Onchocerca volvulus recombinant antigen: physical characterization and clinical correlates with serum reactivity.

An Onchocerca volvulus cDNA clone expressing epitopes found in adult and larval parasites, designated lambda RAL-2, was derived from a 1,000-base message present in adult O. volvulus, which encodes a protein with an apparent molecular weight of 17,000. This protein does not appear to be extensively posttranslationally modified. Serum samples from 52 individuals exposed to O. volvulus were examined for antibodies recognizing the lambda RAL-2 recombinant antigen; 77% produced such antibodies. In addition, individuals producing antibodies recognizing the recombinant antigen were significantly les

Sequence analysis of the wall-associated protein precursor of Streptococcus mutans antigen A.

The nucleotide sequence has been determined for the Streptococcus mutans wall-associated protein A (wapA) gene from serotype c strains Ingbritt and GS5. The nucleotide sequence for each wapA gene was virtually identical, although the gene from strain GS5 contained a 24 base pair deletion. A 29 amino acid signal peptide was specified by each wapA gene with a mature protein of 424 amino acids (Mr, 45,276) for strain Ingbritt and 416 amino acids (Mr, 44,846) for strain GS5. In the C-terminal region of the wall-associated protein A, considerable sequence similarity was found with the membrane anch

An ochre mutation in the vitamin D receptor gene causes hereditary 1,25-dihydroxyvitamin D3-resistant rickets in three families.

Hereditary 1,25-dihydroxyvitamin D3-resistant rickets is a rare autosomal-recessive disease resulting from target-organ resistance to the action of the active hormonal form of vitamin D. Four affected children from three related families with the classical syndrome of hereditary 1,25-dihydroxyvitamin D3-resistant rickets and the absence of detectable binding to the vitamin D receptor (VDR) in cultured fibroblasts or lymphoblasts were examined for genetic abnormalities in the VDR gene. Genomic DNA from Epstein-Barr virus-transformed lymphoblasts of eight family members was isolated and amplifie

Dissemination of trimethoprim-resistant clones of Shigella sonnei in Bulgaria.

Bacterial dysentery due to Shigella sonnei remains a serious public health problem in developed countries, including Bulgaria. At the National Shigella Reference Laboratory in Sofia, 17,126 strains of S. sonnei from epidemics and sporadic cases collected from 1973 to 1987 were studied. Antibiotic susceptibility testing, phage typing, colicin typing, and biotyping were performed for all strains to allow intraspecies differentiation and to track any clonal distribution. Of all strains, 84.3% were resistant to one or more antimicrobials, the most frequent being tetracycline (Tet), streptomycin (S

Specific serodiagnosis of visceral leishmaniasis using a Leishmania donovani antigen identified by expression cloning.

A lambda gt11 expression library was constructed using mRNA from the promastigote form of Leishmania donovani (African isolate MHOM/ET/67/HU3). The library was screened with serum obtained from a patient who contracted visceral leishmaniasis in the Sudan. Several cDNA clones which expressed beta-galactosidase/L. donovani antigen fusion proteins were isolated. One of these clones corresponded to a 60 kDa membrane-associated antigen. By a Western blot assay antibodies against the fusion protein were detected universally in the sera of visceral leishmaniasis patients. They were not detected in se

Molecular analyses of conjugative, gentamicin-resistance plasmids from staphylococcal clinical isolates.

Gentamicin-resistant Staphylococcus aureus and Staphylococcus epidermidis strains which were isolated from infants with staphylococcal bacteremia were analyzed for the presence of self-transmissible gentamicin-resistance (Gmr) plasmids. Conjugative GMr plasmids of approximately 43.8-63 kilobases (kb) were found in all S. aureus strains. Inter- and intra-species transfer of Gmr plasmids by conjugation was observed from S. aureus to S. aureus and to S. epidermidis recipient strains. However, neither inter- nor intra-species transfer of gentamicin resistance by conjugation was observed with nine

The fragile-X syndrome, IV. Progress towards the identification of linked restriction fragment length variants (RFLVs).

