Extracellular application of the N-methyl-D-aspartate receptor allosteric modulator rapastinel acts remotely to regulate Ca2+ inactivation at an intracellular locus.

BACKGROUND: A novel N-methyl-D-aspartate receptor (NMDAR) allosteric modulator, rapastinel (RAP, formerly GLYX-13), elicits long-lasting antidepressant-like effects by enhancing long-term potentiation (LTP) of synaptic transmission. RAP elicits these effects by binding to a unique site in the extracellular region of the NMDAR complex, transiently enhancing NMDAR-gated current in pyramidal neurons of both hippocampus and medial prefrontal cortex. METHODS: We compared efficacy of RAP in modulating Schaffer collateral-evoked NMDAR-currents as a function of kinetics of the Ca2+ chelator in the in

Location and mapping of the human rostromedial tegmental nucleus and associated midbrain inhibitory nuclei regulating dopamine neurons.

Animal experiments reveal distinct GABAergic cell clusters within the dopaminergic midbrain regions in the rostromedial tegmental nucleus (RMTg) and retrorubral fields (RRF) that have yet to be clearly defined in humans. These neurons send prominent inhibitory projections to dopaminergic neurons in the substantia nigra and ventral tegmental area that impact motor, reward and threat processing. We have identified GABAergic RMTg and RRF cell clusters in 6 μm formalin-fixed paraffin-embedded transverse human midbrain sections from ten control cases obtained from the Sydney Brain Bank using immuno

Molecular mechanisms and regulatory networks of salt stress tolerance in plants.

Soil salinity is a major threat to crop productivity, sustainable agriculture, and global food security, with more than 833 million hectares of land affected worldwide. Salt stress restricts plant growth through osmotic stress, ion toxicity, oxidative damage, membrane disruption, reduced photosynthesis, and yield loss. Plants respond through coordinated regulatory networks that connect early stress perception with ion balance, osmotic adjustment, hormone signaling, transcript regulation, and protein modification. Recent advances have identified several sensory and signaling modules involved in

Environmental exposure to dinotefuran disturbs circadian rhythm, thyroid hormones and neurotransmitters, leading to locomotor deficits in Opsariichthys bidens larvae.

Dinotefuran (DIN) is a widely used neonicotinoid insecticide that has been detected in an increasing number of aquatic environments. However, the sublethal effects of DIN on native fish during their early developmental stages remain to be fully elucidated. The present study evaluated the developmental, behavioral, biochemical and transcriptomic responses of Opsariichthys bidens embryos and larvae exposed to three environmental concentrations of DIN (10, 100 and 1000 ng/L). The results evidenced that exposure to 1000 ng/L DIN resulted in a significant reduction in hatching rate at 48 hpf and a

Analyzing the Efficacy and Potential Risks of Nitrous Oxide for Pain Management During Labor: A Narrative Review.

Nitrous oxide has been used worldwide for labor analgesia for over a century. It offers a non-invasive, rapidly acting, and self-administered option for pain and anxiety management. The present investigation examines efficacy, safety, and limitations of nitrous oxide when compared to other labor analgesic modalities, including epidural anesthesia, intravenous opioids, and non-pharmacologic methods. Pharmacologically, nitrous oxide has both analgesic and anxiolytic effects via NMDA receptor inhibition, opioid receptor activation, and GABA-A modulation. While it can be less effective than epidur

Fermented red quinoa (FRQ) effectively mitigates chronic alcohol-induced cognitive impairment and hepatic steatosis through multi-mechanistic pathways.

Long-term alcohol consumption drives systemic damage through metabolites such as acetaldehyde, which trigger oxidative stress, inflammation, and gut dysbiosis. This study evaluated the protective effects of fermented red quinoa (FRQ) in an alcohol-exposed mouse model, with a focus on cognitive function. Male C57BL/6J mice were randomized into three groups for a 28-day study: a normal control, an alcohol-treated group gavaged with ethanol (1 mL/100 g·BW), and a group receiving the same ethanol dose co-administered with FRQ powder (human equivalent dose: 9 g/60 kg·BW). Our results demonstrated t

Activation of GABABR alleviates colitis by reprogramming macrophage polarization via the IRAK-M/NLRP3/NF-κB pathway.

Although the metabotropic GABA type B receptor (GABABR) has emerged as an immune modulator, its specific role in regulating the dysregulated macrophage polarization that drives inflammatory bowel disease (IBD) pathogenesis remains undefined. Utilizing a dextran sulfate sodium-induced colitis model and lipopolysaccharide-stimulated macrophages, we observed that colonic inflammation significantly downregulates GABABR expression concomitant with a pronounced shift toward M1 polarization. Pharmacological activation of GABABR with baclofen ameliorated clinical and histopathological manifestations,

Neural mass modelling of brain stimulation to Alleviate Schizophrenia biomarkers in brain rhythms.

