Diabetic neuropathy.

Diabetic neuropathy (DN) refers to symptoms and signs of neuropathy in a patient with diabetes in whom other causes of neuropathy have been excluded. Distal symmetrical neuropathy is the commonest accounting for 75% DN. Asymmetrical neuropathies may involve cranial nerves, thoracic or limb nerves; are of acute onset resulting from ischaemic infarction of vasa nervosa. Asymmetric neuropathies in diabetic patients should be investigated for entrapment neuropathy. Diabetic amyotrophy, initially considered to result from metabolic changes, and later ischaemia, is now attributed to immunological ch

Aldose reductase activation is a key component of myocardial response to ischemia.

Aldose reductase, a member of the aldo-keto reductase family, has been implicated in the development of vascular and neurological complications in diabetes. Despite recent studies from our laboratory demonstrating protection of ischemic hearts by an aldose reductase inhibitor, the presence and influence of aldose reductase in cardiac tissue remain unknown. Our goal in this study was to isolate and characterize the kinetic properties of cardiac aldose reductase, as well as to study the impact of flux via this enzyme on glucose metabolism and contractile function in hearts subjected to ischemia-

Application of 19F-n.m.r. spectroscopy to the identification of dog urinary metabolites of imirestat, a spirohydantoin aldose reductase inhibitor.

1. Urine from a dog dosed orally at 20 mg/kg with 14C-imirestat, a spirohydantoin aldose reductase inhibitor, contained 17.7 and 12.5% of the administered radioactivity at 0-48 and 48-72 h respectively. 2. Radio-h.p.l.c. of the 0-48 h urine revealed a complex mixture of metabolites and a small proportion of parent drug (1.6% of dose). Direct 19F-n.m.r. spectroscopy of this urine showed the fluoride ion, numerous metabolites which were predominantly glucuronide conjugates and, as a minor component, the parent drug. 3. After incubation with beta-glucuronidase the 0-48 h urine gave a 19F-n.m.r. s

Obligately Tungsten-Dependent Enzymes─Catalytic Mechanisms, Models and Applications.

Tungsten-dependent enzymes incorporate a tungsten ion into their active site in the form of a complex with two pyranometallopterin (MPT) molecules, also known as tungsten cofactor (W-co). W-co-containing enzymes are found in several bacteria and archaea, predominantly in enzymes involved in anaerobic metabolism. While some enzymes occur with either molybdenum or tungsten in their active sites, we concentrate here on enzymes obligately depending on W-co, which are not functional as isoenzymes with Mo-co. These are represented by several subtypes of aldehyde oxidoreductases (AORs), class II benz

NRF2 inhibits RSL3 induced ferroptosis in gastric cancer through regulation of AKR1B1.

Gastric cancer is a malignant tumor associated with a high mortality rate. Recently, emerging evidence has shown that ferroptosis, a regulated form of cell death induced by iron (Fe)-dependent lipid peroxidation. Nuclear factor E2 related factor 2 (NRF2) is a key regulator of intracellular oxidation homeostasis that plays a pivotal role in controlling lipid peroxidation, which is closely related to the process of ferroptosis. However, the molecular mechanism of NRF2 on ferroptosis remains to be investigated in gastric cancer. In our study, NRF 2 was found to transcriptionally activate Aldo-ket

Novel luciferase reporter system for in vitro and organ-specific monitoring of differential gene expression in Listeria monocytogenes.

In this paper we describe construction of a luciferase-based vector, pPL2lux, and use of this vector to study gene expression in Listeria monocytogenes. pPL2lux is a derivative of the listerial integration vector pPL2 and harbors a synthetic luxABCDE operon encoding a fatty acid reductase complex (LuxCDE) involved in synthesis of the fatty aldehyde substrate for the bioluminescence reaction catalyzed by the LuxAB luciferase. We constructed pPL2lux derivatives in which the secA and hlyA promoters were translationally fused to luxABCDE and integrated as a single copy into the chromosome of L. mo

High-dose vitamin therapy stimulates variant enzymes with decreased coenzyme binding affinity (increased K(m)): relevance to genetic disease and polymorphisms.

As many as one-third of mutations in a gene result in the corresponding enzyme having an increased Michaelis constant, or K(m), (decreased binding affinity) for a coenzyme, resulting in a lower rate of reaction. About 50 human genetic dis-eases due to defective enzymes can be remedied or ameliorated by the administration of high doses of the vitamin component of the corresponding coenzyme, which at least partially restores enzymatic activity. Several single-nucleotide polymorphisms, in which the variant amino acid reduces coenzyme binding and thus enzymatic activity, are likely to be remediabl

Induction by xenobiotics of phase I and phase II enzyme activities in the human keratinocyte cell line NCTC 2544.

This study analyses the expression and induction of several drug-metabolising enzyme activities involved in either phase I or phase II biotransformations in NCTC 2544 human keratinocytes. The phase I activities 7-ethoxycoumarin O-deethylase (ECOD), 7-ethoxyresorufin O-deethylase (EROD) and 7-pentoxyresorufin O-depenthylase (PROD) were easily detectable in basal conditions. During incubations lasting up to 144 h in the presence of the classical cytochrome P450 inducers beta-naphthoflavone (BNF), 3-methylcholanthrene (MC) and phenobarbital (PB), a considerable and significant increase in all the

The mechanism of cytochrome P-450-dependent oxidation of ethanol in reconstituted membrane vesicles.

