Immune responses triggered by oral administration of recombinant Bacillus subtilis expressing the E2 protein of classical swine fever virus.

Classical swine fever virus (CSFV) causes an acute, febrile, and highly contagious disease of domesticated pigs and wild boars, resulting in substantial economic losses to the global swine industry. Conventional injectable vaccines fail to induce mucosal immunity and thus cannot block viral invasion via the digestive and respiratory tracts as the primary infection sites. In contrast, oral vaccines mimic the natural infection route and activate mucosal immune responses, thereby emerging as a novel direction for CSFV control. However, the degradation of vaccine antigens by the harsh gastrointest

Evaluation of immunogenicity and protective efficacy of orally delivered Shigella type III secretion system proteins IpaB and IpaD.

Shigella spp. are food- and water-borne pathogens that cause shigellosis, a severe diarrheal and dysenteric disease that is associated with a high morbidity and mortality in resource-poor countries. No licensed vaccine is available to prevent shigellosis. We have recently demonstrated that Shigella invasion plasmid antigens (Ipas), IpaB and IpaD, which are components of the bacterial type III secretion system (TTSS), can prevent infection in a mouse model of intranasal immunization and lethal pulmonary challenge. Because they are conserved across Shigella spp. and highly immunogenic, these pro

In silico analyses of heat shock protein 60 and calreticulin to designing a novel vaccine shifting immune response toward T helper 2 in atherosclerosis.

Recent experiments demonstrated that atherosclerosis is a Th1 dominant autoimmune condition, whereas Th2 cells are rarely detected within the atherosclerotic lesions. Several studies have indicated that Th2 type cytokines could be effective in the reduction and stabilization of atherosclerotic plaque. Therefore, the modulation of the adaptive immune response by shifting immune responses toward Th2 cells by a novel vaccine could represent a promising approach to prevent from progression and thromboembolic events in coronary artery disease. In the present study, an in silico approach was applied

In silico design and testing of a multi-epitope camel mastitis vaccine candidate for development.

INTRODUCTION: Camel mastitis caused by a diverse array of pathogenic bacteria including Staphylococcus aureus and Streptococcus agalactiae is responsible for enormous socioeconomic losses in the arid and semi-arid regions of Africa where camels are an invaluable source of livelihood and nutrition. Although S. aureus and S. agalactiae are environmental contaminants, the latter is an infectious zoonotic agent. The development of a safe, efficacious vaccine for camel mastitis is a compelling priority particularly against the backdrop of climate change and antimicrobial resistance. METHODS: We ha

Replication-Deficient Particles: New Insights into the Next Generation of Bluetongue Virus Vaccines.

UNLABELLED: Bluetongue virus (BTV) is endemic in many parts of the world, often causing severe hemorrhagic disease in livestock. To date, at least 27 different serotypes have been recognized. Vaccination against all serotypes is necessary to protect susceptible animals and to prevent onward spread of the virus by insect vectors. In our previous studies, we generated replication-deficient (disabled infectious single-cycle [DISC]) virus strains for a number of serotypes and reported preliminary data on their protective efficacy in animals. In this report, to advance the DISC vaccines to the mark

Development of a lymph node-homing peptide vaccine targeting C-C chemokine receptor 7-positive dendritic cells with enhanced antitumor immunity.

Targeting dendritic cells via surface receptors presents a promising strategy to enhance antigen delivery and antitumor immunity. C-C Chemokine Receptor 7 (CCR7), as a chemokine receptor, plays a crucial role in dendritic cells migration and homing to lymph node, making it an attractive target for the development of dendritic cell-targeted immunotherapies. In this study, using phage display technology and binding assay, a novel peptide, CRBP3, that specifically targeting CCR7 was identified. In vitro, CRBP3 efficiently bound to and was internalized by both immature and mature dendritic cells.

Bioinformatics-guided vaccine targeting the hemagglutinin protein of avian influenza virus.

Avian influenza viruses (AIV) represent a major zoonotic threat to global public health and agriculture due to their high mutation rates, antigenic drift, and potential for interspecies transmission. This study addresses the urgent need for innovative, broad-spectrum vaccines that can overcome the limitations of traditional approaches, such as slow production and strain-specific protection, by designing and evaluating a novel multi-epitope protein-based vaccine targeting conserved regions of the AIV hemagglutinin (HA) protein through advanced immunoinformatics tools. Using an in silico approac

Mannose-based VZV-gE protein self-adjuvanted hydrogel vaccine enhances immune response.

Herpes zoster (HZ) is an infectious disease caused by the varicella zoster virus. Vaccination, especially recombinant protein vaccine, is an effective preventive method. However, the protein vaccine is easy to degrade, and its immunogenicity is weak, combined adjuvants and delivery systems can improve protein vaccines immune efficacy. In this work, a self-adjuvanted hydrogel vaccine (OHH/gE) was prepared by oxidized mannan and hydroxypropyl chitosan, which could slowly release antigen to continuously stimulate the body to improve immune level and effectively prevent and treat Hz. The results s

A circular mRNA vaccine prototype producing VFLIP-X spike confers a broad neutralization of SARS-CoV-2 variants by mouse sera.

