Centrosome-related gene SPHK1 drives bladder cancer progression and therapeutic vulnerability.

BACKGROUND: Centrosome-related genes (CRGs) regulate cell division and genomic stability and may influence tumor progression, but their prognostic and functional roles in bladder cancer (BLCA) are not fully defined. METHODS: We curated 698 CRGs and identified 12 prognostic genes to construct a centrosome-related signature score (CRSS). The prognostic value of CRSS was evaluated in TCGA and GSE13507 cohorts. Functional enrichment, immune landscape, and drug sensitivity were analyzed through pathway analysis, ESTIMATE, and IC50 prediction. Machine learning and Mendelian randomization were used

Stage-specific EEG spectral signatures of primary insomnia: regional and sex-related differences across NREM and REM sleep.

Sleep architecture is characterized by dynamic transitions between non-rapid eye movement (NREM) and rapid eye movement (REM) stages, each defined by distinct oscillatory signatures. Although quantitative EEG has been widely used to study primary insomnia (PI), reported findings remain inconsistent, partly due to limited consideration of cortical regional differences, sleep-stage specificity, and sex-related variability. This study integrates spectral power analysis with explainable machine learning to provide a comprehensive characterization of neural dynamics in PI. EEG recordings from 100 a

Single-cell and machine learning identify a trihydroxybutyrylation-related prognostic signature in esophageal cancer.

BACKGROUND: Esophageal squamous cell carcinoma (ESCC) is a highly aggressive malignancy with a poor prognosis. This study identifies malignant epithelial subtypes and establishes prognostic biomarkers through single-cell transcriptomics and integrative machine learning approaches. METHODS: We re-analyzed the single-cell RNA sequencing data (GSE188900) and identified malignant epithelial cells using inferCNV. Differentially expressed genes (DEGs) among malignant epithelial subtypes were identified and intersected with lysine β-hydroxybutyrylation (Kbhb)-related genes. Candidate genes were scre

Integrative Transcriptomic Analysis and Single-Cell Characterization Identify RTN4 as a Candidate PBMC-Derived Hub Gene Associated with COPD and Coronary Artery Disease.

Background: Chronic obstructive pulmonary disease (COPD) is associated with systemic inflammation and increased cardiovascular comorbidity, yet peripheral blood molecular markers for cardiovascular comorbidity-related stratification in COPD remain poorly defined. Methods: In this study, PBMC transcriptomic datasets from a COPD cohort (GSE42057) and a CAD cohort (GSE113079) were analyzed using weighted gene co-expression network analysis (WGCNA) to identify disease-associated modules, followed by overlapping gene screening. Machine-learning models were then applied to prioritize the shared gene

Effects of Central Retainer Depth on Stress Distribution and Fracture Resistance in Endocrown-restored Maxillary Premolars: 3D Finite Element and in vitro Analysis.

OBJECTIVE: To explore how central retainer depth affects stress distribution and fracture resistance in endocrown-restored premolars. METHODS: A 3D finite element (FE) model of an intact maxillary premolar was established as a negative control. Five endocrown-restored FE models were established, differing in the extension of the central retainer inside the pulp chamber: 0, 2, 3, 4 and 5 mm. The remaining pulp cavity was filled with composite resin. Stress distributions were analysed under oblique loads. Thirty freshly extracted human maxillary premolars were randomly divided into three groups

Metabolic signatures in osteoporotic cancellous bone: a comprehensive dual-platform metabolomic analysis.

BACKGROUND: Osteoporosis represents a major global health challenge with increasing prevalence and substantial healthcare burden. Current diagnostic approaches relying primarily on bone mineral density measurements have limitations for early disease detection and understanding pathophysiological mechanisms. We conducted comprehensive metabolomic analysis of osteoporotic cancellous bone to identify potential metabolic features and explore disease mechanisms. METHODS: Hip cancellous bone samples from 18 osteoporotic patients and 18 age-matched non-osteoporotic controls were analyzed using dual-

In Vitro Evaluation of the Fracture Resistance of CAD/CAM Materials for Fixed Partial Dentures.

BACKGROUND: Computer-aided design/computer-aided manufacturing (CAD/CAM) materials have revolutionized dental restorations, offering enhanced mechanical properties and esthetic outcomes. However, the fracture resistance of various CAD/CAM materials used in fixed partial dentures (FPDs) remains a crucial determinant of their clinical success. MATERIALS AND METHODS: Three CAD/CAM materials-lithium disilicate, zirconia, and resin-based composite-were selected for the study. A total of 45 three-unit FPD frameworks (15 specimens for each material) were fabricated using standardized CAD/CAM protoco

Automated gall bladder cancer detection using artificial gorilla troops optimizer with transfer learning on ultrasound images.

The gallbladder (GB) is a small pouch and a deep tissue placed under the liver. GB Cancer (GBC) is a deadly illness that is complex to discover in an initial phase. Initial diagnosis can significantly enhance the existence rate. Non-ionizing energy, low cost, and convenience make the US a general non-invasive analytical modality for patients with GB diseases. Automatic recognition of GBC from US imagery is a significant issue that has gained much attention from researchers. Recently, machine learning (ML) techniques dependent on convolutional neural network (CNN) architectures have prepared tr

Machine learning identifies esophageal luminal temperature patterns associated with thermal injury in catheter ablation for atrial fibrillation.

