Tsutsugamushi disease is an infectious disease transmitted to humans through the bite of the Orientia tsutsugamushi-infected chigger mite; however, host-pathogen interactions and the precise mechanisms of damage in O. tsutsugamushi infections have not been fully elucidated. Here, we analyzed the global metabolic effects of O. tsutsugamushi infection on the host using 1H-NMR and UPLC-Q-TOF mass spectroscopy coupled with multivariate statistical analysis. In addition, the effect of O. tsutsugamushi infection on metabolite concentrations over time was analyzed by two-way ANOVAs. Orthogonal partia

BACKGROUND: Scrub typhus is a leading cause of serious febrile illness in rural Southeast Asia. The causative agent, Orientia tsutsugamushi, is an obligate intracellular bacterium that is transmitted to humans by the bite of a Leptotrombidium mite. Research into the basic mechanisms of cell biology and pathogenicity of O. tsutsugamushi has lagged behind that of other important human pathogens. One reason for this is that O. tsutsugamushi is an obligate intracellular bacterium that can only be cultured in mammalian cells and that requires specific methodologies for propagation and analysis. Her…

Tsutsugamushi disease is an acute febrile infectious disease caused by Rickettsia tsutsugamushi. An infection is heralded by the presence of an eschar at the site of the inoculating chigger bite and followed by the development of a disseminated erythematous macular rash. CD30 expression is found in anaplastic large cell lymphoma; however, expression in nonneoplastic cutaneous disorders, such as atopic dermatitis, drug reactions, scabies, and various infectious diseases, has also been reported. Study of the cutaneous histopathology of tsutsugamushi disease has been limited. In this study, we pe

The development and persistence of antibodies to Orientia tsutsugamushi in Rattus rattus and laboratory mice following infection from the bite of naturally infected Leptotrombidium deliense is reported. Antibodies in R. rattus were first detected using an indirect immunoperoxidase method 2 weeks after attachment of an infected mite. The IgM antibody response was mild, and was detected 2, 4 and 6 weeks after infection, while the IgG response was stronger and was detected from 2 to 19 weeks after infection, when the tests ceased. Active rickettsiae were isolated from R. rattus 1-8 weeks after at

OBJECTIVE: In order to provide basic data of tsutsugamushi disease in Shandong province, for the control and prevention of this disease in residents and army-men.
METHODS: Aetiological, serological and epidemiological methods were used to investigate the natural foci of autumn-winter type tsutsugamushi disease.
RESULTS: The peak of tsutsugamushi disease epidemic in Shandong province was in October with cases in this month accounted for 80% of all in a year, that showed autumn-winter type. The main reservoir hosts were Apodemus agrarius, Rattus norvegicus and Cricetulus triton, accounted for

In 1986, investigating tsutsugamushi disease in the suburbs of Hangzhou, We isolated Rickettsia tsutsugamushi from leptotrombidium gaohuense and its reservoir hosts, Rattus confucianus and Rattus fulvescens, which were collected in Yang Shan Wu village, Linan county. It proved there was a natural focus of tsutsugamushi disease in the suburbs of Hang zhou. Which is located at 30 degrees 35'. North latitude, and main forest of the East Tianmu Mountain, covered by wide and needle leaves, with more wide leaves than needle one. The vector is leptotrombidium gaohuense. The host are Rattus confucianu

The first case of tsutsugamushi disease in Ehime Prefecture was experienced in December 1987 with successful isolation of the causative agent. The patient was taken ill twelve days after infection. Immunofluorescent antibody tests using the isolate, Yamazaki strain, and Gilliam, Karp, Kato, Irie and Shimokoshi strains as antigens revealed that the specific antibodies against these antigens appeared and increased in the blood of the patient during the course of the disease. And the antibody titers to the Yamazaki antigen were the highest of these antigens. Agglutinin for Proteus OXK did not app

Scrub typhus is a febrile disease caused by Orientia tsutsugamushi, transmitted by larval stage Trombiculid mites (chiggers), whose primary hosts are small mammals. The phylogenomics of O. tsutsugamushi in chiggers, small mammals and humans remains poorly understood. To combat the limitations imposed by the low relative quantities of pathogen DNA in typical O. tsutsugamushi clinical and ecological samples, along with the technical, safety and cost limitations of cell culture, a novel probe-based target enrichment sequencing protocol was developed. The method was designed to capture variation a…

Many infectious/communicable diseases (IDs) are endemic in Nepal. Until a decade and half ago, IDs were the major cause of both morbidity and mortality accounting 70% for both. However, as a result of various preventive measures implemented by both the state and non-state actors, the overall IDs have shown a changing (declining) trend. The most impressive decline has been seen in the intestinal helminth infection. Though the overall burden of IDs is decreasing, several newer infectious diseases (emerging infections) namely, dengue fever, scrub typhus, influenza (H5N1 and H1N1), and others are …

Orientia tsutsugamushi is an intracellular parasite that causes scrub typhus. After entering the cytoplasm by induced phagocytosis, O. tsutsugamushi escapes from the primary phagosome into the host cytosol, where it replicates slowly. Subsequently, it is released from the host cells by a process resembling viral budding with a remaining bacterial aggregate near the nucleus. Lipid rafts have been implicated in the life cycle of a wide variety of pathogenic microorganisms. We have observed that proteins of O. tsutsugamushi were co-fractionated with the lipid rafts over a sucrose density gradient…

BACKGROUND: Leptospirosis is a zoonotic infection caused by pathogenic Leptospira species and remains a significant cause of acute febrile illness in tropical regions. Its clinical spectrum ranges from mild, self-limiting illness to severe multi-organ dysfunction. The disease is broadly classified into anicteric and icteric (Weil's disease) forms, the latter associated with jaundice and increased morbidity. In India, diagnosis is often challenging due to overlapping features with other endemic infections such as dengue and scrub typhus. Coinfections and changing epidemiological patterns, inclu…
