Identification of a potent palladium-aryldiphosphine catalytic system for high-performance carbonylation of alkenes.

The development of stable and efficient ligands is of vital significance to enhance the catalytic performance of carbonylation reactions of alkenes. Herein, an aryldiphosphine ligand (L11) bearing the [Ph2P(ortho-C6H4)]2CH2 skeleton is reported for palladium-catalyzed regioselective carbonylation of alkenes. Compared with the industrially successful Pd/1,2-bis(di-tert-butylphosphinomethyl)benzene catalyst, catalytic efficiency catalyzed by Pd/L11 on methoxycarbonylation of ethylene is obtained, exhibiting better catalytic performance (TON: >2,390,000; TOF: 100,000 h-1; selectivity: >99%) and s

Ligand-Controlled Copper-Catalyzed Halo-Halodifluoromethylation of Alkenes and Alkynes Using Fluorinated Carboxylic Anhydrides.

Polyhalogenated molecules are often found as bioactive compounds in nature and are used as synthetic building blocks. Fluoroalkyl compounds hold promise for the development of novel pharmaceuticals and agrochemicals, as the introduction of fluoroalkyl groups is known to improve lipophilicity, membrane permeability, and metabolic stability. Three-component 1,2-halo-halodifluoromethylation reactions of alkenes are useful for their synthesis. However, general methods enabling the introduction of halodifluoromethyl (CF2X) and halogen (X') groups in the desired combination of X and X' are lacking.

Visible-Light-Mediated NHC and Tertiary Amine Catalysis Enabling α-H Acylation of Alkenes.

An intermolecular direct α-C-H acylation of alkenes was revealed by the visible-light-mediated N-heterocyclic carbene and quinuclidine catalysis. This convenient protocol provides a facile synthesis toward novel natural products and drug derivatives of α-substituted vinyl ketones. Mechanistic investigations indicated that the transformation proceeded via sequential radical addition, radical coupling, and an elimination process.

1,2-Aminoxyalkylation of alkenes with alkyl iodides and TEMPONa through SET- and XAT-processes.

1,2-Aminoxyalkylation of alkenes with alkyl iodides and TEMPONa in combination with an aryldiazonium salt as an XAT mediator is reported. Various primary, secondary and tertiary alkyl iodides engage as C-radical precursors in the 1,2-aminoxyalkylation with electrophilic alkenes as radical acceptors. The product alkoxyamines are readily transformed to the corresponding alcohols or ketones upon reduction or oxidation, respectively. Mechanistic investigations reveal that aryl radicals, generated through SET-reduction of the aryl diazonium salt with TEMPONa, engage in XAT from unactivated alkyl ha

Three-Component Asymmetric Ni-Catalyzed 1,2-Dicarbofunctionalization of Unactivated Alkenes via Stereoselective Migratory Insertion.

An asymmetric 1,2-dicarbofunctionalization of unactivated alkenes with aryl iodides and aryl/alkenylboronic esters under nickel/bioxazoline catalysis is disclosed. A wide array of aryl and alkenyl nucleophiles are tolerated, furnishing the products in good yield and with high enantioselectivity. In addition to terminal alkenes, 1,2-disubstituted internal alkenes participate in the reaction, establishing two contiguous stereocenters with high diastereoselectivity and moderate enantioselectivity. A combination of experimental and computational techniques shed light on the mechanism of the cataly

Nickel-Catalyzed Multicomponent Coupling: Synthesis of α-Chiral Ketones by Reductive Hydrocarbonylation of Alkenes.

A nickel-catalyzed, multicomponent regio- and enantioselective coupling via sequential hydroformylation and carbonylation from readily available starting materials has been developed. This modular multicomponent hydrofunctionalization strategy enables the straightforward reductive hydrocarbonylation of a broad range of unactivated alkenes to produce a wide variety of unsymmetrical dialkyl ketones bearing a functionalized α-stereocenter, including enantioenriched chiral α-aryl ketones and α-amino ketones. It uses chiral bisoxazoline as a ligand, silane as a reductant, chloroformate as a safe CO

Diastereoselective and Stereodivergent Synthesis of 2-Cinnamylpyrrolines Enabled by Photoredox-Catalyzed Iminoalkenylation of Alkenes.

