Molecular dynamic simulations to assess the structural variability of ClpV from Enterobacter cloacae.

The Enterobacter cloacae complex (ECC) consists of six Enterobacter species (E. cloacae, hormaechei, kobei, ludwigii, nimipressuralis and asburiae) that have emerged as nosocomial pathogens of interest, with E. cloacae and Enterobacter hormachei being the most frequently isolated ECC species in human clinical specimens and intensive care unit (ICU) patients. Many nosocomial outbreaks of E. cloacae have been related to transmission through contaminated surgical equipment and operative cleaning solutions. As this pathogen evades the action of antibiotics, it is important to find alternative targ

Acaricides in agriculture: balancing livestock health and environmental well-being in Trans-Nzoia County, Kenya.

Extensive use of chemicals in food production, although useful, has profound implications. Acaricides, extensively used to control ticks and mites in livestock farming, can leave harmful residues that pose risks to unintended organisms such as plants, insects, people, and other animals. Thus, limiting non-target exposure to acaricides is critical. This study is aimed at assessing acaricide knowledge, use, and disposal methods among farmers and agrochemical dealers in Trans-Nzoia County, Kenya, which is well-known for livestock farming. A purposive survey of stakeholders, including 100 farmers

Genetic Characteristics of Acinetobacter baumannii Isolates Circulating in an Intensive Care Unit of an Infectious Diseases Hospital During the COVID-19 Pandemic.

During the COVID-19 pandemic, a significant increase in the spread of healthcare-associated infections (HAIs) and antimicrobial resistance (AMR) was observed. Acinetobacter baumannii, particularly carbapenem-resistant strains, poses a serious threat in intensive care units (ICUs). This study aimed to genetically characterize A. baumannii isolates from the ICU of an infectious diseases hospital repurposed for COVID-19 patient treatment. Whole-genome sequencing (WGS) was performed on 56 A. baumannii isolates from patients and environmental surfaces using the Illumina MiSeq platform. Bioinformati

Development of an effective simulation model for evaluating smoke leakage during laparoscopic surgery.

The leakage of surgical gas and smoke from the peritoneum during laparoscopy may release noxious aerosols, including potential carcinogens, viruses and other contaminants, into the operating theatre, especially into the breathing zone of the surgical team. Reliable and realistic models and methods that develop and detect surgical smoke in simulated settings are necessary to effectively test devices and strategies intended to reduce such leaks. Here, we report a novel high-fidelity laparoscopic smoke model with innovative imaging methods applicable to the theatre setting, followed by an assessm

Development and optimization of an antibody-free nucleic acid lateral flow assay (AF-NALFA) as part of a molecular toolkit for visual readout of amplified Listeria monocytogenes DNA.

Listeria monocytogenes is a Gram-positive foodborne pathogen responsible for listeriosis, a severe disease with high mortality in immunocompromised individuals. Rapid and accurate detection in food samples is essential for food safety. In this study, we developed and optimized an Antibody-Free Nucleic Acid Lateral Flow Assay (AF-NALFA) as part of a molecular detection toolkit for the visual readout of amplified L. monocytogenes hlyA gene, in combination with ultra-fast asymmetric PCR (aPCR) and oligonucleotide probe hybridization. Three critical parameters were optimized: oligonucleotide probe

Advances in the Detection and Identification of Bacterial Biofilms Through NIR Spectroscopy.

Bacterial biofilms play an important role in the pathogenesis of infectious diseases but are also very relevant in other fields such as the food industry. This fact has led to an increased focus on the early identification of these structures as prophylaxes to prevent biofilm-related contaminations or infections. One of the objectives of the present study was to assess the effectiveness of NIR (Near Infrared) spectroscopy in the detection and differentiation of biofilms from different bacterial species, namely Staphylococcus epidermidis, Staphylococcus aureus, Enterococcus faecium, Salmonella

Detection, survival, and persistence of Staphylococcus capitis NRCS-A in neonatal units in England.

BACKGROUND: The multidrug-resistant Staphylococcus capitis clone, NRCS-A, is increasingly associated with late-onset sepsis in low birthweight newborns in neonatal intensive care units (NICUs) in England and globally. Understanding where this bacterium survives and persists within the NICU environment is key to developing and implementing effective control measures. AIM: To investigate the potential for S. capitis to colonize surfaces within NICUs. METHODS: Surface swabs were collected from four NICUs with and without known NRCS-A colonizations/infections present at the time of sampling. Sam

A systematic literature review of milk consumption and associated bacterial zoonoses in East Africa.

Consumption of unsafe animal-source foods is the major cause of foodborne disease outbreaks in low-income countries. Despite current knowledge of the threat posed by raw milk consumption to human health, people in many countries in East Africa still consume unboiled milk. This literature review explored the association between milk consumption and the occurrence of five milk-borne bacterial zoonoses: brucellosis, salmonellosis, campylobacteriosis, Escherichia coli infections, and tuberculosis. A search for literature published up to 1 October 2021 was conducted through the Web of Science, PubM

Extrahepatic Bile Duct and Gall Bladder Dissection in Nine-Day-Old Mouse Neonates.