Next-generation COVID-19 vaccines are critical due to the ongoing evolution of SARS-CoV-2 virus and rapid waning duration of the neutralizing antibody response against current vaccines. The mRNA vaccines mRNA-1273 and BNT162b2 were developed using linear transcripts encoding the prefusion-stabilized trimers (S-2P) of the wildtype spike, which have shown a reduced neutralizing activity against the variants of concern B.1.617.2 and B.1.1.529. Recently, a new version of spike trimer, termed VFLIP (five (V) prolines, Flexibly-Linked, Inter-Protomer disulfide) was developed. Based on the original a

Ganglioside enhances the immunogenicity of nanoparticles displaying short synthetic tumor neoepitopes and epitopes.

RATIONALE: Short major histocompatibility class I epitopes can be coupled to the surface of immunogenic liposomes to elicit antigen-specific CD8+ T cells with vaccination. Here, we examined whether the co-incorporation of GM3 ganglioside, a lipid which targets CD169, a sialic acid-binding receptor, would further improve the immunogenicity of this approach. METHODS: Liposomes were formed with GM3 incorporated and were assessed for CD169 targeting and anti-tumor immunogenicity in murine models. Antigen-specific CD8+ T cell populations and characteristics were investigated to examine the effect

Dendritic Lipopeptide Nanovaccines Orchestrate Multi-Pattern Recognition Receptors Activation and Potentiate Antitumor Immunity.

Precise control over the spatial and temporal delivery of antigens and adjuvants is essential for eliciting effective and durable immune responses. Here, we report a self-assembling nanovaccine platform based on dendritic lipopeptides (DLPs) that integrate delivery and immune-stimulation through a structurally defined design. DLPs are constructed by conjugating structurally optimized lipid tails to second-generation lysine- or arginine-rich dendritic scaffolds. Systematic engineering of the lipid domain allows for precise fine-tuning of the amphiphilic structure, yielding molecular candidates

Immunotherapy Treatment of Colorectal Cancer and Lung Metastasis with Genetically Modified Outer Membrane Vesicles Delivering Poly-Neoepitopes in a Mouse Model.

Targeting multiple neoepitopes by vaccines have the potential to circumvent intratumor clonal heterogeneities and minimize the chance of tumor escape. Due to immunostimulatory properties of outer membrane vesicles, their ability to target lymph nodes, as well as unaltered conformation of the loaded antigens in recombinant OMVs (rOMVs), they offer as promising vaccine platforms. Herein, we aimed to evaluate the therapeutic and antimetastatic potential of ClearColiTM-derived rOMVCT−26 polytope encoding CT-26 polytope and compare the induced immunity by the CT-26 polytope as a recombinant protein

Reverse vaccinology-based design of a universal multiepitope vaccine against chikungunya virus: Phylogenetic and immunoinformatics approaches.

Chikungunya virus (CHIKV) infection is a re-emerging arboviral disease in tropical and subtropical regions. In addition to acute febrile syndrome, CHIKV infection may lead to chronic articular manifestations that significantly affect a long-term quality of life. This study aimed to design a universal vaccine candidate covering all circulating genotypes of CHIKV based on conserved multiepitope platform. We employed a large scale phylogenetic and immunoinformatic approach to identify conserved regions of the open reading frames (ORF2) region encoding viral structural proteins. This study ultimat

Preclinical Evaluation of a Phage T4-Based Multi-Epitope Nanoparticle Vaccine Against Porcine Epidemic Diarrhea Virus.

Porcine epidemic diarrhea virus (PEDV) continues to threaten the global swine industry, and currently available vaccines often provide incomplete protection against emerging virulent strains. In this study, we developed a phage T4-based multi-epitope nanoparticle vaccine targeting PEDV. B and T cell epitopes derived from the PEDV spike (S) protein were identified through immunoinformatic analyses and assembled into a tandem epitope construct (Pep) using flexible linkers. In silico analyses predicted favorable antigenic properties of Pep and a stable interaction with Toll-like receptor 3 (TLR3)

Fucosyltransferase 4-derived peptide bioconjugates on carbon nanotubes enhance antitumor immunity in an ovarian cancer mouse model.

Ovarian cancer is characterized by a profoundly immunosuppressive tumor microenvironment, which has limited the clinical success of immunotherapy approaches. Antigen-based nanovaccines represent a promising strategy to enhance antitumor immunity by improving antigen delivery and immune activation. In this study, we evaluated carbon nanotubes (CNTs) bioconjugated with a peptide derived from tumor-associated antigen fucosyltransferase 4 (FUT4) as an immunomodulating platform in an immunocompetent ovarian cancer mouse model. FUT4 expression was confirmed in the ID8-Defb29/Vegf-a (ID8DVLuc) ovaria
