Neuronal Parasitism, Early Myenteric Neurons Depopulation and Continuous Axonal Networking Damage as Underlying Mechanisms of the Experimental Intestinal Chagas' Disease.

There is a growing consensus that the balance between the persistence of infection and the host immune response is crucial for chronification of Chagas heart disease. Extrapolation for chagasic megacolon is hampered because research in humans and animal models that reproduce intestinal pathology is lacking. The parasite-host relationship and its consequence to the disease are not well-known. Our model describes the temporal changes in the mice intestine wall throughout the infection, parasitism, and the development of megacolon. It also presents the consequence of the infection of primary myen

Temporospatial Analysis and New Players in the Immunology of Amyotrophic Lateral Sclerosis.

Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disorder characterized by progressive loss of lower and upper motor neurons (MN) leading to muscle weakness, paralysis and eventually death. Although a highly varied etiology results in ALS, it broadly manifests itself as sporadic and familial forms that have evident similarities in clinical symptoms and disease progression. There is a tremendous amount of knowledge on molecular mechanisms leading to loss of MNs and neuromuscular junctions (NMJ) as major determinants of disease onset, severity and progression in ALS. Specifically, two

Neuronal overexpression of human VAPB slows motor impairment and neuromuscular denervation in a mouse model of ALS.

Four mutations in the VAMP/synaptobrevin-associated protein B (VAPB) gene have been linked to amyotrophic lateral sclerosis (ALS) type 8. The mechanism by which VAPB mutations cause motor neuron disease is unclear, but studies of the most common P56S variant suggest both loss of function and dominant-negative sequestration of wild-type protein. Diminished levels of VAPB and its proteolytic cleavage fragment have also been reported in sporadic ALS cases, suggesting that VAPB loss of function may be a common mechanism of disease. Here, we tested whether neuronal overexpression of wild-type human

Acute motor axonal neuropathy cases in Van region.

Acute motor axonal neuropathy (AMAN) is a form of Guillain Barré Syndrome (GBS) seen in summer months in Northern China to cause epidemics. This form of the disease, which is also sporadically observed in other countries, constitutes less than 5% of GBS in Western countries. It usually develops with motor findings. No sensory findings are observed. In some of the cases, the severe impairments in tissues improve however slowly and inadequately. In the motor conduction studies of cases with AMAN, motor action potential values are lowered. On needle electromyography (EMG), motor unit potential (M

Botulinum toxin B treatment in children with spastic movement disorders: a pilot study.

The treatment of adult and pediatric patients suffering from movement disorders with elevated muscle tone includes the application of focally denervating botulinum toxins. Dystonic movement disorders in adult patients have been treated successfully using botulinum toxin type B (NeuroBloc). Thus far, there has been no systematic treatment of children with botulinum toxin type B. This study reports on the treatment of 29 children with spastic or dystonic movement disorders using botulinum toxin type B in an open-label pilot study. Sixty-two treatment sessions were performed. In 33 of these sessi

Effect of subepineurial dehydroepiandrosterone treatment on healing of transected nerves repaired with the epineurial sleeve technique.

The epineurial sleeve technique for nerve repair is designed in part to protect a healing nerve from external humoral influences, but research suggests that the external factor dehydroepiandrosterone (DHEA) may actually improve nerve healing in crush injuries. To test the effect of DHEA, we injected it into the epineurial chambers created to repair transected rat sciatic nerves. In 18 control rats, the nerve was transected and repaired without DHEA treatment. Eighteen animals received subepineurial injections of propylene glycol vehicle, and 18 received subepineurial injections of about 0.2 ml

[Influence of adrenergic denervation on uterine contractility of pregnant rats in vitro].

The aim of the present study was to investigate the functional changes in the sympathetic nerves in the rat myometrium during pregnancy by electric field stimulation (EFS) in vitro. In our experiments the contractility of the rat myometrium were registered at 5, 10, 15, 18, 20, 22 (term) day of pregnancy, in non pregnant rats and in 6-OH-dopamine pretreated rats. The parameters of EFS were the following: pulse width 0.6 ms, frequency range of nerve stimulation 1-70 Hz, supramaximal voltage 40 V. We concluded that low pulse width and low frequency stimulation is selective for adrenergic nerves.

Practical guidelines to safe surgery about the subscapularis.

Surgical mobilization of an adherent or retracted subscapularis tendon is often necessary whenever an attempt is made to restore function to the glenohumeral joint subsequent to failed anterior reconstructive surgery. Surgical guidelines drawn from this study emphasize how a surgeon might accomplish this task effectively and avoid denervating the subscapularis, a muscle that is essential to anterior glenohumeral stability and strength. In this study we examined subscapularis innervation in 11 fresh-frozen cadaveric shoulders. The position of the subscapular nerve insertion points were recorded

Widespread neural excitation initiated from cardiac spinal afferent nerves.

Reflex effects of cardiac sympathetic afferent nerve excitation on cardiac and renal components of sympathetic outflow have been well investigated, but the extent of these cardiac afferent influences on other sympathetic nerves or on respiratory or cortical neural systems is unknown. Therefore such influences were investigated electrophysiologically in anesthetized, vagotomized, baroreceptor-denervated cats. Stimulation of cardiac sympathetic spinal afferent neurons by the noxious substance bradykinin caused excitation of cardiac, renal, splenic, gastrohepatic, adrenal, and deep peroneal (musc

Post-traumatic facial nerve paralysis: three cases of delayed temporal bone exploration with recovery.

