A tributyrin-monolaurin complex protects piglets against PEDV infection via viral inhibition, lipid metabolism restoration, and microbiota modulation.

Porcine epidemic diarrhea virus (PEDV) is a major pathogen causing severe diarrhea and intestinal damage in piglets. Monolaurin (GML) and tributyrin (TB) are well-documented functional lipids with intestinal health benefits, but their combined efficacy against PEDV infection remains unclear. The present study first investigated the anti-PEDV effect of GML, TB, and their mixtures using PEDV-infected Vero cells. Then, the effects of a microencapsulated TB-GML complex (PE) were evaluated in PEDV-infected piglets, focusing on its protective mechanisms. Vero cells were simultaneously treated with P

Multi-omics integration reveals the ameliorative effects and underlying mechanisms of Astragalus membranaceus (Huangqi)-Fuzhuan brick tea on nonalcoholic fatty liver disease.

ETHNOPHARMACOLOGICAL RELEVANCE: Nonalcoholic fatty liver disease (NAFLD) is a major clinical challenge and a growing global public health burden, yet no pharmacological therapy specific to this disease has been approved to date. Notably, Astragalus membranaceus (Huangqi, HQ) is incorporated into approximately 80% of multi-herb formulations employed for treating liver diseases. Fuzhuan brick tea, a distinctive Chinese fermented tea, is widely recognized for its unique fermentation process and hypolipidemic properties. However, whether co-fermentation with HQ enhances its lipid-lowering efficacy

Sodium-glucose cotransporter 2 inhibitors in breast cancer patients on endocrine therapy: a targeted strategy to mitigate long-term cardiometabolic risk.

BACKGROUND: Hormone receptor-positive breast cancer (HR+ BC) represents the most common breast cancer subtype and is increasingly characterized by long-term survivorship complicated by substantial cardiometabolic and cardiovascular morbidity. Endocrine therapies, including aromatase inhibitors, selective estrogen receptor modulators and degraders, and ovarian suppression, remain central to disease control but promote metabolic dysfunction, vascular injury, and systemic inflammation, thereby increasing cardiovascular disease (CVD) risk and potentially contributing to endocrine resistance. These

Liver microRNA transcriptome reveals miR-182 as link between type 2 diabetes and fatty liver disease in obesity.

BACKGROUND: The development of obesity-associated comorbidities such as type 2 diabetes (T2D) and hepatic steatosis has been linked to selected microRNAs in individual studies; however, an unbiased genome-wide approach to map T2D induced changes in the miRNAs landscape in human liver samples, and a subsequent robust identification and validation of target genes are still missing. METHODS: Liver biopsies from age- and gender-matched obese individuals with (n=20) or without (n=20) T2D were used for microRNA microarray analysis. The candidate microRNA and target genes were validated in 85 human

Exposure to high fructose corn syrup during adolescence in the mouse alters hepatic metabolism and the microbiome in a sex-specific manner.

KEY POINTS: The prevalence of obesity and non-alcoholic fatty liver disease in children is dramatically increasing at the same time as consumption of foods with a high sugar content. Intake of high fructose corn syrup (HFCS) is a possible aetiology as it is thought to be more lipogenic than glucose. In a mouse model, HFCS intake during adolescence increased fat mass and hepatic lipid levels in male and female mice. However, only males showed impaired glucose tolerance. Multiple metabolites including lipids, bile acids, carbohydrates and amino acids were altered in liver in a sex-specific manne

Lipid metabolic reprogramming of tumor-associated macrophages drives resistance to immune checkpoint blockade in lung cancer: a narrative review of mechanisms and therapeutic strategies.

BACKGROUND AND OBJECTIVE: Immune checkpoint inhibitors (ICIs), represented by programmed cell death protein 1 (PD-1)/programmed death-ligand 1 (PD-L1), have shown remarkable efficacy in non-small cell lung cancer (NSCLC); however, many patients still develop resistance to immunotherapy. Although small cell lung cancer (SCLC) is also an important histological type of lung cancer, NSCLC accounts for the majority of lung cancer cases. Current research on ICI development, first-line treatment efficacy, and the mechanisms of lipid metabolism in tumor-associated macrophages (TAMs) is predominantly f

Dietary copper supplementation enhances lipolysis in Rex rabbits.

