Micro/nanoplastics (MNPs), detected in human tissues including the placenta, raise significant concerns regarding their potential impact on early human development. However, the mechanisms underlying their developmental toxicity remain poorly understood. To address this, we applied an Integrated Approaches to Testing and Assessment (IATA) framework to evaluate the developmental toxicity of polystyrene (PS) MNPs by combining adverse outcome pathway (AOP) development, experimental testing within an Integrated Testing Strategy (ITS), and literature-based data integration. As part of the ITS, expe

Organoids have emerged as innovative three-dimensional (3D) in vitro models capable of reproducing the essential structural and functional characteristics of human organs. They offer an alternative between traditional two dimensions (2D) cell cultures and animal models. Derived from stem cells, organoids have an intrinsic capacity for self-organisation and morphogenesis, recapitulating the processes of embryonic development. Three elements are crucial for their generation: the origin of the stem cells, the extracellular matrix, and controlled exposure to morphogens. Among the most promising ap

The efficient establishment of in vitro neural models that accurately mimic the structural and functional connectivity of neural networks is critical in neuroscience research. 3D bioprinting shows great potential for constructing sophisticated in vitro models with high freedom of design. However, mature neurons are delicate and susceptible to manipulation. Here, extrusion-based 3D bioprinting is employed to fabricate gelatin methacryloyl (GelMA)-based constructs containing embryonic day 18 (E18) rat cortical neurons, referred to as 3D neuMatrix. 3D neuMatrix displays favorable neuronal viabili

Memristor has shown significant application prospects in the post Moore's electron era, but its application in the field of reproductive medicine is still blank. In this work, a memristor with Ag/C40H56-PEDOT:PSS/FTO structure was prepared by using a heterojunction of natural biomaterial carotene (C40H56) and Poly(3,4-ethylenedioxythiophene):poly(styrenesulfonate) (PEDOT:PSS) as the functional layer of the device, which demonstrates remarkable resistance switching capabilities under a certain bias voltage. Subsequently, this memristor was employed to monitor the follicular fluid within the ova

The aim of this work was a detailed radiological and histopathological evaluation of a solid tumour that was diagnosed in a 2-year-old goose (Anser domesticus). The radiograph examination showed an osseous change involving the cervical vertebrae. The tumour measuring 15 cm × 10 cm × 9 cm was dense and had well-defined borders, suggesting the presence of calcified bone tissue. Histopathology revealed a well-defined benign neoplasm derived from bone that consisted largely of irregular, disorganized bone trabeculae surrounded by a layer of osteoblasts. The tumour has been classified as an osteoma

Insect reproductive capacity is a key determinant of population fitness, though its regulatory mechanisms remain limited. Here, we identified an op50 silkworm mutant (derived from p50 strain) exhibiting female-specific reproductive defects, which provided an important model for studying insect reproductive regulation mechanism. We found that Bombyx mori xanthine dehydrogenase 1 (BmXDH1) in op50 contained a single-base insertion causing premature termination, deleting 154 C-terminal amino acids and losing catalytic activity. ΔBmXDH1 mutants recapitulated the op50 phenotype with 50 % reduced ovi

Enterovirus 71 (EV71), a major causative agent of hand, foot, and mouth disease (HFMD), is often associated with severe clinical outcomes. In this study, three rare cyano-containing alkaloids, cyanodaphcalycines A-C (1-3), featuring an unusual 5/6/6/7/5/5-hexacyclic scaffold, and an undescribed daphmanidin A-type alkaloid, daphmanidin G (4), were isolated from the leaves and stems of the medicinal plant Daphniphyllum calycinum Benth., along with three known Daphniphyllum alkaloids (5-7). The previously undescribed structures and their absolute configurations were determined using a combination

BACKGROUND: Rhabdomyosarcoma (RMS) is the most prevalent soft tissue sarcoma in children, with approximately 30% of head and neck RMS occurring in the orbit. The management of orbital RMS is complex, requiring a multidisciplinary approach and careful surgical planning. The objective of the present paper is to provide the neurosurgeon with an update on this challenging tumor.
CASE DESCRIPTION: A 12-year-old boy was admitted to our department after the onset of diplopia in his right eye, associated with quickly worsening ipsilateral eyelid edema and mild proptosis. MRI revealed an anterior supe

Alterations to cilia are responsible for a wide range of severe disease; however, understanding of the transcriptional control of ciliogenesis remains incomplete. In this study we investigated whether altered cilia-mediated signaling contributes to the pleiotropic phenotypes caused by the Forkhead transcription factor FOXC1. Here, we show that patients with FOXC1-attributable Axenfeld-Rieger Syndrome (ARS) have a prevalence of ciliopathy-associated phenotypes comparable to syndromic ciliopathies. We demonstrate that altering the level of Foxc1 protein, via shRNA mediated inhibition, CRISPR/Cas

