Recent advances in pretreatment and detection technologies for ergot alkaloids.

Ergot alkaloids (EAs) are a class of mycotoxins widely present in complex matrices such as grains, pharmaceuticals, and biological samples. EAs possess vasoconstrictive and neurotoxic properties, which can lead to severe health issues such as gangrene and hallucinations. However, significant matrix effects, numerous co-existing interferents, and tedious sample pretreatment procedures in complex matrices pose substantial challenges for high-sensitivity and high-selectivity analysis. Accurate analysis of EAs is crucial for food safety and public health. This paper systematically reviews the rese…

Cytotoxic sesquiterpene alkaloids isolated from Nuphar pumila (Timm) de Candolle.

Five undescribed nupharidines, namely amidonupumones A-C (1-3), amidonupumol A (4), and dinupharolutine (7), along with twelve known analogues (5, 6, 8-17) were isolated from Nuphar pumila. Their structures were elucidated by extensive spectroscopic data, ECD, NMR calculations, and X-ray diffraction. Among them, compounds 1-4 represent the first alkaloids containing an amide structure isolated from N. pumila. In vitro bioassays revealed that compounds 7 and 11 exhibited broad-spectrum cytotoxic activity at a concentration of 40 μM.

Two new C19-diterpenoid alkaloids from Delphinium potaninii var. bonvalotii.

Two previously unreported C19-diterpenoid alkaloids, bonvalotizines A and B (1-2), together with fourteen previously identified compounds (3-16) were obtained from the plant of Delphinium potaninii var. bonvalotii (Franch.) W.T. Wang. Structural characterisation of the new compounds was achieved through spectroscopic analysis, including NMR (1D and 2D), HR-ESI-MS, and IR techniques. Biological evaluation revealed that none of the isolated compounds exhibited notable suppression of NO production in RAW 264.7 macrophages induced with lipopolysaccharide (LPS).

Asymmetric Total Syntheses of Strychnos Alkaloids via Selective Fischer Indolization.

The complex structures and important biological functions of Strychnos alkaloids have attracted a great deal of attention from synthetic chemists. Herein, we describe the concise asymmetric total syntheses of the Strychnos alkaloids, (-)-dehydrotubifoline, (-)-tubifoline, and (-)-tubifolidine, as well as the formal total synthesis of (-)-strychnine. Our strategy features the construction of the common tetracyclic pyrrolo[2,3-d]carbazole structure using regioselective Fischer indolization on unsymmetrical cyclic ketones and late-stage functionalization for divergent synthesis. We developed a st…
