Preglomerular microcirculation expresses the cAMP-adenosine pathway.

UNLABELLED: The purpose of this study was to investigate whether the extracellular cAMP-adenosine pathway (i.e., transport of cAMP out of cells followed by extracellular conversion of cAMP to adenosine) exists in preglomerular microvessels (PGMVs). Incubation of PGMVs for 1 h with 30 microM cAMP increased the amount of extracellular adenosine from 163 +/- 18.6 (n = 18) to 9810 +/- 604 (n = 12) pmol/mg of protein (P < 10(-6)). The phosphodiesterase inhibitor 3-isobutyl-1-methylxanthine (IBMX; 1 mM; n = 6) and the ecto-phosphodiesterase inhibitor 1, 3-dipropyl-8-p-sulfophenylxanthine (DPSPX; 1 m

Adenosine triphosphate: established and potential clinical applications.

Adenosine 5'-triphosphate (ATP) is a purine nucleotide found in every cell of the human body. In addition to its well established role in cellular metabolism, extracellular ATP and its breakdown product adenosine, exert pronounced effects in a variety of biological processes including neurotransmission, muscle contraction, cardiac function, platelet function, vasodilatation and liver glycogen metabolism. These effects are mediated by both P1 and P2 receptors. A cascade of ectonucleotidases plays a role in the effective regulation of these processes and may also have a protective function by ke

Adenosine-induced atrial arrhythmia: a prospective analysis.

BACKGROUND: Adenosine is considered safe and effective for paroxysmal supraventricular tachycardia (PSVT), but anecdotal experience suggests that adenosine can precipitate atrial arrhythmias. OBJECTIVES: To determine the frequency and mechanisms of adenosine-induced atrial arrhythmias. SETTING: Clinical electrophysiology laboratory at a university medical center. DESIGN: Prospective observational study. PATIENTS: 200 consecutive patients with PSVT undergoing an electrophysiology procedure. INTERVENTION: During PSVT, 12 mg of adenosine was administered centrally through the femoral vein.

Adenosine-induced non-sustained polymorphic ventricular tachycardia.

Adenosine has become widely used because of its diagnostic and therapeutic value in the emergency management of arrhythmias. It produces transient heart block by slowing conduction through the AV node and thus terminates supraventricular tachycardias that involve the atrioventricular node. Bradyarrhythmias of short duration are common side effects of the use of this drug. Premature atrial and ventricular beats have also been reported. The very short half-life and lack of serious adverse effects generally lead to the consideration that adenosine is a safe drug. We describe a 56-year-old woman w

Regional adenosine attenuates postischemic spinal cord injury.

PURPOSE: Operations on the thoracic and thoracoabdominal aorta are associated with postoperative paraplegia rates as high as 20% to 30%. Attempts to reduce this complication have focused on decreasing the energy needs of the spinal cord with protective agents such as hypothermia and barbiturates or on increasing blood flow with various shunts. This study explores the hypothesis that perfusion of the spinal cord with hypothermic solutions or with adenosine will prevent or ameliorate paraplegia. METHODS: New Zealand white rabbits underwent 40 minutes of infrarenal aortic isolation. The infraren

Methyl xanthine, adenosine, and human taste responsivity.

The influences of three methyl xanthines (MX) on human taste responsivity, caffeine, theophylline, and theobromine, were examined using blind control procedures. Taste responsivity in the same subjects was determined using the matching procedure described by Schiffman (Study 1) and the whole-mouth procedure described by Sheperd (Study 2). In each study, the duration of MX pretreatment necessary to enhance taste responsivity was examined. No potentiation of overall- and taste-quality specific intensity ratings was observed for any tastant, independent of test procedure, type, and concentration

Adenosine: novel antiarrhythmic therapy for supraventricular tachycardia.

Adenosine (Adenocard) is a unique new agent for the acute treatment of paroxysmal supraventricular tachycardia (PSVT). Administered by intravenous bolus, it has an onset and duration measured in seconds and greater than 90% efficacy. Its primary effect is to slow atrioventricular nodal conduction, thus converting reentrant forms of PSVT to normal sinus rhythm. Side effects quickly dissipate without treatment because of the short duration of action. Other uses include diagnosis of broad or wide QRS complex tachycardias and controlled intraoperative hypotension. Its short duration and high effic

Lymphocytic adenosine deaminase activity in typhoid fevers.

Lymphocyte adenosine deaminase (L-ADA) activity, a measure of lymphocyte activity, was estimated in 10 healthy controls and 30 patients with typhoid fever (20 uncomplicated and 10 complicated) at the time of admission, at onset of complications and weekly until recovery. Mean L-ADA activity in healthy controls was 20.49 +/- 3.62 mU/10(6) cells. In uncomplicated patients L-ADA activity was 36.33 +/- 5.09 mU/10(6) cells at time of admission, which is significantly raised as compared to controls. It remained high at the height of the fever and at defervescence. In complicated patients L-ADA activ

Adenosine decreases neurotransmitter release at central synapses.

