CTLA-4 gene polymorphism at position 49 in exon 1 reduces the inhibitory function of CTLA-4 and contributes to the pathogenesis of Graves' disease.

Activation of T cells requires at least two signals transduced by the Ag-specific TCR and a costimulatory ligand such as CD28. CTLA-4, expressed on activated T cells, binds to B7 present on APCs and functions as a negative regulator of T cell activation. Our laboratory previously reported the association of Graves' disease (GD) with a specific CTLA-4 gene polymorphism. In theory, reduced expression or function of CTLA-4 might augment autoimmunity. In the present study, we categorized autoimmune thyroid disease patients and normal controls (NC) by genotyping a CTLA-4 exon 1 polymorphism and inv

Novel mechanism for presynaptic inhibition: GABA(A) receptors affect the release machinery.

Presynaptic inhibition is produced by increasing Cl(-) conductance, resulting in an action potential of a smaller amplitude at the excitatory axon terminals. This, in turn, reduces Ca(2+) entry to produce a smaller release. For this mechanism to operate, the "inhibitory" effect of shunting should last during the arrival of the "excitatory" action potential to its terminals, and to achieve that, the inhibitory action potential should precede the excitatory action potential. Using the crayfish neuromuscular preparation which is innervated by one excitatory axon and one inhibitory axon, we found,

Involvement of Ca2+ influx in the mechanism of tamoxifen-induced apoptosis in HepG2 human hepatoblastoma cells.

The signaling mechanism of tamoxifen (TAM)-induced apoptosis was investigated in HepG2 human hepatoblastoma cells which do not express the estrogen receptor (ER). TAM induced cytotoxicity and DNA fragmentation, a hallmark of apoptosis, in a dose-dependent manner. TAM increased the intracellular concentration of Ca2+. This effect was completely inhibited by the extracellular Ca2+ chelation with EGTA. TAM also induced a Mn2+ influx, indicating that TAM activated Ca2+ influx pathways. This action of TAM was significantly inhibited by flufenamic acid (FA), a known non-selective cation channel bloc

FXY2/MID2, a gene related to the X-linked Opitz syndrome gene FXY/MID1, maps to Xq22 and encodes a FNIII domain-containing protein that associates with microtubules.

Opitz G/BBB syndrome (OS) is a genetically heterogeneous disorder with an X-linked locus and an autosomal locus linked to 22q11.2. OS affects multiple organ systems with often variable severity even between siblings. The clinical features, which include hypertelorism, cleft lip and palate, defects of cardiac septation, hypospadias, and anorectal anomalies, indicate an underlying disturbance of the developing ventral midline of the embryo. The gene responsible for X-linked OS, FXY/MID1, is located on the short arm of the human X chromosome within Xp22.3 and encodes a protein with both an RBCC (

Predominant HLA-class II bound self-peptides of a hematopoietic progenitor cell line are derived from intracellular proteins.

Human myeloid progenitor cells temporarily express HLA class II molecules during the differentiation pathway to granulocytes and macrophages. The significance of major histocompatibility complex (MHC) class II molecules at this stage of development is unknown. As a first stop of inquiry into their function, we have characterized the profile of major self-peptides bound to the HLA-DR molecules expressed by KG-1 cells, a line that shares many of the phenotypic characteristics of colony-forming unit-granulocyte-macrophage progenitors. Searches of protein data bases showed that all matching peptid

On the regulation of the renal response blood volume expansion by vascular parameters in the rat.

The dynamic patterns of body fluid volume distribution, of cardiovascular variables and of renal water, sodium and potassium excretion were studied in the anaesthetized rat following acute, iso-oncotic blood volume expansion. The increased renal excretion following expansion was soon reversed and its pattern to termination was not correlated with changes in pulse and mean arterial pressure, in plasma electrolyte concentration or in haematocrit. At the end of the renal response the change in extracellular fluid volume was not usually corrected, the blood volume was always well above its control

Effects of Ampelopsis brevipedunculata on Ovalbumin-Induced Allergic Airway Inflammation via Regulating Mast Cell Activation and PLCγ/Akt/NF-κB Signaling Pathway.