We present our strategy and progress towards the identification of rare restriction fragment length variants (RFLVs) segregating with the fragile-X syndrome. DNA from a carrier mother and two retarded sons was digested with 7 restriction endonucleases and Southern blots were probed with cloned unique X-chromosomal sequences. Two of 17 cloned segments tested revealed RFLVs between the X-chromosomes of the carrier mother. One of them detected variants using Pvu II and Msp I. The Pvu II and Msp I alleles found on the X-chromosome bearing the fragile-X mutation were not found in 31 and 22 random X

Multi-agent game theory for precision psychiatry.

Psychiatry has learned to measure and map the brain in exquisite detail, yet it still lacks a theory of what happens between minds. Most current frameworks treat cognition as an individual process, studied through static and non-interactive tasks, creating a fundamental mismatch with real-world social behavior, which is dynamic and reciprocal. In restricted psychiatric conditions such as autism spectrum disorder (ASD) and schizophrenia with social phenotypes, core symptoms emerge most clearly during interaction. Here I argue that these differences are better understood as failures of coordinat

Advances in genetically engineered and humanized mouse models for dissecting sepsis mechanisms and therapeutic response.

Sepsis remains a major cause of global mortality, yet translation from preclinical studies to effective targeted therapies has been limited. A key barrier is that conventional animal models incompletely capture human immune circuitry and patient heterogeneity. Recent progress in mouse genetics, CRISPR/Cas-based genome editing, conditional alleles (Cre-loxP), and next-generation humanized platforms has expanded the toolkit for functional interrogation of host pathways that shape sepsis outcomes. Germline and cell-type-restricted perturbations have helped map roles for pattern-recognition signal

Mechanistic insights and methodological challenges in the study of glycosylated RNA: A review.

Glycosylated RNA (GlycoRNA) constitutes a novel category of cell-surface nucleic acids characterized by intricate glycan modifications, thereby extending the scope of glycosylation beyond proteins and lipids and challenging the traditional notion that RNA is restricted to intracellular compartments. This review encapsulates recent advances in the field of GlycoRNA biology, including biochemical findings that 3-(3-amino-3-carboxypropyl) uridine to serve as an anchor covalent RNA-N-glycan conjugates. It also highlights advancements in methodologies such as metabolic labeling, spatial imaging, an

Modeling spasticity: a systematic review.

Spasticity, a type of hypertonia characterized by a velocity-dependent increase in muscle tone, is associated with damage to the brain and/or spinal cord in different neurological conditions. However, secondary non-neurological factors, such as soft tissue changes, can complicate the assessment and differentiation of the underlying causes. Accurate assessment is crucial for effective treatment planning, with clinicians relying on passive movement to grade the 'feel' of the spastic limb. This review aims to identify and evaluate the feasibility of spasticity models or simulations for clinical t

[Potential of imaging in the clinical routine : Focus on cardiovascular MRI].

Cardiac magnetic resonance imaging (CMR) provides decisive advantages, particularly in coronary heart disease, myocarditis and cardiomyopathy. It accurately detects ischemia, scarring, edema and microvascular disorders, enables reliable risk stratification and supports treatment decisions such as revascularization or medication adjustments. Modern quantitative perfusion methods and artificial intelligence (AI)-based analyses further increase the diagnostic accuracy. In inflammatory myocardial and pericardial diseases, CMR using mapping techniques and late gadolinium enhancement (LGE) forms the

CRISPR-screen informed engineered T cell therapies.

Adoptive T cell therapies can deliver curative responses for refractory patients with B cell malignancies, yet clinical activity in solid tumors remains inconsistent. Tumor-intrinsic barriers dominating this inconsistency include the immunosuppressive solid tumor microenvironment (TME) imposing chronic inhibitory cues to T cells and the scarcity of patient-shared and uniformly expressed tumor-restricted antigens for T cells to target. CRISPR-based forward genetics screens enable mapping of the functional genome regulating T cell anti-tumor activity. Here, we review recent insights from pooled

Beyond Sparsity: Receptive Field Expansion and Cross-Task Fusion for LiDAR Multi-Task Perception.

