assessed placental nutrient transfer and umbilical artery (UA) blood flow during late gestation. At embryonic day (E) 15.5, we observed a decline in the trans-placental flux of glucose and system A amino acids (by using 3H-MeG a

Involvement of kinesins in skeletal dysplasia: a review.

Skeletal dysplasias are group of rare genetic diseases resulting from mutations in genes encoding structural proteins of the cartilage extracellular matrix (ECM), signaling molecules, transcription factors, epigenetic modifiers, and several intracellular proteins. Cell division, organelle maintenance, and intracellular transport are all orchestrated by the cytoskeleton-associated proteins, and intracellular processes affected through microtubule-associated movement are important for the function of skeletal cells. Among microtubule-associated motor proteins, kinesins in particular have been sh

A rare case of a toddler with unilateral cryptorchidism-related cranial suspensory ligament remnant.

KEY CLINICAL MESSAGE: Several animal experiment studies have shown that insufficient testicular descent to the scrotum can be caused by persistence of cranial suspensory ligament (CSL). We report a case of right cryptorchidism in a male toddler surgically treated with an orchidopexy possibly associated with CSL persistence based on intraoperative and pathological findings. This case would be a precious source to further investigate the etiopathogenesis of cryptorchidism. ABSTRACT: The CSL anchors embryonic gonads to the dorsal abdominal wall during antenatal mammalian development. Although it

High-Performance Thin-Layer Chromatography-Densitometry-Tandem ESI-MS to Evaluate Phospholipid Content in Exosomes of Cancer Cells.

The question of whether exosome lipids can be considered as potential cancer biomarkers faces our current limited knowledge of their composition. This is due to the difficulty in isolating pure exosomes, the variability of the biological sources from which they are extracted, and the uncertainty of the methods for lipid characterization. Here, we present a procedure to isolate exosomes and obtain a deep, repeatable, and rapid phospholipid (PL) composition of their lipid extracts, from embryonic murine fibroblasts (NIH-3T3 cell line) and none (B16-F1) and high (B16-F10) metastatic murine skin m

Cerium oxide decorated 5-fluorouracil loaded chitosan nanoparticles for treatment of hepatocellular carcinoma.

Reactive oxygen species (ROS) play a crucial role in the mammalian system in both normal and pathological conditions. Hence, this work prepared and characterized the ROS responsive cerium oxide nanoparticles (CeO2 NPs) decorated 5-fluorouracil (5FU) loaded chitosan (CS) nanoparticles (CS-5FU NPs) for enhanced anticancer activity in hepatocellular carcinoma (HepG2 cells). CeO2 NPs decorated CS-5FU NPs were found to be spherical in shape and black dense aggregated particles sized 200 nm. The functional properties and cubic crystalline structure of CeO2 NPs were studied by Fourier-transform infra