We present a neural mass model (NMM) of the brain thalamo-cortico-thalamic (TCT) network to understand the effectiveness of non-invasive treatment with transcranial Direct Current Stimulation (tDCS) in reversing the anomalous electroencephalogram (EEG) oscillations in Schizophrenia. Our TCT NMM consists of twelve neural populations representing the thalamus and cortex modules of the visual pathway connected in a closed loop; the synaptic pathways are modelled with a 3-state kinetic framework allowing the inclusion of the slow excitatory N-methyl-D-aspartate-receptors (NMDAR). Indeed, a popular

Oxytocin-mediated neuroprotection in ischemic stroke: molecular mechanisms, therapeutic potential and clinical translational prospects.

Ischemic stroke (IS), also known as ischemic cerebrovascular accident, is a serious consequence of cerebral ischemia characterized by high morbidity, disability, and mortality. The primary causes of IS include atherosclerosis, cardiogenic embolism, large artery occlusion, and small artery disease. The occurrence of IS involves multiple cellular death mechanisms such as vascular obstruction, inflammatory response, excitotoxicity, oxidative stress, as well as neuronal apoptosis, necroptosis, and pyroptosis. Despite the application of current drugs and therapeutic strategies in the treatment of I

A novel fungal alkaloid protects retinal ganglion cells against glutamate-induced excitotoxicity in vitro and in vivo via activation of GABA pathway.

Retinal ganglion cells (RGCs) degeneration drives irreversible vision loss in glaucoma, ischemic/hereditary optic neuropathies, and demyelinating disorders, yet current therapies inadequately prevent RGC death. While fungal-derived compounds have revolutionized drug discovery, their neuroprotective potential remains underexplored. This study investigates the soil-derived fungus Umbelopsis sp. DWS 372 for novel neuroprotective agents targeting RGC preservation. Ten previously uncharacterized alkaloids and polyketides (1-10), including a new natural product (10), were isolated from rice-fermente

Systemic Glucose Regulation by a Hindbrain Inhibitory Circuit in a Mouse Model of Type 1 Diabetes.

INTRODUCTION: Previous work showed that increasing the electrical activity of inhibitory neurons in the dorsal vagal complex (DVC) is sufficient to increase whole-body glucose concentration in normoglycemic mice. Here we tested the hypothesis that deactivating GABAergic neurons in the dorsal hindbrain of hyperglycemic mice decreases synaptic inhibition of parasympathetic motor neurons in the dorsal motor nucleus of the vagus (DMV) and reduces systemic glucose levels. METHODS: Chemogenetic activation or inactivation of GABAergic neurons in the nucleus tractus solitarius (NTS) was used to asses

Hypothalamic Paraventricular Stimulation Inhibits Nociceptive Wide Dynamic Range Trigeminocervical Complex Cells via Oxytocinergic Transmission.

Oxytocinergic transmission blocks nociception at the peripheral, spinal, and supraspinal levels through the oxytocin receptor (OTR). Indeed, a neuronal pathway from the hypothalamic paraventricular nucleus (PVN) to the spinal cord and trigeminal nucleus caudalis (Sp5c) has been described. Hence, although the trigeminocervical complex (TCC), an anatomical area spanning the Sp5c, C1, and C2 regions, plays a role in some pain disorders associated with craniofacial structures (e.g., migraine), the role of oxytocinergic transmission in modulating nociception at this level has been poorly explored.

Anxiolytic effects of N-(4,5-dihydro-5-oxo-1,2-dithiolo-[4,3,b]-pyrrole-6-yl)-N-methylformamide, a pyrroloformamide isolated from a marine Streptomyces sp., in adult zebrafish by the 5-HT system.

General anxiety disorders are among the most prevalent mental health problems worldwide. The emergence and development of anxiety disorders can be due to genetic (30-50%) or non-genetic (50-70%) factors. Despite medical progress, available pharmacotherapies are sometimes ineffective or can cause undesirable side effects. Thus, it becomes necessary to discover new safe and effective drugs against anxiety. This study evaluated the anxiolytic effect in adult zebrafish (Danio rerio) of a natural pyrroloformamide (PFD), N-(4,5-dihydro-5-oxo-1,2-dithiolo-[4,3,b]-pyrrole-6-yl)-N-methylformamide, isol

Bilateral optogenetic activation of inhibitory cells favors ictogenesis.

Mesial temporal lobe epilepsy (MTLE) is the most common type of focal refractory epilepsy and is characterized by recurring seizures that are often refractory to medication. Since parvalbumin-positive (PV) interneurons were recently shown to play significant roles in ictogenesis, we established here how bilateral optogenetic stimulation of these interneurons in the hippocampus CA3 regions modulates seizures, interictal spikes and high-frequency oscillations (HFOs; ripples: 80-200 Hz, fast ripples: 250-500 Hz) in the pilocarpine model of MTLE.