LiDAR-based multi-task perception for autonomous driving requires efficient integration of spatial context and cross-task information, yet existing methods often suffer from restricted receptive fields and suboptimal task interaction. This paper presents a novel multi-task framework that goes beyond sparsity constraints, leveraging receptive field expansion and cross-task fusion to enhance 3D object detection and semantic segmentation. We introduce the Spatial Density-Invariant Multi-scale Integrator (SDIMI), which adaptively fuses multi-resolution contextual features using density-agnostic st

Endemic Mycoses in 2026: can we keep the pace of redrawing maps?

PURPOSE OF REVIEW: The dimorphic fungal infections were classically grouped as endemic mycoses due to their restricted geographic distribution. However, their original geographic distribution dates from the twentieth century and true prevalence and distribution of these infections is limited by subclinical presentations, lack of healthcare provider awareness, lack of diagnostic capacity, and incomplete reporting. In this review, we discuss the new geographic distribution of endemic mycoses and the factors influencing its expansion. RECENT FINDINGS: There has been an increase in incidence and

A‑iMPO: A Design-Time Prediction Toolbox for Blood Brain Barrier Permeation Using an Integrated and Interpretable Multiparameter Optimization Framework.

Design time prioritization of central nervous system (CNS) drug candidates remains a challenge due to the restrictive nature of the blood-brain barrier (BBB) governing brain exposure. Although various multiparameter optimization (MPO) strategies have guided CNS medicinal chemistry for over a decade, existing frameworks rely heavily on heuristic cutoffs and offer limited interpretability across chemically diverse scaffolds. Here, we introduce a next generation CNS-MPO frameworkAragen-iMPO, (A-iMPO)which was developed using 5,129 curated compounds through an integrated workflow combining expla

Deep peptide recognition profiling decodes TCR specificity and enables disease-associated antigen discovery.

Predicting T cell receptor (TCR) specificity on the basis of sequence is challenging because TCRs of similar sequence can recognize entirely different antigens, whereas TCRs of different sequence can recognize the same antigens. Here we present a system that integrates high-throughput yeast display with fine-tuned protein language models (pLMs) to generate deep peptide recognition profiles (PRPs) for individual TCRs, each detailing binding against millions of peptides. We provide detailed PRPs for a panel of HLA-B*27:05-restricted TCRs from persons with ankylosing spondylitis and acute anterio

Barriers to addiction-related care during pregnancy in Nigeria: a scoping review and health system policy analysis.

Substance-related exposures in pregnancy are documented across Nigeria, but their integration into antenatal care remains unclear. This scoping review and health-system policy analysis mapped empirical evidence on these exposures among pregnant women and examined barriers to perinatal addiction-related care. Addiction-related care is understood here as screening, risk assessment, counselling, documentation, referral, and treatment. Five databases were searched on 6 April 2026; 23 empirical studies were synthesised with national policy documents and clinical guidelines using the WHO health-syst

An inflammasome-driven differentiation program in intestinal stem cells protects against Salmonella infection.

Intestinal stem cells (ISCs) are essential for sustaining epithelial renewal and barrier integrity, yet their role in orchestrating defense against enteric pathogens remains unclear. Here we identify a stem cell-intrinsic immune mechanism whereby Lgr5+ ISCs detect intracellular Salmonella enterica and activate an inflammasome-dependent differentiation program. Using fluorescent-labeled S. enterica, single-cell transcriptomics, fate mapping, organoid models, and genetic perturbations, we show that invaded ISCs undergo rapid reprogramming toward antimicrobial peptide-enriched Paneth cells via ap

Coronavirus protein interaction mapping in bat and human cells identifies molecular and genetic switches for immune evasion and replication.

Coronaviruses, including SARS-CoV-2, can cause severe disease in humans, whereas reservoir hosts like Rhinolophus bats remain asymptomatic. To investigate how host-specific protein-protein interactions (PPIs) influence infection, we generated comparative PPI maps for SARS-CoV-2 and its bat-origin relative RaTG13 using affinity purification-mass spectrometry (AP-MS) in human and Rhinolophus ferrumequinum (RFe) bat cells. This approach identified both conserved and virus- and host-specific interactions that regulate infection dynamics. Notably, SARS-CoV-2 required a non-synonymous mutation in nu
