Soft tissue-mimicking hydrogel stiffness modulates polarisation of human monocyte-derived macrophages.

Minimising the foreign body response (FBR) to implants remains an unmet clinical challenge. The mechanical properties of biomaterials, particularly stiffness, have been shown to influence macrophage activation and FBR. However, current literature on the effects of material stiffness on macrophage activation presents conflicting results. These inconsistencies may stem from the use of cell lines or murine primary cells, which do not fully represent the behaviour of primary human monocyte-derived macrophages. Additionally, many previous studies have focused on stiffness values that fall outside t

Pulmonary drug delivery applications of natural polysaccharide polymer derived nano/micro-carrier systems: A review.

Respiratory distress syndrome and pneumothorax are the foremost causes of death as a result of the changing lifestyle and increasing air pollution. Numerous approaches have been studied for the pulmonary delivery of drugs, proteins as well as peptides using meso/nanoparticles, nanocrystals, and liposomes. These nano/microcarrier systems (NMCs) loaded with drug provide better systemic as well as local action. Furthermore, natural polysaccharide-based polymers such as chitosan (CS), alginate (AG), hyaluronic acid, dextran, and cellulose are highly used for the preparation of nanoparticles and de

Plasma Bead Entrapped Liposomes as a Potential Drug Delivery System to Combat Fungal Infections.

Fibrin-based systems offer promises in drug and gene delivery as well as tissue engineering. We established earlier a fibrin-based plasma beads (PB) system as an efficient carrier of drugs and antigens. In the present work, attempts were made to further improve its therapeutic efficacy exploiting innovative ideas, including the use of plasma alginate composite matrices, proteolytic inhibitors, cross linkers, and dual entrapment in various liposomal formulations. In vitro efficacy of the different formulations was examined. Pharmacokinetics of the formulations encapsulating Amphotericin B (AmpB

Effect of peroxymonosulfate oxidation activated by powdered activated carbon for mitigating ultrafiltration membrane fouling caused by different natural organic matter fractions.

Powdered activated carbon (PAC) adsorption has been widely applied prior to ultrafiltration membrane for potable water production. However, the impact of PAC adsorption on membrane fouling was still controversial. To solve this problem, combined PAC and peroxymonosulfate (PMS) pretreatment was proposed in this study. The application of PAC/PMS for mitigating membrane fouling by natural organic matter (NOM) has been evaluated, and compared with PMS oxidation or PAC adsorption alone. The influence of NOM fractions on the control efficiency was also investigated using humic acid (HA), bovine seru

Fabrication of food-derived bigels beads for the co-delivery of hydrophobic and hydrophilic bioactives: insights into environmental responsiveness and release mechanisms.

Food-derived bioactives often suffer from poor physicochemical stability during processing and storage. To address this, we developed a novel dual-network bigel bead system for the efficient co-delivery of hydrophilic (l-ascorbic acid) and lipophilic (β-Carotene) compounds. Formulated by integrating sodium alginate (SA) or cellulose nanofiber (CNF) hydrogels with a monoglyceride oleogel, the system's structural characteristics, environmental responsiveness, and digestive behavior were systematically evaluated. Structural analyses (FTIR and DSC) revealed that the oleogel phase fundamentally rec

DLP-bioprinted ultrabiomimetic trachea with spatiotemporal angiogenesis regulation for segmental airway reconstruction.

Segmental airway reconstruction requires tracheal grafts that emulate the hierarchical structure and biochemical functions of native trachea. However, achieving both biomimetic fidelity and efficient vascularization remains challenging. Here, we present a digitally light-processed tissue-engineered trachea featuring anatomically biomimetic C-shaped cartilage rings, alternating fibrous rings, and dorsal fibrous bands, precisely replicating the structural hierarchy of native trachea. To promote rapid neovascularization, we develop a stress-relaxing and degradable alginate-based hydrogel capable

Multifunctional Hydrogel Dressing Based on Glycosylated Collagen-Curcumin Nanoparticles Effectively Accelerates Wound Healing.

Multifunctional hydrogels have aroused great interest in accelerating wound healing. In this study, a hydrogel dressing with antioxidant, antibacterial, anti-inflammatory, and wound healing properties is developed using xylose (Xy) glycated Chinese giant salamander collagen (CGSPXy), curcumin (Cur), sodium alginate (SA), and chitosan (CS). Specifically, the CGSP/CGSPXy-Cur nanoparticle is mixed with SA through hydrogen bonding interactions to form a hydrogel framework. Subsequently, the CGSP/CGSPXy-Cur-SA is immersed in a solution containing Ca2⁺ and CS, forming an ion-crosslinked semi-interpe

Programmable Tissue Folding Patterns in Structured Hydrogels.

Folding of mucosal tissues, such as the tissue within the epithelium of the upper respiratory airways, is critical for organ function. Studying the influence of folded tissue patterns on cellular function is challenging mainly due to the lack of suitable cell culture platforms that can recreate dynamic tissue folding in vitro. Here, a bilayer hydrogel folding system, composed of alginate/polyacrylamide double-network (DN) and hyaluronic acid (HA) hydrogels, to generate static folding patterns based on mechanical instabilities, is described. By encapsulating human fibroblasts into patterned HA

Hollow Microfiber Assembly-Based Endocrine Pancreas-on-a-Chip for Sugar Substitute Evaluation.

With the increasing demand for low-sugar, low-calorie healthy diets, artificial sweeteners are widely used as substitutes for sugar in the food industry. Therefore, developing models that can better predict the effects of sugar substitutes on the human body is necessary. Here, a new type of endocrine pancreas-on-a-chip is developed based on a microfiber assembly and its stimulation of pancreatic secretion by glucose or sugar substitutes is evaluated. This new endocrine pancreas-on-a-chip is assembled using two components: (1) a cell-loaded hollow methacrylate gelatin (GelMA)/calcium alginate (

Cartilage is a connective tissue which a limited capacity for healing and repairing. In this context, osteoarthritis (OA) disease may be developed with high prevalence in which the use of scaffolds may be a promising treatment. In addition, three-dimensional (3D) bioprinting has become an emerging additive manufacturing technology because of its rapid prototyping capacity and the possibility of creating complex structures. This study is focused on the development of nanocellulose-alginate (NC-Alg) based bioinks for 3D bioprinting for cartilage regeneration to which it is added chondroitin sulf

Fabrication of Konjac glucomannan-based composite hydrogel crosslinked by calcium hydroxide for promising lacrimal plugging purpose.

Swelling, mechanical strength, flexibility, and toughness are important parameters in hydrogel preparation for application in the human body. Herein, composite hydrogels were prepared using a mix of Konjac glucomannan (KGM), sodium alginate (SA), and polyvinyl alcohol (PVA) cross-linked by calcium hydroxide. The PVA/KGM/SA composite hydrogel showed a suitable swelling ratio and rate, as well as elasticity and flexibility. In addition, the elongation at break was 660.3%, with a breaking strength of 87.25 kPa and a compression modulus of 1.660 MPa. Rheological studies showed that the composite h