No consensus exists today on the management of immediate-onset post-traumatic facial nerve paralysis. Controversy surrounds the timing of surgical intervention and the role of electrophysiologic testing. Three patients are presented who sustained immediate, complete facial paralysis following closed head trauma. They did not have prompt facial nerve decompression. In each case, electroneurography and electromyography showed complete nerve degeneration and denervated muscle. Despite the results of the electrophysiologic tests, all patients underwent late surgical decompression of the nerve: one

Control of genioglossus muscle inspiratory activity.

Tonic and phasic inspiratory genioglossus (GG) electromyographic activity (EMG) was recorded from 13 anesthetized rabbits during unstimulated breathing. Integrated GG EMG peaked earlier in inspiration and presented a more rounded contour than integrated diaphragmatic (DIA) EMG. Spontaneous augmented deep inspirations (sighs) showed a biphasic pattern in both GG and DIA EMGs. Hyperventilation abolished phasic inspiratory activity in the GG before the DIA, suggesting that the GG has a higher CO2 apneic point. Hypercapnia increased both EMGs; however, GG EMG increased more, as a percent of base l

Orbicularis oculi reflex and facial muscle electromyography. Pre- and postoperative evaluation of posterior fossa space-occupying lesions.

The electrically evoked orbicularis oculi reflex was analyzed in 26 patients with proven posterior fossa space-occupying lesions (11 cerebellopontine angle and 15 cerebellar hemispheric masses). In the former group, although only six had clinical evidence of trigeminal or facial involvement, the reflex analysis showed both nerves to be involved in all. Electromyography (EMG) of the orbicularis oculi showed profound denervation of this muscle while only minimal abnormalities of the masseter were seen. The sensory component of the trigeminal nerve was always more involved than its motor componen

Defining Endogenous Mitochondrial Transfer in Muscle After Rotator Cuff Injury.

BACKGROUND: Rotator cuff muscle degeneration leads to poor clinical outcomes for patients with rotator cuff tears. Fibroadipogenic progenitors (FAPs) are resident muscle stem cells with the ability to differentiate into fibroblasts as well as white and beige adipose tissue. Induction of the beige adipose phenotype in FAPs has been shown to improve muscle quality after rotator cuff tears, but the mechanisms of how FAPs exert their beneficial effects have not been fully elucidated. PURPOSE: To study the horizontal transfer of mitochondria from FAPs to myogenic cells and examine the effects of β

Longitudinal transcriptomic analysis of mouse sciatic nerve reveals pathways associated with age-related muscle pathology.

BACKGROUND: Sarcopenia, the age-associated decline in skeletal muscle mass and strength, has long been considered a disease of muscle only, but accumulating evidence suggests that sarcopenia could originate from the neural components controlling muscles. To identify early molecular changes in nerves that may drive sarcopenia initiation, we performed a longitudinal transcriptomic analysis of the sciatic nerve, which governs lower limb muscles, in aging mice. METHODS: Sciatic nerve and gastrocnemius muscle were obtained from female C57BL/6JN mice aged 5, 18, 21 and 24 months old (n = 6 per age

Neuromuscular effects of repeated exposure to the pesticide methyl bromide.

Methyl bromide (MB) is a fumigant pesticide that remains in use for quarantine and pre-shipment applications. Although acute intoxication is associated with motor dysfunction, the effects of repeated occupational inhalation exposure on neuromuscular structures remain poorly characterized. This study evaluated functional and anatomical consequences of repeated MB inhalation exposure in adult male Sprague-Dawley rats. Animals were exposed to either clean air or 300 ppm MB by nose-only inhalation for 2 hours a day, 5 days a week, over 4 weeks. Neurobehavioral assessments included functional obser

Effectiveness and safety of early intramuscular botulinum toxin injections to prevent shoulder deformity in babies with brachial plexus birth injury (POPB-TOX), a randomised controlled trial: study protocol.

INTRODUCTION: In children with brachial plexus birth injury (BPBI), denervation of the shoulder muscles leads to bony deformity in the first months of life, reducing active and passive range of motion (ROM) and causing activity limitation. The aim of this multicentre randomised controlled trial is to evaluate the effectiveness of botulinum toxin injections (BTI) in the shoulder internal rotator muscles of 12-month-old babies in limiting the progression of posterior subluxation of the glenohumeral joint, compared with a sham procedure mimicking BTI. The secondary aims are to evaluate the effect

Changes in electromyographic results of patients with lumbar radiculopathy: a follow-up study.

OBJECTIVE: To assess the neurophysiologic changes in a group of patients with lumbar radiculopathy 5 to 12 months after their first electromyographic examination. DESIGN: A prospective group of patients with a case definition of lumbar radiculopathy was reassessed between 5 and 12 months after their first clinical, functional, imaging, and neurophysiologic evaluation. Both the lumbar paraspinals (in which the mini-mapping technique was used) and the same lower limb muscles were explored in every patient. Relevant abnormalities were (1) positive sharp waves/fibrillation potentials, (2) polypha

Nonpoliovirus poliomyelitis simulating Guillain-Barré syndrome.

BACKGROUND: Paralytic poliomyelitis due to the wild-type poliovirus has been eradicated in the United States because of effective immunization programs. In the postvaccination era, most cases are caused by other RNA viruses, such as coxsackievirus or echovirus. The condition usually begins with a fever and upper respiratory tract or gastrointestinal tract symptoms that progress to a "paralytic" phase characterized by limb weakness, areflexia, and, occasionally, respiratory failure that superficially resemble Guillain-Barré syndrome. OBJECTIVE: To describe 2 patients with nonpoliovirus poliomy