The mechanism of cytochrome P-450-dependent oxidation of ethanol has been investigated using reconstituted phospholipid vesicles containing purified preparations of rabbit liver microsomal NADPH-cytochrome P-450 reductase and cytochrome P-450 LM2. Incorporation of cytochrome b5 into the vesicles resulted in a 5-fold enhancement of cytochrome P-450-catalyzed O-dealkylation of 7-ethoxycoumarin, whereas the cytochrome P-450-dependent ethanol oxidation was slightly inhibited. Superoxide dismutase, added in increasing amounts to the vesicles, inhibited the formation of superoxide anions and, in a c

Mitochondrial Quality Regulation Genes as Prognostic Markers in Hepatocellular Carcinoma: Tumor Microenvironment, Therapeutic Response, and Drug Sensitivity Analysis.

Aberrant expression of mitochondrial quality regulation genes (MQRGs) is intricately linked to mitochondrial dysfunction and the progression of hepatocellular carcinoma (HCC), highlighting the urgent need for reliable prognostic biomarkers. In this study, we aimed to identify differentially expressed MQRGs from 20 candidate genes by analyzing transcriptomic profiles and clinical records from the TCGA (n = 371) and GEO (n = 167) datasets. The identified prognostic MQRGs, along with associated subtype differentially expressed genes (DEGs, n = 156), underwent LASSO and multivariate Cox regression

Asborin inhibits Aldo/Keto reductase 1A1.

Asborin is the carbaborane analogue of aspirin. Replacement of the phenyl ring in aspirin by ortho-carbaborane was found to change the pharmacological profile of the compound remarkably. Unlike aspirin, asborin cannot selectively acetylate a single serine residue in the active site of cyclooxygenase, and as a result inhibitory potency is reduced. Activation of the acetyl group and the presence of the hydrophobic and bulky cluster therefore did not meet the requirements for cyclooxygenase inhibition. Both features, however, match perfectly for inhibition of the aldo/keto reductase family. Herei

Metabolomics-driven insights into the multi-target antibacterial mechanisms of 2-methoxycinnamaldehyde against Bacillus cereus and its application in pork preservation.

Bacillus cereus is a major foodborne pathogen characterized by robust biofilm formation and increasing antimicrobial resistance. This study identified 2-Methoxycinnamaldehyde (MCA) from Toona sinensis as the principal antibacterial compound and evaluated its inhibitory mechanisms and preservation potential. MCA exhibited potent activity against B. cereus ATCC 11778 (MIC = 150 μg/mL; MBC = 200 μg/mL) and resistant strains. In vitro assays demonstrated that MCA induced concentration-dependent membrane disruption, DNA damage, and intracellular protein leakage. Furthermore, treatment at 1 MIC and

Breaking the Growth-Defense Trade-Off: bHLH Transcription Factors as Integrators of Development, Metabolism, and Yield in Artemisia annua.

Artemisinin, a sesquiterpene lactone produced by Artemisia annua, is synthesized and stored predominantly in glandular secretory trichomes (GSTs). Despite substantial advances in elucidating its biosynthetic pathway and transcriptional regulation, a key challenge remains: how developmental processes, hormone signaling, and metabolic pathways are coordinately integrated to overcome the trade-off between trichome density, metabolic flux, and vegetative biomass, which ultimately limits whole-plant yield. Recent studies have identified basic helix-loop-helix (bHLH) transcription factors (TFs) as c

Effects of intrauterine devices on proteins in the uterine lavage fluid of mares.

Intrauterine devices block luteolysis in cyclic mares, but the underlying mechanism is unknown. To clarify the mechanisms, the protein profile of the endometrial secretome was analyzed using two-dimensional difference gel electrophoresis (2D-DIGE). Twenty-seven mares were classified according to whether they were inseminated (AI) or had an intrauterine device (IUD), a water-filled plastic sphere, inserted into the uterus on Day 3 after ovulation. Uterine lavage fluids were collected on Day 15 from pregnant inseminated mares (AI-P; n = 8), non-pregnant inseminated mares (AI-N; n = 4), and mares

Characterization of the final step in the conversion of phytol into phytanic acid.

Phytol is a branched-chain fatty alcohol that is a naturally occurring precursor of phytanic acid, a fatty acid involved in the pathogenesis of Refsum disease. The conversion of phytol into phytanic acid is generally believed to take place via three enzymatic steps that involve 1) oxidation to its aldehyde, 2) further oxidation to phytenic acid, and 3) reduction of the double bond at the 2,3 position, yielding phytanic acid. Our recent investigations of this mechanism have elucidated the enzymatic steps leading to phytenic acid production, but the final step of the pathway has not been investi

A mechanically-induced colon cancer cell population shows increased metastatic potential.

BACKGROUND: Metastasis accounts for the majority of deaths from cancer. Although tumor microenvironment has been shown to have a significant impact on the initiation and/or promotion of metastasis, the mechanism remains elusive. We previously reported that HCT-8 colon cancer cells underwent a phenotypic transition from an adhesive epithelial type (E-cell) to a rounded dissociated type (R-cell) via soft substrate culture, which resembled the initiation of metastasis. The objective of current study was to investigate the molecular and metabolic mechanisms of the E-R transition. METHODS: Global