Asthma is a chronic allergic airway inflammation that requires ongoing therapeutic intervention and reduces the patient's quality of life. Therefore, safe and efficacious drugs for the treatment of asthma are urgently needed. This study investigated whether the ethyl acetate fraction derived from a 95% (v/v) ethanol extract of dried Ampelopsis brevipedunculata (ABE) alleviates allergic airway inflammation. To investigate the anti-allergic and anti-inflammatory effects of ABE, a mouse model of ovalbumin (OVA)-induced allergic airway inflammation was used and clarified their mechanisms in mast c

Changes in segmental bioimpedance following eccentric plantar flexor exercise taken to volitional failure.

Segmental bioelectrical impedance analysis (BIA) evaluates cellular health through phase angle (PhA), resistance (R), and reactance (Xc), providing insights into localized muscle and fluid balance changes during exercise recovery. This exploratory study examined localized BIA responses following an eccentric plantar flexor exercise protocol combined with functional testing. Twenty male participants (82.6 ± 16 kg; 177 ± 8.7 cm; 23.8 ± 3.45 years) completed unilateral eccentric plantar flexor resistance training (40% of body mass as a weighted vest, four sets of fifty repetitions, fifth set to f

Mitochondrial dysfunction and glycolytic shift define the metabolic profile of hydatid fluid-stimulated dendritic cells.

Dendritic cells (DCs) require substantial metabolic reprogramming to mount an immune response against parasites. In the context of Echinococcus granulosus infection, parasite antigens can modulate host immune responses; however, their specific impact on DC metabolism remains poorly defined. We measured mitochondrial membrane potential (ΔΨM), intracellular reactive oxygen species (ROS), and nitric oxide (NO) production in FMS-like tyrosine kinase 3 ligand (FLT3-L)-derived DC (FL-DCs) stimulated with hydatid fluid (HF) or purified laminar layer (pLL) from E. granulosus. Diagnostic limitations of oropharyngeal swab cultures and inconsistent presence of secondary bacterial infection in canine aspiration pneumonia.

OBJECTIVE: In dogs with aspiration pneumonia (AP), our objectives were to determine whether oropharyngeal (OP) swab isolates would match bronchoalveolar lavage fluid (BALF) isolates and determine the frequency of secondary bacterial infection (SBI) in AP by use of 4 definitions. METHODS: 30 client-owned dogs with clinical diagnosis of AP (cough or fever, and radiographic alveolar pattern with historical vomiting or regurgitation) with OP and BALF cultures were prospectively enrolled from 2019 to 2023. Exclusion criteria included antimicrobial administration within 14 days of presentation or i

PLGA-particle-based vaccine induces SARS-CoV-2-specific antibody and T-cell responses.

BACKGROUND AND PURPOSE: Current coronavirus disease 2019 (COVID-19) vaccines effectively prevent severe disease but induce primarily systemic immunity without mucosal protection in the respiratory tract, which is mandatory for protection from severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection and the prevention of viral transmission. Vaccination strategies capable of inducing local immunity at the site of infection are therefore needed. Here, we evaluated a poly (lactic-co-glycolic acid) (PLGA) microparticle-based vaccine co-encapsulating the receptor binding domain (RBD) o

In vivo-measured Lewy body pathology is associated with neuropsychiatric symptoms across the Alzheimer's disease continuum.

