The heterogeneity of the mesostriatal dopaminergic system as revealed in normal and parkinsonian monkeys.

The present immunohistochemical study has shown that the mesostriatal DA system in primates is a highly heterogeneous entity composed of several subsystems displaying variable degrees of vulnerability to MPTP. These subsystems were shown to affect the expression of neuropeptide in the striatofugal neurons in a manner that varies according to the location of the cell bodies in the various striatal territories. Also, a distinct subset of DA neurons in the SN/VTA complex was found to specifically innervate GPi and to be relatively resistant to the neurotoxic effect of MPTP. Furthermore, DA neuron

Sex differences in subcortical auditory processing emerge across development.

Human subcortical auditory processing is sexually dimorphic. The prevailing view - that sex differences arise from cochlear differences - remains unproven, and the extent to which these differences reflect distinct auditory processes is unknown. To determine the origin of subcortical sex differences, we mapped their emergence onto the peripheral-to-central maturation of the auditory system in 516 participants (250 female) across three age groups: 3-5, 14-15, and 22-26 years. To examine whether these sex differences arise from distinct processes, we compared developmental trajectories of each e

Fetal Cerebral Ventricular Dilatation: Etiopathogenic Study of 130 Observations.

BACKGROUND: Fetal cerebral ventricular dilatation (CVD) is a common abnormal prenatal finding that often predicts a poor prognosis. The etiology involves both genetic and nongenetic factors with diverse pathogenic mechanisms. We describe the neuropathological features of CVD in a large cohort of fetuses. The goals are to determine the physiopathological mechanisms and etiologies. METHODS: We retrospectively analyzed a series of 130 fetuses examined at the Necker University Hospital following termination of pregnancy between January 2000 and December 2014. Chiari II and Dandy-Walker malformati

Effect of itraconazole on mouse mesencephalic neurons.

Neuronal cells have complex geometrical shapes, long processes such as axons and dendrites, and as a response to specific stimuli, they go through polarized neuronal migration that influences connectivity and information processing. Recently, it has been discovered that itraconazole, a widely used systemic antifungal drug, has an effect on cell morphology, acting as an inhibitor of the morphogen Sonic Hedgehog (Shh) and of the mammalian target of rapamycin mTOR pathways. In this paper we evaluated the effect of itraconazole on mouse mesencephalic dopaminergic neurons following their neurite ou

Perimesencephalic subarachnoid hemorrhage: risk factors, clinical presentations, and outcome.

BACKGROUND: Perimesencephalic nonaneurysmal subarachnoid hemorrhage (PNSH) appears to have an origin and natural history distinct from aneurysm rupture. However, the risk factors and complications of this pattern are still in debate. We performed a study with goals of comparing PNSH risk factors and clinical presentations with other sorts of spontaneous subarachnoid hemorrhages (SAH) and exhibit the PNSH outcome and prognosis. METHODS: Retrospective review of patients who experienced SAH between May 2006 and July 2008 in the First Affiliated Hospital of Chongqing Medical University was undert

Non-invasive evaluation of nigrostriatal neuropathology in a proteasome inhibitor rodent model of Parkinson's disease.

BACKGROUND: Predominantly, magnetic resonance imaging (MRI) studies in animal models of Parkinson's disease (PD) have focused on alterations in T2 water 1H relaxation or 1H MR spectroscopy (MRS), whilst potential morphological changes and their relationship to histological or behavioural outcomes have not been appropriately addressed. Therefore, in this study we have utilised MRI to scan in vivo brains from rodents bearing a nigrostriatal lesion induced by intranigral injection of the proteasome inhibitor lactacystin. RESULTS: Lactacystin induced parkinsonian-like behaviour, characterised by

Progressive supranuclear palsy syndrome presenting as progressive nonfluent aphasia: a neuropsychological and neuroimaging analysis.

There is currently considerable interest in the clinical spectrum of progressive nonfluent aphasia (PNFA) and progressive supranuclear palsy (PSP) and the intersection of these two entities. Here, we undertook a detailed prospective clinical, neuropsychological, and neuroimaging analysis of 14 consecutive patients presenting with PNFA to identify cases meeting clinical criteria for PSP. These patients had further detailed assessment of extrapyramidal and oculomotor functions. All patients had high-resolution MR brain volumetry and a cortical thickness analysis was undertaken on the brain image

Cabergoline decreases alcohol drinking and seeking behaviors via glial cell line-derived neurotrophic factor.

BACKGROUND: Cabergoline is an ergotamine derivative that increases the expression of glial cell line-derived neurotrophic factor (GDNF) in vitro. We recently showed that GDNF in the ventral tegmental area (VTA) reduces the motivation to consume alcohol. We therefore set out to determine whether cabergoline administration decreases alcohol-drinking and -seeking behaviors via GDNF. METHODS: Reverse transcription polymerase chain reaction (RT-PCR) and Enzyme-Linked ImmunoSorbent Assay (ELISA) were used to measure GDNF levels. Western blot analysis was used for phosphorylation experiments. Operan

Methamphetamine preconditioning alters midbrain transcriptional responses to methamphetamine-induced injury in the rat striatum.

Methamphetamine (METH) is an illicit drug which is neurotoxic to the mammalian brain. Numerous studies have revealed significant decreases in dopamine and serotonin levels in the brains of animals exposed to moderate-to-large METH doses given within short intervals of time. In contrast, repeated injections of small nontoxic doses of the drug followed by a challenge with toxic METH doses afford significant protection against monoamine depletion. The present study was undertaken to test the possibility that repeated injections of the drug might be accompanied by transcriptional changes involved

Infusions of 3alpha,5alpha-THP to the VTA enhance exploratory, anti-anxiety, social, and sexual behavior and increase levels of 3alpha,5alpha-THP in midbrain, hippocampus, diencephalon, and cortex of female rats.