INTRODUCTION: Luminal esophageal temperature (LET) monitoring during atrial fibrillation (AF) ablation is widely used to reduce the incidence of endoscopically detected esophageal lesion (EDEL). We sought to assess whether specific patterns of LET variation are associated with EDEL. METHODS: A high-fidelity multisensor probe was used to record LET in AF patients undergoing radiofrequency ablation (RFA) or cryoballoon ablation (CBA). Explainable machine learning and SHapley Additive exPlanations (SHAP) analysis were used to predict EDEL and assess feature importance. RESULTS: A total of 94 pa

Lifelong olfactory deprivation-dependent cortical reorganization restricted to orbitofrontal cortex.

Prolonged sensory deprivation has repeatedly been linked to cortical reorganization. We recently demonstrated that individuals with congenital anosmia (CA, complete olfactory deprivation since birth) have seemingly normal morphology in piriform (olfactory) cortex despite profound morphological deviations in the orbitofrontal cortex (OFC), a finding contradictory to both the known effects of blindness on visual cortex and to the sparse literature on brain morphology in anosmia. To establish whether these unexpected findings reflect the true brain morphology in CA, we first performed a direct re

Self-calibrated brain network estimation and joint non-convex multi-task learning for identification of early Alzheimer's disease.

Detection of early stages of Alzheimer's disease (AD) (i.e., mild cognitive impairment (MCI)) is important to maximize the chances to delay or prevent progression to AD. Brain connectivity networks inferred from medical imaging data have been commonly used to distinguish MCI patients from normal controls (NC). However, existing methods still suffer from limited performance, and classification remains mainly based on single modality data. This paper proposes a new model to automatically diagnosing MCI (early MCI (EMCI) and late MCI (LMCI)) and its earlier stages (i.e., significant memory concer

Separability of motor imagery of the self from interpretation of motor intentions of others at the single trial level: an EEG study.

BACKGROUND: We aimed to investigate the separability of the neural correlates of 2 types of motor imagery, self and third person (actions owned by the participant himself vs. another individual). If possible this would allow for the development of BCI interfaces to train disorders of action and intention understanding beyond simple imitation, such as autism. METHODS: We used EEG recordings from 20 healthy participants, as well as electrocorticography (ECoG) in one, based on a virtual reality setup. To test feasibility of discrimination between each type of imagery at the single trial level, t

Bisphenol A exposure in myasthenia gravis: Potential targets and mechanisms revealed by network toxicology and molecular dynamics.

BACKGROUND: Myasthenia gravis (MG) is a B-cell-mediated autoimmune disease characterized by impaired neuromuscular transmission. Although genetic predisposition and thymic abnormalities are well recognized, they cannot fully explain the increasing incidence and regional heterogeneity of MG, highlighting the potential contribution of environmental factors. Bisphenol A (BPA), a ubiquitous endocrine-disrupting chemical, exhibits estrogenic activity, immunomodulatory effects, and mitochondrial toxicity, and has been implicated in multiple autoimmune disorders. However, the potential role of BPA in

Methylome profiling reveals context-dependent chemo-resistance mechanisms and enhances risk stratification in AML.

BACKGROUND: Acute Myeloid Leukemia (AML) is the second most lethal hematologic malignancy, with approximately 30% of patients being refractory to first-line induction therapy. While current risk stratification systems, such as European LeukemiaNet (ELN), use cytogenetic and molecular markers to guide treatment decisions, their predictive accuracy remains suboptimal, particularly in forecasting treatment response. METHODS: In this study, we performed integrated methylome and transcriptome analysis of diagnostic samples from 146 AML patients, including methylome data on 25 patients, to investiga

Polyethylene terephthalate microplastics exposure enhances the risk of ulcerative colitis: insights from multiomics integration, machine learning, and molecular docking reveal intestinal toxicity mechanisms.

BACKGROUND: In recent decades, the widespread use of polyethylene terephthalate (PET) has led to the proliferation of PET microplastics (PET-MPs) in the environment, posing potential threats to human health, including ulcerative colitis (UC). However, the underlying molecular mechanisms remain unclear. METHODS: Data sourced from public databases were harnessed to predict the targets of PET-MPs and identify UC-related differentially expressed genes. Functional enrichment analyses were conducted to uncover relevant biological pathways. Three machine learning (ML) algorithms were utilized to scr

Prenatal-to-childhood urinary phenol and metal biomarkers in relation to BMI trajectories in a 10-year birth cohort.

Early-life exposure to phenols and metals is widespread, but evidence linking repeated exposure across development to long-term growth patterns remains limited. We analyzed 509 mother-child pairs from the Sheyang Mini Birth Cohort Study, in which urinary concentrations of three phenols (BPA, triclosan, and BP-3) and four metals (Pb, Cd, As, and Cr) were measured during pregnancy and at ages 1, 3, 8, and 10 years. BMI z-score trajectories were identified using latent class trajectory modeling, and associations were evaluated using Firth's logistic regression. Bayesian kernel machine regression

Association of sleep symptom burden with default mode network homogeneity in first-episode drug-naïve adolescents with major depressive disorder.

BACKGROUND: The default mode network (DMN) plays an important role in depression and sleep regulation. However, whether sleep symptoms are associated with alterations in DMN homogeneity in adolescent major depressive disorder (MDD) remains unclear. This study aimed to investigate whether sleep symptom burden is associated with alterations in DMN homogeneity in adolescents with MDD. METHODS: Following quality control procedures, the final sample comprised 162 first-episode, drug-naïve adolescents with MDD, including 86 participants with higher sleep symptom burden (MDD-S) and 76 participants w