Intracellular alpha-synuclein aggregates, known as Lewy bodies (LB), are commonly observed in Alzheimer's disease (AD) dementia. Post-mortem studies have shown a higher frequency of neuropsychiatric symptoms among individuals with AD and LB co-pathology. However, the effects of in vivo-measured LB pathology on neuropsychiatric symptoms in AD remain underexplored. This study aimed to evaluate cross-sectional and longitudinal effects of in vivo-measured LB pathology on neuropsychiatric symptoms across the AD continuum. We analyzed data from 1,169 participants from the Alzheimer's Disease Neuroim

Elevated Flotillin-1 in Saliva and Salivary Glands: A Novel Non-Invasive Biomarker in an Alzheimer's Disease Mouse Model.

Background/Objectives: Alzheimer's disease (AD) is currently diagnosed using established biomarkers, such as reduced cerebrospinal fluid (CSF) Aβ42, increased phosphorylated tau, and cerebral amyloid levels detected by PiB-PET. Because these methods are invasive or require specialized facilities, less invasive and easily detectable biomarkers are needed. Flotillin-1 concentrations are reduced in the CSF and serum of patients with AD. This study examined whether flotillin-1 in saliva, a less invasive specimen than blood, could serve as a biomarker. Methods: Wild-type (WT) and AppNL-G-F (APP kno

CSF Proteomics and Machine Learning Reveal Distinct Stages Across the Alzheimer's Disease Continuum.

Alzheimer's disease (AD) is a neurodegenerative disorder characterized by heterogeneous pathophysiological changes that begin years before symptoms emerge. Existing biomarkers like Aβ and pTau capture only fragments of this complexity, limiting diagnosis and therapeutic development. Leveraging high-resolution cerebrospinal fluid (CSF) proteomics, quantifying 2,492 proteins using tandem-mass-tag mass spectrometry (TMT-MS), in 1,104 ADNI participants, we identified pathways reflecting AD pathogenesis and stage-specific molecular events in-vivo. In biomarker-positive MCI (due-to-AD) and AD Dement

NAT10 induces mitochondrial dysfunction in lung epithelial cells by acetylating HMGB1 to exacerbate Pseudomonas aeruginosa-induced acute lung injury.

BACKGROUND: Pseudomonas aeruginosa (PA) is a major pathogen that causes pneumonia and acute lung injury (ALI). Dysregulated NAT10 expression is associated with inflammatory and infectious diseases, but its role in PA-induced ALI remains unclear. METHODS: A mouse pneumonia model was established by intratracheal injection of PA, and lentivirus-mediated NAT10 interference and HMGB1 overexpression vectors were administered via the tail vein. Lung mechanics, protein content, total cell counts, neutrophil counts, inflammatory factor levels in bronchoalveolar lavage fluid (BALF), and lung bacterial

Effects of Dietary Sodium Modulation on Circulating Extracellular Vesicles.

BACKGROUND: Sodium is involved in osmoregulation, fluid balance, and immune system function. High sodium (HS) consumption is linked to endothelial dysfunction, hypertension, and mortality. Extracellular vesicles (EVs), nano-sized particles reflecting cellular status, could link dietary sodium to vascular and immune changes. This study aimed to characterize circulating EVs and assess their functional role on endothelial cells after dietary sodium modulation. METHODS: Fifty subjects underwent 2 dietary sodium interventions, each lasting 5 to 7 days: sodium-restriction (10-40 mmol/d, 0.23-0.92 g

Exosomal Communication Between Cumulus-Oocyte Complexes and Granulosa Cells: A New Molecular Axis for Oocyte Competence in Human-Assisted Reproduction.

Exosomal microRNAs (ex-miRs), encapsulated in extracellular vesicles (EVs), play a vital role in facilitating paracrine communication among granulosa cells (GCs), cumulus cells (CCs), and the oocyte inside follicular fluid (FF). These small non-coding RNAs are crucial for regulating folliculogenesis, oocyte maturation, and early embryonic development via modulating intracellular signaling networks. Dysregulation o has been associated with reproductive disorders such as polycystic ovarian syndrome (PCOS), diminished ovarian reserve (DOR), and inadequate ovarian response (POR), impacting oocyte
