Effect of piperonyl butoxide on cell replication and xenobiotic metabolism in the livers of CD-1 mice and F344 rats.

Male CD- 1 mice were fed diets containing 0 (control), 10, 30, 100, and 300 mg/kg/day piperonyl butoxide (PBO) and 0.05% sodium phenobarbital (NaPB) and male F344 rats were fed diets containing 0 (control), 100, 550, 1050, and 1850 mg/kg/day PBO and 0.5% NaPB for periods of 7 and 42 days. In both species PBO and NaPB increased relative liver weight and whereas PBO produced a midzonal (mouse) or periportal/midzonal (rat) hypertrophy, NaPB produced a centrilobular hypertrophy. In the rat, individual cell necrosis was also observed at 42 days after high doses of PBO. Replicative DNA synthesis, as

Morphine causes distinct changes in the lipidome throughout the brain and body after acute or chronic administration: implications for novel endogenous lipid signaling systems involved in opioid reward and withdrawal.

A growing body of evidence demonstrates that signaling pathways of endogenous lipids (endolipids) modulate the reinforcing properties of opioids including reward and withdrawal. Many of these studies are focused on the endocannabinoid (eCB) system, and its primary eCB ligands, Anandamide (AEA) and 2-arachidonoylglycerol (2-AG). The central hypothesis is that modulation of the eCB system through eCB receptors and enzymes may improve therapeutic outcomes for opioid use disorder (OUD). However, outcomes in preclinical and clinical studies using the low efficacy, CB1/CB2 orthosteric agonist, THC,

Heat-Treated Limosilactobacillus fermentum PS150 Improves Sleep Quality with Severity-Dependent Benefits: A Randomized, Placebo-Controlled Trial.

Background: Insomnia is prevalent and difficult to treat safely over the long term. Given the role of the microbiota-gut-brain axis in melatonin and hypothalamic-pituitary-adrenal (HPA) regulation, and preclinical evidence for Limosilactobacillus fermentum PS150, we evaluated whether a heat-treated formulation (HT-PS150) could improve sleep and modulate endocrine/circadian markers in adults with poor sleep. Methods: In a randomized, double-blind, placebo-controlled trial, 84 adults aged 20-60 years with PSQI ≥ 5 and ISI < 22 were assigned to receive either placebo or HT-PS150 for eight weeks.

Rosmarinic acid- and epigallocatechin gallate-enriched extracts improve insomnia and sleep-wake balance in caffeine-and p-chlorophenylalanine-induced insomnia models.

This study evaluated the effects of a rosmarinic acid- and epigallocatechin gallate-enriched extract mixture (RE-GTE) on insomnia and sleep-wake disturbances induced by excessive caffeine intake or serotonin depletion using p-chlorophenylalanine (PCPA) in mice. RE-GTE was administered in both caffeine- and PCPA-induced insomnia models, and sleep-related behaviors were assessed using pentobarbital-induced sleep tests and behavioral analyses of anxiety-related and locomotor parameters. In addition, sleep-related neurotransmitters and receptor expression were examined using molecular and biochemi

NAcLat-DBS reduces GABAB expression of dopamine neurons to correct depressive symptoms in CMS mice.

Hypodopaminergic activity within the mesolimbic circuitry is recognized as a typical characteristic in severe and refractory depression. Deep brain stimulation (DBS) of the nucleus accumbens (NAc) has shown therapeutic potential for treatment-resistant depression (TRD), yet its underlying molecular mechanism remains incompletely defined. We used chronic mild stress (CMS) combining fiber photometry recording, pharmacological interventions and genetic manipulations in freely moving mice to examine the variation of dopamine (DA) activity in the ventral tegmental area (VTA) and the potential roles

Zolpidem restores sleep and decreases amyloid in a mouse model.

INTRODUCTION: Deficits in non-rapid eye movement (NREM) sleep facilitate Alzheimer's disease (AD) progression. Enhancing gamma-aminobutyric acid-ergic (GABAergic) signaling can restore sleep. Unbiased computational analysis identified zolpidem as high-affinity GABA receptor modulator facilitating chloride transport that could slow AD. METHODS: Zolpidem's effects on sleep and Alzheimer's progression were evaluated in young APP/PS1 (amyloid precursor protein/presenilin 1) mice. Sleep was monitored with electroencephalography/electromyography (EEG/EMG) telemetry. Wide-field imaging with voltage-

The α1 subunit-containing GABAA receptor-mediated inhibitory transmission in the lateral orbitofrontal cortex contributes to anxiety- and depression-like behaviors in Parkinsonian rats.

The lateral orbitofrontal cortex (LO) is critically involved in the modulation of anxiety and depression. However, the involvement of GABAA receptor-mediated inhibitory transmission within the LO in Parkinson's disease (PD)-associated anxiety and depression remains unclear. In this study, unilateral 6-hydroxydopamine (6-OHDA) lesions of the substantia nigra pars compacta (SNc) in adult male Sprague-Dawley rats induced anxiety- and depression-like behaviors, accompanied by hyperactivity of glutamatergic neurons, decreased synthesis and release of GABA and increased release of glutamate, as well

Transcriptomes of higher order thalamic nuclei in obsessive compulsive disorder.