17beta-Estradiol (E2) and progesterone (P4) influence the onset and duration of sexual behavior and are also associated with changes in behaviors that may contribute to mating, such as exploration, anxiety, and social behaviors (socio-sexual behaviors). In the midbrain ventral tegmental area (VTA), the P4 metabolite, 5alpha-pregnan-3alpha-ol-20-one (3alpha,5alpha-THP), modulates lordosis of E2-primed rodents; 3alpha,5alpha-THP can also influence anxiety and social behaviors. To examine if 3alpha,5alpha-THP in the VTA mediates socio-sexual behaviors, we infused 3alpha,5alpha-THP to the VTA of d

[Pseudo-tumoral neuro-Behçet's disease].

INTRODUCTION: Clinical involvement of the central nervous system occurs in about 10 to 30 percent of patients with Behçet's disease. Neurological pseudo tumoral presentation is rare. CASE REPORT: We report a case of pseudo tumoral neuro-Behçet disease in a 38-year-old man, with past history of facial and dorsal folliculitis, who presented suddenly headache, right hemiplegia, aphasia and disturbed consciousness. Neuroradiological investigations showed a pseudo tumoral lesion in the left capsulo-thalamic region extending to the homolateral peduncle. The patient improved with steroid and immunos

Apoptosis inducing factor mediates caspase-independent 1-methyl-4-phenylpyridinium toxicity in dopaminergic cells.

Parkinson's disease is a debilitating neurodegenerative disease characterized by loss of midbrain dopaminergic neurons. These neurons are particularly sensitive to the neurotoxin 1-methyl-4-phenylpyridinium (MPP+), the active metabolite of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP), which causes parkinsonian syndromes in humans, monkeys and rodents. Although apoptotic cell death has been implicated in MPTP/MPP+ toxicity, several recent studies have challenged the role of caspase-dependent apoptosis in dopaminergic neurons. Using the midbrain-derived MN9D dopaminergic cell line, we fou

Effects of olanzapine infusions to the ventral tegmental area on lordosis and midbrain 3alpha,5alpha-THP concentrations in rats.

RATIONALE: The progesterone metabolite and neurosteroid 5alpha-pregnane-3alpha-ol-20-one (3alpha,5alpha-THP) facilitates sexual behavior of estradiol-primed rodents through its actions in the ventral tegmental area (VTA). Olanzapine, an atypical antipsychotic, may exert some of its actions by increasing 3alpha,5alpha-THP levels. OBJECTIVE: If olanzapine has effects by increasing 3alpha,5alpha-THP levels, then olanzapine administration to the VTA should facilitate feminine sexual behavior of estradiol-primed rodents concomitant with increasing midbrain levels of 3alpha,5alpha-THP. METHODS. In

Neonatal ablation of the nigrostriatal dopamine pathway does not influence limb development in rats.

Hemiparkinson-hemiatrophy syndrome (HP-HA) is associated with skeletal hemiatrophy and the later development of parkinsonism. It is generally assumed that this phenotype is due to the combination of dysfunction of the basal ganglia (e.g., substantia nigra compacta and/or other related structures), causing parkinsonism, and of other areas (e.g., cerebral cortex), causing hemiatrophy. The occurrence of asymmetry of limb size in a patient with very asymmetric involvement of dopa-responsive dystonia encouraged Greene et al. (2000, Mov. Disord. 15: 537-541) to propose that lifelong deficits in nigr

CEPU-1 expression in the early embryonic chick brain.

We describe the expression pattern of CEPU-1, a cell adhesion molecule of the immunoglobulin superfamily, in the early chick embryo brain. An initially broad domain of expression, encompassing forebrain, midbrain and anterior hindbrain, is subsequently narrowed down to a ring-shaped domain at the midbrain-hindbrain boundary, co-localizing precisely with the expression of Wnt1 at the isthmus. In addition, CEPU-1 is expressed in the dorsal aspect of rhombomere 4 and its emigrating neural crest cells. Later in development, we also find CEPU-1 expression in other parts of the developing nervous sy

Modulation of MK-801-induced behaviour by noradrenergic agents in mice.

RATIONALE: Inhibition of glutamatergic N-methyl-D-aspartate (NMDA) receptors following the administration of NMDA receptor antagonists results in psychotic-like behaviour. Whereas it is known that pharmacological manipulation of dopaminergic and serotonergic pathways affect this drug-induced psychosis, a role for noradrenaline has not yet been clearly defined. OBJECTIVES: Thus, in the present study, we assessed a possible role for noradrenaline in the behavioural response to the non-competitive NMDA receptor anatgonist, MK-801, in male CD-I mice. RESULTS: MK-801 (0.02-1.28 mg/kg; ED50 0.2 mg

Metabolic energy capacity of dopaminergic grafts and the implanted striatum in parkinsonian nonhuman primates as visualized with cytochrome oxidase histochemistry.

Histochemistry for visualization of the mitochondrial enzyme cytochrome oxidase has been detect cellular and regional differences in brain metabolism. We have examined the pattern of cytochrome oxidase (CO) staining in grafts of embryonic ventral mesencephalic tissue, and in the implanted striatum, of MPTP-treated monkeys as one index of the functional activity of grafted tissue and its influence on the host brain. Four monkeys were selected for study based on interesting variations in dopamine (DA) neuron content of their bilateral grafts as demonstrated with tyrosine-hydroxylase (TH) immunoc
