Alzheimer's disease (AD) remains the leading cause of dementia worldwide, imposing an enormous and growing societal burden with more than 55 million people affected globally. Despite decades of intensive investigation, existing therapeutic options provide only modest symptomatic relief and fail to prevent or slow disease progression, emphasizing the critical need for interventions that target the fundamental molecular mechanisms of neurodegeneration. Pathologically, Alzheimer's disease is characterized by extracellular accumulation of amyloid-β plaques, intracellular neurofibrillary tangles fo

Dengue is an arboviral disease of global significance caused by Orthoflavivirus denguei (DENV), which has four antigenically distinct serotypes. The envelope (E) protein plays a critical role in viral entry and eliciting immune responses. This study aimed to compare the biochemical and immunological properties of the E protein across the four DENV serotypes using in silico approaches. E protein reference sequences were retrieved from RefSeq and analyzed with various bioinformatics tools. Sequence alignment revealed identities ranging from 63.08% to 77.69%. Biochemical analysis showed minimal v

Human papillomaviruses (HPVs), particularly types 16 and 18, are major contributors to cervical cancer through the oncogenic activities of the E6 and E7 proteins. These viral proteins inactivate the tumour suppressors p53 and pRB, driving uncontrolled cellular proliferation. In this study, we investigated the interaction between the HPV E7 protein and the R2TP complex, a co-chaperone involved in essential cellular functions, including ribosome biogenesis, transcription, and macromolecular assembly. We identified PIH1D1, a core R2TP subunit, as an interacting partner of HPV16 and HPV18 E7 prote

Eosinophilic esophagitis (EoE) has transitioned from an isolated gastrointestinal disorder to a recognized type 2 immune-mediated allergic disease, most likely representing a late manifestation of the atopic march. This comprehensive review examines the complex inflammatory pathogenesis linking EoE and food allergy, and critically discusses the mechanisms of disease induction, dietary treatments, and emerging clinical challenges. While genome-wide association studies identify shared susceptibility loci with classic atopy, EoE exhibits distinct tissue-specific pathways, particularly dominated b

Excessive lipid absorption is a key factor in obesity. Lipids are solubilized in the gut via bile acid (BA) micelles, where pancreatic lipase hydrolyzes them for absorption. This study aimed to enrich pancreatic lipase inhibitory (PLI) peptides from food protein hydrolysates and clarify their inhibition mechanisms. We used heat-treated porous silica gel (HTSG) to selectively enrich basic and hydrophobic peptides through adsorption-desorption. While HTSG has previously enriched PLI peptides, the mechanism remained unclear. Since basic and hydrophobic peptides can bind strongly to BAs like tauro

Aortic valve stenosis (AVS) is a growing healthcare burden. Aortic valve replacement (AVR) remains the only effective treatment to eliminate pressure overload and triggers myocardial reverse remodelling (RR), with regression of hypertrophy, fibrosis and diastolic function normalisation. However, many patients show an incomplete RR, being at higher risk of death. We aimed to uncover pathways and new therapeutic targets for incomplete RR through myocardial (phospho)proteomics. AVS patients were categorised based on left ventricle mass regression (LVM): complete RR (≥ 15%) or incomplete RR (≤ 5%)

Hetero-trans-β-glucanase (HTG) is an Equisetum plant enzyme that covalently grafts cellulose (donor substrate) to xyloglucan heptasaccharide (XXXG; Xyl3.Glc4 as acceptor substrate). Commercial exploitation of this technology requires bulk XXXG production. Novel acceptor substrates bearing two or more XXXG moieties could potentially cross-link cellulose chains. We explored bulk XXXG production, the synthesis of novel (including multimeric) XXXG conjugates and novel coloured derivatives, and explored new activators of HTG action. Commercial tamarind gum (crude xyloglucan) was digested with xylog

The diagnosis of IgE-mediated cow's milk allergy (CMA) involves serum-specific IgE (spIgE), skin prick tests (SPT), and the gold standard oral food challenge (OFC). SpIgE levels are measured using methods such as ImmunoCAP and IMMULITE, with most cutoff data derived from ImmunoCAP. This study aims to determine IMMULITE-specific cutoff values to predict OFC positivity. Patients diagnosed with CMA via OFC between 2019 and 2023 were retrospectively analyzed. Data on demographics, eosinophil counts, total and specific IgE levels, and SPT results were collected. OFC was conducted using yogurt, milk

BACKGROUND: Bone nonunion is a major complication in fracture treatment. Despite considerable advancements in fracture care, 5-10% of fractures result in nonunion; This demonstrates the requirement for novel molecules to enhance fracture healing. Brain-derived neurotrophic factor (BDNF) has been increasingly recognized for its role in fracture healing; however, the underlying molecular mechanisms remain unclear. Osteoblast proliferation and migration are critical to this process. The nuclear casein kinase and cyclin-dependent kinase substrate 1 gene (NUCKS1), located on human chromosome 1 at 1

In 1974, Van den Berghe et al described a distinct hematologic disorder associated with acquired, interstitial deletion of part of the long arm of chromosome 5. This condition is now classified as myelodysplastic syndrome (MDS) with isolated deletion 5q, or MDS-del(5q). The common deletion region 5q32-5q33 contains several genes and microRNAs whose expression levels are reduced in hematopoietic cells, consistent with the loss of 1 allele. Haploinsufficiency production of multiple gene transcripts, primarily involving CSNK1A1, RPS14, MIR145, and MIR146A, results in myelodysplastic hematopoiesis