Obsessive-compulsive disorder (OCD) is a chronic psychiatric illness associated with altered function in cortico-striatal-thalamo-cortical (CSTC) circuits. In this pilot study, we examined differential RNA expression in the thalamus using postmortem human brain tissue samples from 11 subjects with OCD and 10 unaffected subjects. We individually dissected the mediodorsal magnocellular, mediodorsal parvocellular, and ventral anterior nuclei, which participate in orbitofrontal and anterior cingulate CSTC circuits most frequently associated with OCD, and the posterior ventrolateral nucleus, which

Neuropeptide S alleviates neuropathic pain through lateral hypothalamic orexinergic circuit in rats.

Neuropathic pain poses a significant clinical challenge due to limited treatment options. Neuropeptide S (NPS) system is known to modulate pain and related affective states; however, the underlying mechanisms remain unexplained. NPS activates lateral hypothalamic (LH) orexinergic neurons involved in feeding, drug seeking, and stress-induced analgesia. Herein, we test the hypothesis that pain modulatory action of NPS is mediated by the LH-orexinergic system. Neuropathic pain was induced in rats via chronic constriction injury (CCI) of the sciatic nerve and the response was investigated using pa

Lactobacillus reuteri ATG-F4 alleviates behavioral impairment induced by chronic stress.

Chronic stress impairs quality of life by disturbing physical and psychological functioning, and increases susceptibility to a wide range of diseases. Lactobacillus reuteri ATG-F4 isolated from the fecal samples of a newborn baby alleviated chronic stress-induced anhedonia by altering the expression of neurometabolites and neurotransmitter receptors. In this study, we examined whether ATG-F4 can improve behavioral deterioration induced by chronic stress using the Laboratory Animal Behavioral Observation Registration and Analysis System (LABORAS). The continuous behavior of the mice was examine

Low 5HT1B and 5HT4 Receptor Neurotransmission as a Potential Link Between Cholesterol Metabolism and Suicide Risk.

Elevated levels of the inflammatory cytokine IL-6 and decreased levels of cholesterol in blood and brain tissue have been reported in studies of individuals who attempted or completed suicide. The mechanisms underlying these effects remain unclear. In this review, we discuss a potential mechanistic link between these observations involving lipid raft function and serotonergic signaling. Reduced cholesterol availability may affect lipid raft function and could, potentially through reduced levels of the lipid raft protein S100A10 (p11), result in diminished cell-surface expression of the seroton

Synergistic regulation of itch behaviors by corticotropin-releasing hormone neurons in the medial prefrontal cortex.

The medial prefrontal cortex (mPFC), which abundantly expresses corticotropin-releasing hormone (CRH) and its receptors, is a key region for integrating somatosensory, cognitive, and emotional information. Although CRH has been implicated in itch modulation, the cellular and local-circuit roles of CRH-positive neurons in the mPFC remain poorly defined. Here, we combined behavioral assays, in vivo fiber photometry, chemogenetics, optogenetics, immunofluorescence, and ex vivo whole-cell patch-clamp recordings to determine how mPFC CRH neurons regulate itch-related scratching in mice. mPFC CRH ne

Timing of exercise differentially modulates fear memory and hippocampal neurotransmitters in male rats.

Exercise promotes neurogenesis and enhances memory consolidation while reducing the retention of aversive memories and anxiety-like behaviors. While our previous work found that acute exercise alters neurotransmitter concentrations, including dopamine and serotonin, in a time-of-day-dependent manner, the long-term effects of chronically timed exercise on neurotransmitter dynamics and behavioral phenotypes remain unclear. To examine whether the daily timing of a chronic exercise intervention modulates its impact on neurotransmitter profiles and fear responses, male rats were conditioned using a

Effects of carbamazepine and phenytoin in Krushinsky-Molodkina rats: further validation of the audiogenic kindling model of epilepsy.

In the present study, we evaluated the effects of the clinically approved anti-seizure medications (ASMs) carbamazepine (CBZ) and phenytoin (PHT) on brainstem audiogenic seizures (AGS) and limbic seizures in Krushinsky-Molodkina (KM) audiogenic rats. In order to analyze the effects of ASMs on the standard AGS, KM rats received single intraperitoneal injection of CBZ (50 mg/kg), PHT (25 mg/kg), or vehicle prior to the single AGS stimulation. The further three groups of KM rats were exposed to 14 daily AGS stimulations (audiogenic kindling), resulting in the stable development of limbic clonus.

Alteration of BDNF and noradrenergic markers in locus coeruleus in a mouse model of cancer-induced bone pain.

The locus coeruleus (LC) is the principal source of noradrenaline (NA) in the central nervous system. In neuropathic pain states, nociceptive stimuli activate LC. This study examined the expression and localization of BDNF and NE neuron-specific proteins in the LC of mice with cancer-induced bone pain (CIBP). Behavioral experiments demonstrated that CIBP mice exhibited persistent spontaneous pain, mechanical and thermal hyperalgesia, and deficits in locomotor activity and motor coordination. H&E and TRAP staining revealed trabecular bone destruction and increased in osteoclast activity. Immuno

Embracing Diversity: Sedative effects vary across drug chemistry and frequency domains.

Moderate sedation can balance patients' comfort and preserve brain activities during deep brain stimulation neurosurgical procedures; however, the sub-cortical effects of different sedatives are rarely explored. We simultaneously recorded EEG, local field potential (LFP), and spikes (multi- and single-unit) from the dorsolateral frontal cortex and external globus pallidus (GPe), the central nucleus of the basal ganglia, of two female non-human primates. These recordings were carried out after titration of each drug and animal to achieve moderate sedative effects. The recording sessions include