Despite the strong anticancer activity of SN38 (7-ethyl-10-hydroxy-camptothecin), the severe side effects and loss of anticancer activity caused by the lack of selectivity to cancer cells and hydrolysis of ring E prevent its clinical application. To address the issue, herein a multifunctional SN38 derivative (compound 9) containing biotin (tumor-targeting group) and valproic acid (histone deacetylase inhibitor, HDACi) was synthesized via click chemistry and evaluated using MTT assay. The in vitro cytotoxicity study showed that compound 9 exhibited superior cytotoxicity than irinotecan against

The developing brain has a well-organized anatomical structure comprising different types of neural and non-neural cells. Stem cells, progenitors and newborn neurons tightly interact with their neighbouring cells and tissue microenvironment, and this intricate interplay ultimately shapes the output of neurogenesis. Given the relevance of spatial cues during brain development, we acknowledge the necessity for a spatial transcriptomics map accessible to the neurodevelopmental community. To fulfil this need, we generated spatially resolved RNA sequencing (RNAseq) data from embryonic day 13.5 mous

Sonic hedgehog signaling regulates processes of embryonic development across multiple tissues, yet factors regulating context-specific Shh signaling remain poorly understood. Exome sequencing of families with polymicrogyria (disordered cortical folding) revealed multiple individuals with biallelic deleterious variants in TMEM161B, which encodes a multi-pass transmembrane protein of unknown function. Tmem161b null mice demonstrated holoprosencephaly, craniofacial midline defects, eye defects, and spinal cord patterning changes consistent with impaired Shh signaling, but were without limb defect

This study investigated the prevalence of embryonic and connective tissue elements in the filum terminale (FT) of patients with tethered cord syndrome (TCS), examining both typical and pathological histology. The FT specimens from 288 patients who underwent spinal cord detethering from 2013 to 2021 were analyzed. The histopathological examination involved routine hematoxylin and eosin staining and specific immunohistochemistry when needed. The patient details were extracted from electronic medical records. The study found that 97.6% of the FT specimens had peripheral nerves, and 70.8% had regu

Mucinous neoplasms are commonly seen in the ovaries and pancreas. Their occurrence in the retroperitoneum is uncommon. We present a case of a retroperitoneal mucinous cystadenocarcinoma in a 54-year-old female who presented with right flank pain. Imaging demonstrated an 8.6 × 7.9 cm mass at the anterior surface of the lower pole of the right kidney, suspicious for renal cell carcinoma. Serum tumor markers carbohydrate antigen 19-9 (CA 19-9) and cancer embryonic antigen (CEA) were within normal limits, and cancer antigen 125 (CA 125) was elevated. Surgical resection of the mass was performed. I

The hereditary ataxias are a heterogenous group of disorders with an increasing number of causative genes being described. Due to the clinical and genetic heterogeneity seen in these conditions, the majority of such individuals endure a diagnostic odyssey or remain undiagnosed. Defining the molecular etiology can bring insights into the responsible molecular pathways and eventually the identification of therapeutic targets. Here, we describe the identification of biallelic variants in the GEMIN5 gene among seven unrelated families with nine affected individuals presenting with spastic ataxia a

The defective human survival motor neuron 1 (SMN1) gene leads to spinal muscular atrophy (SMA), the most common genetic cause of infant mortality. We previously reported that loss of SMN results in rapid differentiation of Drosophila germline stem cells and mouse embryonic stem cells (ESCs), indicating that SMN also plays important roles in germ cell development and stem cell biology. Here, we show that in healthy mice, SMN is highly expressed in the gonadal tissues, prepubertal spermatogonia, and adult spermatocytes, whereas low SMN expression is found in differentiated spermatid and sperm. Development of the complex structure of the vertebrate limb requires carefully orchestrated interactions between multiple regulatory pathways and proteins. Among these, precise regulation of 5' Hox transcription factor expression is essential for proper limb bud patterning and elaboration of distinct limb skeletal elements. Here, we identified Geminin (Gmnn) as a novel regulator of this process. A conditional model of Gmnn deficiency resulted in loss or severe reduction of forelimb skeletal elements, while both the forelimb autopod and hindlimb were unaffected. 5' Hox gene expression expanded

OBJECTIVE: The embryonic mesentery of the ascending and descending colons as well as the pancreas disappears due to peritoneal fusion, but there might be no or few photographic demonstrations of the intermediate morphologies during the process. The aims of this study were to characterize the morphological relationship of the interface between the renal fascia and peritoneum.
METHODS: Fourteen late-stage fetuses with crown rump lengths (CRLs) of 250-325 mm (gestational age: 30-38 weeks) were histologically examined.
RESULTS: The renal fascia, a thick or thin layer consisting of densely-distri

Structural maintenance of chromosomes flexible hinge domain-containing 1 (SMCHD1) is an architectural factor critical for X-chromosome inactivation (XCI) and the repression of select autosomal gene clusters. In mice, homozygous nonsense mutations in Smchd1 cause female-specific embryonic lethality due to an XCI defect. However, although human mutations in SMCHD1 are associated with congenital arhinia and facioscapulohumeral muscular dystrophy type 2 (FSHD2), the diseases do not show a sex-specific bias, despite the essential nature of XCI in humans. To investigate whether there is a dosage imb
