Oxygen Availability and Metabolic Dynamics During Mycobacterium tuberculosis Latency.

OBJECTIVE: In vitro models of Mycobacterium tuberculosis (Mtb) nonreplicating persistence (NRP) suggest the rate of oxygen ( O2 ) depletion is a significant determinant in persistence. However, few studies have characterized the metabolic association between slow and rapid O2 depletion rates and successful versus failed persistence. METHODS: We developed a theoretical model of Mtb metabolic adaptation that includes O2 driven genetic modulation of enzymes in the tricarboxylic acid cycle, energy, and redox recycling pathways. We conducted an in silico study of Mtb adaptation, and investigated t

Vacated niches, competitive release and the community ecology of pathogen eradication.

A recurring theme in the epidemiological literature on disease eradication is that each pathogen occupies an ecological niche, and eradication of one pathogen leaves a vacant niche that favours the emergence of new pathogens to replace it. However, eminent figures have rejected this view unequivocally, stating that there is no basis to fear pathogen replacement and even that pathogen niches do not exist. After exploring the roots of this controversy, I propose resolutions to disputed issues by drawing on broader ecological theory, and advance a new consensus based on robust mechanistic princip

The maximum attainable body size of herbivorous mammals: morphophysiological constraints on foregut, and adaptations of hindgut fermenters.

An oft-cited nutritional advantage of large body size is that larger animals have lower relative energy requirements and that, due to their increased gastrointestinal tract (GIT) capacity, they achieve longer ingesta passage rates, which allows them to use forage of lower quality. However, the fermentation of plant material cannot be optimized endlessly; there is a time when plant fibre is totally fermented, and another when energy losses due to methanogenic bacteria become punitive. Therefore, very large herbivores would need to evolve adaptations for a comparative acceleration of ingesta pas

The crucible of resilience: hormesis as the unifying principle of evolution, genetics, and epigenetics.

The traditional linear no-threshold model of toxicity and stress fails to capture the complex, adaptive nature of biological systems. Hormesis-a biphasic dose-response phenomenon where low-dose stress induces beneficial, adaptive responses while high doses cause harm-offers a more comprehensive paradigm. This article asserts that hormesis is a major, unifying explanatory principle of life that efficiently integrates evolutionary theory, genetics, and epigenetics. From an evolutionary perspective, hormesis is an energy-efficient strategy of adaptive homeostasis, permitting organisms to build bi

Regulatory Roles of a Novel MarR-like Protein Lmo0840 in Tetracycline-Associated Growth Phenotype and Virulence of Listeria monocytogenes.

Listeria monocytogenes (L. monocytogenes) is a zoonotic foodborne pathogen that triggers life-threatening invasive illnesses. MarR-type transcriptional regulators widely control bacterial stress adaptation, antibiotic tolerance and virulence, playing crucial roles in the pathogenesis of diverse bacterial pathogens. Lmo0840 is a conserved member of this family; however, its physiological functions and regulatory networks in L. monocytogenes remain completely unexplored. To this end, we constructed an lmo0840 deletion mutant and its complemented strain in the L. monocytogenes EGD-e background an

Effects of morphine withdrawal on the membrane properties of medium spiny neurons in the nucleus accumbens shell.

Medium spiny neurons (MSNs) in the nucleus accumbens (NAc) undergo persistent alterations in their biological and physiological characteristics upon exposure to drugs of abuse. Previous studies demonstrated that the biochemical, morphological, and intrinsic physiological properties of MSNs are heterogeneous and provided new insights into the physiological and molecular roles of individual MSNs in addictive behaviors. However, it remains unclear whether MSNs in the NAc shell (NAcSh), an important region for mediating behavioral sensitization, are electrophysiologically heterogeneous and how suc

Dynamic remodeling of membrane proteins for temperature adaptation in oysters.

Rising ocean temperatures due to global warming pose severe threats to marine organism survival. Membrane proteins perform important biological functions and serve as sensitive environmental stress sensors. However, the characterization of membrane protein content in response to temperature adaptation remains poorly understood in marine organisms. Here, we performed reciprocal transplantation experiments between the northern/cold and southern/warm natural habitats of two allopatric but closely related oyster species, Crassostrea gigas and Crassostrea angulata. Comparative membrane proteomics c

[Confronting the Health-Related Challenges of Climate Change: Nursing Education for the Future].

Climate change is the greatest threat to public health in the 21st century. The increasing health impact of heat waves, the increasing magnitudes and spatial expansions of vector and water-borne diseases epidemics, and the increasing medical burdens of biological allergic illnesses, worsening local air pollution, and other related issues are expected to continue to increase in severity in the near future. All of these issues are global problems that must be faced. Adaptation strategies and action plans related to climate change are needed and emerging. Moreover, integrating the basic concepts,

The Eye's Armour: Case Report of Cartilage in the Sclera of the Himalayan Golden Eagle and Focused Review of Vertebrate Ocular Skeletal Adaptations to Visual Ecology.

The vertebrate eye, while commonly perceived as a soft-tissue structure, is reinforced in numerous taxa by an internal ocular skeleton composed of scleral cartilage and ossicles. This manuscript presents a histological case report documenting scleral cartilage in the Himalayan Golden Eagle (Aquila chrysaetos daphanea) and integrates these findings with a comprehensive synthesis of current understanding of the ocular skeleton's evolution, development and functional morphology. These findings are consistent with previously described avian scleral architecture. Although based on a single specimen

Deep origins, distinct adaptations, and species-level status indicated for a glacial relict seal.