Adenosine, at concentrations ranging from 5 to 100 microM, decreases the efficacy of transmission at the perforant path synapses on dentate granule cells. We have used whole cell recording from these cells in slices to determine the mechanism of the reduced synaptic strength. We find that size of miniature excitatory postsynaptic currents (mepscs) is unaffected by adenosine at concentrations up to 100 microM, an observation that indicates adenosine's mode of action is not through a decreased postsynaptic sensitivity to neurotransmitter. A quantal analysis indicates, however, that the quantity

Serum adenosine deaminase: isoenzymes and diagnostic application.

Human adenosine deaminase (ADA; EC 3.5.4.4) consists of three isoenzymes: ADA1, ADA1+CP, and ADA2. We developed an electrophoretic technique to distinguish between these three isoenzymes. The isoenzyme pattern was studied in tissue and cell homogenates, as well as in serum from normal subjects and from patients with increased serum ADA who had either hepatitis, infectious mononucleosis, tuberculosis, pneumonia, rheumatoid arthritis, or acute lymphoblastic leukemia (ALL). The highest ADA activity was found in lymphocytes and monocytes. ADA2 could be detected only in monocytes (18% of total ADA

Behavioral characteristics of centrally administered adenosine analogs.

Mice were implanted with chronic indwelling cannulae in the lateral cerebral ventricle. A series of adenosine analogs and related compounds were injected into the lateral ventricle (ICVT) and their effects on spontaneous locomotor activity recorded. All analogs produced dose-related decreases in locomotor activity. 5'-N6-ethyl-carboxamidoadenosine (NECA) was the most potent compound tested, with a number of N6-substituted analogs also being effective depressants of activity. Caffeine, administered either intracerebroventricularly or intraperitoneally, antagonized the depressant effects of the

Sick sinus syndrome: an adenosine-mediated disease.

It is suggested that adenosine is a mediator of the paroxysmal bradycardias and tachycardias characterising the sick sinus syndrome. Increased sensitivity to adenosine could account for the arrhythmias, which might therefore be suppressed by an adenosine antagonist such as theophylline.

Adenosine diphosphate ribose pyrophosphohydrolase in human skin.

Adenosine diphosphate ribose (ADPR) pyrophosphohydrolase (ADPR-PPase), which catalyzes the hydrolysis of ADPR to yield adenosine monophosphate (AMP) and ribose-5'-phosphate, was assayed in human penile foreskin. Since ADPR is formed from nicotinamide adenine dinucleotide (NAD) by NAD glycohydrolase (NADase), NADase was also assayed in human skin. The skin tissue obtained by circumcision was separated into three layers; epidermis of the outer prepuce, epidermis of the inner prepuce, and dermis. ADPR-PPase was found to be present in all of the three layers with nearly equal activity. NADase was

Adenosine 5'-Carboxamide-Based Inhibitors of METTL1.

METTL1 is the human RNA methyltransferase that catalyzes the methylation of N7-guanosine in RNA. Overexpression of METTL1 has been linked to cancer, and increasing evidence supports the therapeutic potential of METTL1 inhibition in oncology. In this study, we have optimized a series of adenosine 5'-carboxamide derivatives as METTL1 inhibitors through structure-guided modifications of a previously discovered hit compound. The advanced inhibitor B22 shows an IC50 of 1 μM in an enzymatic assay, which is a 178-fold improvement with respect to the initial hit. The crystal structure of the des-methy

Updated Management Guidelines for Adenosine Deaminase Deficiency.

Inherited defects in the adenosine deaminase (ADA) gene typically cause severe combined immunodeficiency. In addition to infections, ADA-deficient patients can present with neurodevelopmental, behavioral, hearing, skeletal, lung, heart, skin, kidney, urogenital, and liver abnormalities. Some patients also suffer from autoimmunity and malignancies. In recent years, there have been remarkable advances in the management of ADA deficiency. Most ADA-deficient patients can be identified by newborn screening for severe combined immunodeficiency, which facilitates early diagnosis and treatment of asym

Deficiency of adenosine deaminase 2 (DADA2): Review.

The deficiency of adenosine deaminase 2 (DADA2) is an autosomal recessive disease caused by loss-of-function (LOF) mutations in the ADA2 gene and was first described in 2014. Initially, it was described as vasculopathy/vasculitis that mostly affected infants and young children and closely resembled polyarteritis nodosa (PAN). Skin rash and ischemic/hemorrhagic stroke are predominant symptoms. However, the clinical spectrum of DADA2 has continued to expand since then. It has now been reported in adults as well. Besides vasculitis-related manifestations, hematological, immunological, and autoinf

Structure of Klebsiella pneumoniae adenosine monophosphate nucleosidase.

Klebsiella pneumoniae is a bacterial pathogen that is increasingly responsible for hospital-acquired pneumonia and sepsis. Progressive development of antibiotic resistance has led to higher mortality rates and creates a need for novel treatments. Because of the essential role that nucleotides play in many bacterial processes, enzymes involved in purine and pyrimidine metabolism and transport are ideal targets for the development of novel antibiotics. Herein we describe the structure of K. pneumoniae adenosine monophosphate nucleosidase (KpAmn), a purine salvage enzyme unique to bacteria, as de