BACKGROUND: Copper is an important regulator of lipid metabolism in mammals, as a cofactor of many enzymes and is involved in the lipolysis. Copper deficiency has been considered as a significant factor in human diseases related to abnormal lipid metabolism, while adding copper to the diet seems to be the simplest and most effective way to prevent copper deficiency. AIMS: The aim of this study was to investigate the effects of dietary copper level on lipid metabolism in Rex Rabbits. METHODS: A total of 120 90-d-old Rex Rabbits were randomly allotted into three treatments, with 40 replicates

Dysregulation of epicardial adipose tissue in cachexia due to heart failure: the role of natriuretic peptides and cardiolipin.

BACKGROUND: Cachexia worsens long-term prognosis of patients with heart failure (HF). Effective treatment of cachexia is missing. We seek to characterize mechanisms of cachexia in adipose tissue, which could serve as novel targets for the treatment. METHODS: The study was conducted in advanced HF patients (n = 52; 83% male patients) undergoing heart transplantation. Patients with ≥7.5% non-intentional body weight (BW) loss during the last 6 months were rated cachectic. Clinical characteristics and circulating markers were compared between cachectic (n = 17) and the remaining, BW-stable patien

Characterising nitric oxide-mediated metabolic benefits of low-dose ultraviolet radiation in the mouse: a focus on brown adipose tissue.

AIMS/HYPOTHESIS: Exposure to sunlight has the potential to suppress metabolic dysfunction and obesity. We previously demonstrated that regular exposure to low-doses of ultraviolet radiation (UVR) reduced weight gain and signs of diabetes in male mice fed a high-fat diet, in part via release of nitric oxide from skin. Here, we explore further mechanistic pathways through which low-dose UVR exerts these beneficial effects. METHODS: We fed mice with a luciferase-tagged Ucp1 gene (which encodes uncoupling protein-1 [UCP-1]), referred to here as the Ucp1 luciferase transgenic mouse ('Thermomouse')

Mutant p53-R280K hijacks SREBP1 to promote fatty acid synthesis and breast cancer progression via FASN.

BACKGROUND: Lipid metabolic reprogramming is a hallmark of cancer, in which fatty acid synthesis is crucial for the rapid proliferation and metastasis of cancer cells. The p53-R280K mutation drives aggressive cancer, yet its role in fatty acid synthesis remains unclear. METHODS: The CRISPR/CasRx technique was applied to knockdown endogenous mutant p53-R280K in MDA-MB-231 cells and wild-type p53 (WT-p53) in ZR75-1 cells. Luciferase reporter and ChIP assays demonstrated mutant p53-R280K binding to the FASN promoter and regulating its activity. Oil red O staining, flow cytometry, and triglycerid

SREBP-1c impairs ULK1 sulfhydration-mediated autophagic flux to promote hepatic steatosis in high-fat-diet-fed mice.

A metabolic imbalance between lipid synthesis and degradation can lead to hepatic lipid accumulation, a characteristic of patients with non-alcoholic fatty liver disease (NAFLD). Here, we report that high-fat-diet-induced sterol regulatory element-binding protein (SREBP)-1c, a key transcription factor that regulates lipid biosynthesis, impairs autophagic lipid catabolism via altered H2S signaling. SREBP-1c reduced cystathionine gamma-lyase (CSE) via miR-216a, which in turn decreased hepatic H2S levels and sulfhydration-dependent activation of Unc-51-like autophagy-activating kinase 1 (ULK1). Inhibition of Δ24-dehydrocholesterol reductase activates pro-resolving lipid mediator biosynthesis and inflammation resolution.

Targeting metabolism through bioactive key metabolites is an upcoming future therapeutic strategy. We questioned how modifying intracellular lipid metabolism could be a possible means for alleviating inflammation. Using a recently developed chemical probe (SH42), we inhibited distal cholesterol biosynthesis through selective inhibition of Δ24-dehydrocholesterol reductase (DHCR24). Inhibition of DHCR24 led to an antiinflammatory/proresolving phenotype in a murine peritonitis model. Subsequently, we investigated several omics layers in order to link our phenotypic observations with key metabolic

Genistein affects adipose tissue deposition in a dose-dependent and gender-specific manner.

The soy isoflavone genistein targets adipose tissue and elicits physiological effects that may vary based on dietary intake. We hypothesized that the adipose effects of genistein are dose and gender dependent. Four-week-old C57BL/6 male and female mice received daily oral doses of genistein (50-200,000 microg/kg.d) or 17beta-estradiol (E2) (5 microg/kg.d) for 15 d or a diet containing 800 ppm genistein. Genistein increased epididymal and renal fat pad and adipocyte size at doses up to 50,000 microg/kg.d or at 800 ppm in the diet in males but not in females. The alteration in adipocity correlat