Isolated populations of postglacial relicts are known from many regions and are typically found on mountains for terrestrial species and in lakes for aquatic species. Among the few aquatic mammalian relicts, the Saimaa ringed seal (Pusa hispida saimensis) has been landlocked in Lake Saimaa, Finland, for the last 10,000 y. Saimaa ringed seals show genetic, behavioral, and morphological differences from the other ringed seal subspecies, but the extent these differences stem from the end of the last glacial period remains unclear. Here, we demonstrate with comprehensive sampling and state-of-the-

Moving in an Uncertain World: Robust and Adaptive Control of Locomotion from Organisms to Machine Intelligence.

Whether walking, running, slithering, or flying, organisms display a remarkable ability to move through complex and uncertain environments. In particular, animals have evolved to cope with a host of uncertainties-both of internal and external origin-to maintain adequate performance in an ever-changing world. In this review, we present mathematical methods in engineering to highlight emerging principles of robust and adaptive control of organismal locomotion. Specifically, by drawing on the mathematical framework of control theory, we decompose the robust and adaptive hierarchical structure of

Endocranial anatomy and phylogenetic position of the crocodylian Eosuchus lerichei from the late Paleocene of northwestern Europe and potential adaptations for transoceanic dispersal in gavialoids.

Eosuchus lerichei is a gavialoid crocodylian from late Paleocene marine deposits of northwestern Europe, known from a skull and lower jaws, as well as postcrania. Its sister taxon relationship with the approximately contemporaneous species Eosuchus minor from the east coast of the USA has been explained through transoceanic dispersal, indicating a capability for salt excretion that is absent in extant gavialoids. However, there is currently no anatomical evidence to support marine adaptation in extinct gavialoids. Furthermore, the placement of Eosuchus within Gavialoidea is labile, with some a

The multifactor pelvis: An alternative to the adaptationist approach of the obstetrical dilemma.

The obstetrical dilemma describes the competing demands that a bipedally adapted pelvis and a large-brained neonate place on human childbirth and is the predominant model within which hypotheses about the evolution of the pelvis are framed. I argue the obstetrical dilemma follows the adaptationist program outlined by Gould and Lewontin in 1979 and should be replaced with a new model, the multifactor pelvis. This change will allow thorough consideration of nonadaptive explanations for the evolution of the human pelvis and avoid negative social impacts from considering human childbirth inherentl

Elucidation of the coping strategy in an OMP homozygous knockout mutant of Synechocystis 6803 defective in iron uptake.

The TonB-dependent outer membrane (OM) transporters import iron (Fe) under Fe-deficient growth conditions in the unicellular cyanobacterium Synechocystis sp. strain PCC 6803 (S. 6803 or WT). The present study characterises the mechanisms employed by a homozygous Δslr1908 (Slr1908, an OM protein) mutant of S. 6803 to adapt to intracellular iron deficiency imposed due to lack of Slr1908 when grown under Fe sufficient conditions for short (1 week) and long term (3 weeks). Although the homozygous Δslr1908 cells showed symptoms of stress such as slow growth, high ROS and altered ultrastructure, the

From Empirical to Pathogenesis-Based Treatments for Psoriasis.

Since the foundation of the European Society for Dermatological Research, pathogenesis of psoriasis has been studied by many research groups, focusing on various compartments of the skin. Understanding of the pathogenesis of psoriasis has evolved into a branching model of innate and acquired immunity. Insights in the genetics of psoriasis proved to be compatible with this model. Inspired by these insights, pathogenesis-based treatments have emerged with unprecedented efficacy and sustainability. In particular, the cytokine network harbors major treatment targets for biologics with TNF-α, the I

Competition for Manganese at the Host-Pathogen Interface.

Transition metals such as manganese are essential nutrients for both pathogen and host. Vertebrates exploit this necessity to combat invading microbes by restricting access to these critical nutrients, a defense known as nutritional immunity. During infection, the host uses several mechanisms to impose manganese limitation. These include removal of manganese from the phagolysosome, sequestration of extracellular manganese, and utilization of other metals to prevent bacterial acquisition of manganese. In order to cause disease, pathogens employ a variety of mechanisms that enable them to adapt

Postnatal development of the skull of Dinilysia patagonica (Squamata-stem Serpentes).

The snake skull represents a profound transformation of the ancestral squamate cranium in which dermal skull roof bones were integrated with the braincase, in a manner convergent with that which occurred during the origin of mammals. However, the ontogeny of snake characters at the origin of the clade has until now been inaccessible. Here we describe a postnatal ontogenetic series of the Late Cretaceous stem snake Dinilysia patagonica and compare it to that of extant lizards and snakes. Comparative analysis indicates notable ontogenetic changes, including advanced state of ossification, isomet

Elastic modulus of cetacean auditory ossicles.

In order to model the hearing capabilities of marine mammals (cetaceans), it is necessary to understand the mechanical properties, such as elastic modulus, of the middle ear bones in these species. Biologically realistic models can be used to investigate the biomechanics of hearing in cetaceans, much of which is currently unknown. In the present study, the elastic moduli of the auditory ossicles (malleus, incus, and stapes) of eight species of cetacean, two baleen whales (mysticete) and six toothed whales (odontocete), were measured using nanoindentation. The two groups of mysticete ossicles o
