Rickettsial infections of the central nervous system.

As a result of migrations and globalization, people may face a possible increase in the incidence of central nervous system rickettsial infections (CNS R). These diseases, caused by Rickettsia species and transmitted to humans by arthropod bites, are putatively lethal. However, the diagnosis of CNS R is challenging and often delayed due to their nonspecific clinical presentation and the strict intracellular nature of rickettsiae. Furthermore, transfer of rickettsiae to the brain parenchyma is not yet understood. The aim of this review is to analyze and summarize the features and correlated fin

High levels of antisense transcription from numerous genes in the obligate intracellular bacterium Orientia tsutsugamushi.

Orientia tsutsugamushi (Ot) is a cytoplasmic, gram-negative, obligate intracellular bacterium that causes the human disease scrub typhus. It has a highly repetitive genome, whereby approximately 50% is composed of multiple copies of a proliferated integrative and conjugative element, the Rickettsial amplified Genetic Element (RAGE). Previous RNA sequencing analysis revealed a high level of antisense transcription in Ot, particularly from RAGE-encoded genes, with 18% of all genes having a sense:antisense ratio <1 and 5% with a ratio <0.1. In our current study, we have confirmed the earlier RNA …

Development of One Health-based community surveillance system to study zoonotic diseases: a study protocol.

Animals are responsible for a substantial burden of infectious diseases and pose a serious threat to human health, particularly in peri-urban areas where human-animal-environment interfaces are intensified due to rapid urban expansion. This protocol aims to develop a One Health-based community surveillance system to assess the prevalence, transmission dynamics and risk factors associated with brucellosis, leptospirosis and scrub typhus in a peri-urban setting in Odisha, India. We will conduct a prospective mixed-methods cohort study over 3 years in two peri-urban blocks of Khordha district. Ab…

The intracellular bacterium Orientia tsutsugamushi uses the autotransporter ScaC to activate BICD adaptors for dynein-based motility.

The intracellular bacterium Orientia tsutsugamushi relies on the microtubule cytoskeleton and the motor protein dynein to traffic to the perinuclear region within infected cells. However, it remains unclear how the bacterium is coupled to the dynein machinery and how transport is regulated. Here, we discover that O. tsutsugamushi uses its autotransporter protein ScaC to recruit the dynein adaptors BICD1 and BICD2 for movement to the perinucleus. We show that ScaC is sufficient to engage dynein-based motility in the absence of other bacterial proteins and that BICD1 and BICD2 are required for e

[Role of Leptotrombidium (L.) scutellare in transmission of human diseases].

A series of cases of hemorrhagic fever with renal syndrome (HFRS) in Shaanxi province and autumn-type tsutsugamushi disease in Jiangsu province were studied during 1988 to 1990 and 1986 to 1993 to explore the role of Leptotrombidium (L.) scutellare in transmission of these diseases. Results showed that L. (L.) scutellare is a dominant strain of mites inhabiting in rats and its seasonal fluctuation conformed to the seasonal distribution of these diseases. L. (L.) scutellare could naturally infect with HFRS virus and Rickettsia tsutsugamushi and transmit them by stinging and transovarial route.

Predicting the potential distribution areas of Leptotrombidium rubellum under current and future climate change.

BACKGROUND: Leptotrombidium rubellum (L. rubellum), a confirmed vector of scrub typhus, was historically restricted to southeastern coastal China but has recently been detected in southwestern regions. Species distribution modeling was applied to predict its current and future potential distribution areas under multiple climate scenarios, identify high-priority surveillance areas, and determine key environmental drivers. The results may facilitate a transition from passive to proactive vector monitoring. METHODS: Fifty-seven potential influencing factors were evaluated. The maximum entropy (M…

Inhibition of eukaryotic translation by tetratricopeptide-repeat proteins of Orientia tsutsugamushi.

Orientia tsutsugamushi, an obligate intracellular bacterium, is the causative agent of scrub typhus. The genome of Orientia tsutsugamushi has revealed multiple ORFs encoding tetratricopeptide-repeat (TPR) proteins. The TPR protein family has been shown to be involved in a diverse spectrum of cellular functions such as cell cycle control, transcription, protein transport, and protein folding, especially in eukaryotic cells. However, little is known about the function of the TPR proteins in O. tsutsugamushi. To investigate the potential role of TPR proteins in host-pathogen interaction, two orie…