BACKGROUND: Atherosclerosis is a pathophysiological process common to a range of cardiovascular diseases. We reasoned that considering clinical presentations of atherosclerosis across the coronary, peripheral, and cerebrovasculature as a single entity would enhance statistical power to identify rare genetic variation driving pathological processes across multiple vascular beds.
METHODS: We performed an exome-wide association study of atherosclerotic cardiovascular disease in 434 438 UK Biobank participants of European ancestry.
RESULTS: We identified rare, predicted damaging variants in HTRA

BACKGROUND: Congenital vascular malformations in the oral and maxillofacial region present significant diagnostic and therapeutic challenges, particularly in pediatric patients. These lesions-most commonly low-flow venous malformations-can compromise oral function, aesthetics, and routine dental care due to their vascular nature and bleeding risk.
CASE PRESENTATION: We report the case of a 3.5-year-old girl with a congenital vascular malformation involving the left cheek, upper lip, vestibule, hard palate, soft palate, and anterior maxilla. The lesion was present at birth and gradually enlarg

Okur-Chung neurodevelopmental syndrome (OCNDS) is an ultra-rare disorder caused by variants in the CSNK2A1 gene. CSNK2A1 encodes for the alpha subunit of casein kinase 2 (CK2), a serine/threonine kinase critical in neural development. CK2 is implicated in many human pathologies, including viral infections, cancer, inflammation, cardiovascular, neurodegenerative, and psychiatric diseases. However, the mechanism of action for the CSNK2A1 variants observed in OCNDS is not fully understood, although studies suggest a loss of function or altered substrate specificity. There are no approved treatmen

Sleep deprivation (SD) has emerged as a critical concern impacting human health, leading to significant damage to the cardiovascular system. However, the underlying mechanisms are still unclear, and the development of targeted drugs is lagging. Here, we used mice to explore the effects of prolonged SD on cardiac structure and function. Echocardiography analysis revealed that cardiac function was significantly decreased in mice after five weeks of SD. Real-time quantitative PCR (RT-q-PCR) and Masson staining analysis showed that cardiac remodeling marker gene Anp (atrial natriuretic peptide) an

Gastric cancer (GC) is a frequently occurring malignancy with poor prognosis. Casein kinase 2 interacting protein-1 (CKIP-1) is a PH domain-containing protein implicated in regulating tumorigenesis and macrophage homeostasis. This study aimed to elucidate the role and potential mechanism of CKIP-1 in the progression of GC. CKIP-1 expression in GC tumor and para-carcinoma tissues was detected using RT-qPCR. Then, human monocyte cell line THP-1 was treated with PMA, interleukin (IL)-4 and IL-13 to induce M2-polarized macrophages. CD206, arginase-1 (Arg-1) and transforming growth factorβ1 (TGFβ1)

BACKGROUND & AIMS: Sleep disorders (SDs) are common in chronic liver diseases (CLDs). Some SDs arise from impaired internal clock and are, hence, circadian rhythm SDs (CRSDs). Bile acids (BAs), whose levels are increased in many CLDs, reciprocally interact with circadian rhythm. This study explores the mechanisms underlying CRSDs in CLDs and novel therapies.
METHODS: We monitored the sleep of patients with CLD using actigraphic watch and established male mouse cholemia models by feeding with BA or bile duct ligation. Sleep-wake cycle and circadian rhythm were analyzed by electroencephalogram-

OBJECTIVE: Although nearly half of preterm survivors display persistent neurobehavioral dysfunction including memory impairment without overt gray matter injury, the underlying mechanisms of neuronal or glial dysfunction, and their relationship to commonly observed cerebral white matter injury are unclear. We developed a mouse model to test the hypothesis that mild hypoxia during preterm equivalence is sufficient to persistently disrupt hippocampal neuronal maturation related to adult cellular mechanisms of learning and memory. Methods: Neonatal (P2) mice were exposed to mild hypoxia (8%O 2 )

In recent decades, scientific methodologies applied in theCultural Heritage field have been growing, due to their pivotal role in guiding informed decisions concerning conservation strategies and daily maintenance. To achieve this goal, minimally/non-invasive quantitative and qualitative analyses are needed. However, the non-invasive and selective identification of proteinaceous binders and coatings in artworks represent an open issue in Cultural Heritage science. Herein, a novel miniaturized system is introduced, which consists of a label-free electrochemical immunosensor integrated with bioc

Snake venom metalloproteases (SVMPs) are hydrolytic enzymes dependent on metal binding, primarily zinc (Zn2+), at their catalytic site. They are classified into three classes (P-I to P-III). BjussuMP-II, a P-I SVMP isolated from Bothrops jararacussu snake venom, has a molecular mass of 24 kDa. It exhibits inhibitory activity on platelet aggregation and hydrolyzes fibrinogen. TNF-α upregulates the expression of adhesion molecules on endothelial cell surfaces, promoting leukocyte adhesion and migration during inflammation. Literature indicates that SVMPs may cleave the TNF-α precursor, possibly

BACKGROUND: IgE-mediated cow's milk allergy (IgE-CMA) is one of the first allergies to arise in early childhood and may result from exposure to various milk allergens, of which β-lactoglobulin (BLG) and casein are the most important. Understanding the underlying mechanisms behind IgE-CMA is imperative for the discovery of novel biomarkers and the design of innovative treatment and prevention strategies.
METHODS: We report a longitudinal in vivo murine model, in which two mice strains (BALB/c and C57Bl/6) were sensitized to BLG using either cholera toxin or an oil emulsion (n = 6 per group). A