The dissection of murine neonatal bile ducts has been described as difficult. The main aim of the described standard operating procedure is the isolation of the extrahepatic bile duct (EBD) in mouse neonates without damaging the bile duct during preparation. Because of its exceptionally close preparation compared to the cholangiocytes cell line and harvesting of the entire extrahepatic bile duct system (EBDS), the described approach is extremely useful in researching animal models of newborn bile duct disorders, such as biliary atresia. After euthanasia, the peritoneal cavity was accessed, and

Personal Protective Equipment and Donning and Doffing Techniques in the Cardiac Catheterization Laboratory During the COVID-19 Pandemic: Insights From an Internet Search for Protocols.

Background: Due to the ongoing coronavirus disease 2019 (COVID-19) pandemic, a need for precise donning and doffing protocols for personal protective equipment (PPE) among healthcare infrastructures is paramount. Procedures involving the cardiac catheterization laboratory (CCL) are routinely non-aerosolizing but have the potential for rapid patient deterioration, creating the need for aerosolizing generating procedures. Multiple societal and governmental guidelines on the use of PPE during medical procedures are available on Internet websites; however, there is limited literature available in

Re-purposing of domestic steam disinfectors within the hospital-at-home setting.

BACKGROUND: Vulnerable patients being cared for in hospital-at-home settings require safe disinfection of their medical devices, including nebulisers and other respiratory equipment. The scale of patients now being cared for in hospital-at-home settings as a result of COVID19 places huge pressure on hospital central sterile services departments (CSSDs) to provide consumable items to safely support such patients' care. This places new importance on the disinfection of mundane objects, including crockery, cutlery and frequently touched objects in the home environment. This study examined tempera

Hepatitis C antibody prevalence among Mexico City prisoners injecting legal and illegal substances.

BACKGROUND: Hepatitis C virus (HCV) infection is highly prevalent among prisoners and this prevalence estimates reach 64% among prisoners who inject illicit drugs. Prisons are important sites for HCV transmission in the absence of access to sterile injecting equipment; hence, it can be transmitted between prisoners who share contaminated needles and syringes. We aimed to estimate the prevalence of risk factors for anti-HCV prevalence, with particular interest on injecting behavior, and to assess correlates of anti-HCV positivity among Mexico City prisoners. METHODS: Cross-sectional study base

Motor deficits, impaired response inhibition, and blunted response to methylphenidate following neonatal exposure to decabromodiphenyl ether.

Decabromodiphenyl ether (decaBDE) is an applied brominated flame retardant that is widely-used in electronic equipment. After decades of use, decaBDE and other members of its polybrominated diphenyl ether class have become globally-distributed environmental contaminants that can be measured in the atmosphere, water bodies, wildlife, food staples and human breastmilk. Although it has been banned in Europe and voluntarily withdrawn from the U.S. market, it is still used in Asian countries. Evidence from epidemiological and animal studies indicate that decaBDE exposure targets brain development a

Sensitivity and specificity of real-time reverse transcription polymerase chain reaction, histopathology, and immunohistochemical labeling for the detection of Rift Valley fever virus in naturally infected cattle and sheep.

Real-time reverse transcription polymerase chain reaction (real-time RT-PCR), histopathology, and immunohistochemical labeling (IHC) were performed on liver specimens from 380 naturally infected cattle and sheep necropsied during the 2010 Rift Valley fever (RVF) epidemic in South Africa. Sensitivity (Se) and specificity (Sp) of real-time RT-PCR, histopathology, and IHC were estimated in a latent-class model using a Bayesian framework. The Se and Sp of real-time RT-PCR were estimated as 97.4% (95% confidence interval [CI] = 95.2-98.8%) and 71.7% (95% CI = 65-77.9%) respectively. The Se and Sp o

Control of blood-transmitted infections in dentistry.

As knowledge has accumulated on the blood-transmitted pathogenic agents, the contact with biological fluids (blood, plasma, saliva, etc.) from apparently healthy individuals has started to be regarded as a real professional risk for dentists. Theoretically, exposure to a contaminated biological specimen may have as a consequence transmission of infection from patient to dentist, from dentist to patient and from patient to patient via inadequately decontaminated and sterilized dental equipment. The present study is concerned with the analysis of the specific conditions that favor the occurrence

Social and environmental risk factors in the emergence of infectious diseases.

Fifty years ago, the age-old scourge of infectious disease was receding in the developed world in response to improved public health measures, while the advent of antibiotics, better vaccines, insecticides and improved surveillance held the promise of eradicating residual problems. By the late twentieth century, however, an increase in the emergence and re-emergence of infectious diseases was evident in many parts of the world. This upturn looms as the fourth major transition in human-microbe relationships since the advent of agriculture around 10,000 years ago. About 30 new diseases have been