A photoredox-catalyzed iminoalkenylation of γ-alkenyl O-acyl oximes has been developed. Readily available alkenylboronic acids serve as alkenylation reagents, leading to densely functionalized pyrrolines. Both (E)- and (Z)-cinnamylpyrrolines are accessible depending on the reaction solvent. In dichloromethane, (E)-cinnamylpyrrolines are produced through a photoredox-mediated single-electron-transfer process. In tetrahydrofuran, (Z)-cinnamylpyrrolines are generated by photocatalytic contra-thermodynamic E-to-Z isomerization of (E)-cinnamylpyrrolines though an energy-transfer pathway. Two stereo

Few-layer Black Phosphorous Catalyzes Radical Additions to Alkenes Faster than Low-valence Metals.

The substitution of catalytic metals by p-block main elements has a tremendous impact not only in the fundamentals but also in the economic and ecological fingerprint of organic reactions. Here we show that few-layer black phosphorous (FL-BP), a recently discovered and now readily available 2D material, catalyzes different radical additions to alkenes with an initial turnover frequency (TOF0) up to two orders of magnitude higher than representative state-of-the-art metal complex catalysts at room temperature. The corresponding electron-rich BP intercalation compound (BPIC) KP6 shows a nearly t

Enzymatic transformation of mercapturic acids derived from halogenated alkenes to reactive and mutagenic intermediates.

The metabolism of the mercapturic acids S-pentachlorobutadienyl-N-acetylcysteine (N-Ac-PCBC), S-trichlorovinyl-N-acetylcysteine (N-Ac-TCVC) and S-dichlorovinyl-N-acetylcysteine (N-Ac-DCVC) by subcellular fractions from male rat liver and kidney homogenates was studied. As a model compound, N-Ac-PCBC, 14C labelled, was synthesised. It was intensively metabolised by cytosolic but not by microsomal enzymes from rat liver and kidney. The major metabolite identified by GC/MS was pentachlorobutadienylcysteine, the amount produced being highest in kidney cytosol. Metabolic conversion of 14C-N-Ac-PCBC

A Bio-Based Chiroptical Overcrowded Alkene From Flavanone: Photoreactivity and Theoretical Investigation.

The design of light-driven overcrowded alkenes often relies on complex synthetic strategies to achieve precise control over rotational motion. In this study, we demonstrate a sustainable approach to design an overcrowded alkene and an O-heterocyclic phenanthrene derivative using a bio-based enantiopure flavanone, (-)-isolonchocarpin, as starting material. The synthesis involves a McMurry deoxygenative homocoupling reaction to generate a sterically hindered overcrowded alkene, which can further undergo photochemical (E/Z)-isomerization and oxidative 6π-electrocyclization. The oxidation product

Azide-Alkene Cycloaddition and Visible-Light-Mediated Wolff Rearrangement of Sugar Enones.

The synthesis of branched carbohydrate β-amino esters is described using the Wolff rearrangement of a diazoketone derived from the pyrolysis product (-)-levoglucosenone (LGO). An azide-alkene cycloaddition on LGO catalyzed by methanesulfonic acid directly afforded β-amino diazoketones via the triazoline. A subsequent visible-light-mediated Wolff rearrangement resulted in ring-contraction, giving a series of branched deoxyamino sugars with excellent diastereoselectivity (>20:1). The use of 2,2,2-trifluoroethanol as solvent at subambient temperature improved the yields of the ester relative to r

Transition Metal-Free Borata-Alkene/Alkyne Metathesis Enables Access to Cyclopentenes.

Metathesis reactions are fundamental transformations in organic synthesis, enabling efficient reorganization of unsaturated bonds and underpinning broad applications in pharmaceuticals, functional materials, and polymer science. To date, the conceptual and methodological development of metathesis transformations has been focused predominantly on carbon-carbon unsaturated bonds, and strategies that combine unsaturated bond reorganization with the direct incorporation of heteroatoms remain rare. Additionally, the established metathesis methodologies have been developed typically within the frame

Shedding Light on Photochemical Activation and Catalytic Mechanism of Cobalt-Catalyzed Alkene Hydroaminocarbonylation.

In a recent experimental study, a cobalt-catalyzed, light-induced synthesis of amides, achieving 100% atom economy by coupling amines and alkenes under 390 nm LED illumination, was reported. Despite its practical appeal, important mechanistic questions remain, particularly regarding the photoinduced generation of the active catalyst and the subsequent catalytic cycle. Herein, we shed light onto the mechanism of this transformation. Our results revealed how vibronic and spin-orbit coupling effects synergistically enhance the photoabsorption of the precatalyst at 390 nm, driving the formation of

Photocatalytic Cross-Coupling of Aldehydes and Alkenes for Aryl Vinyl Ketones by a Single Catalyst.

Herein is the first example of photocatalytic cross-coupling of alkenes with aldehydes by a single catalyst without an external photosensitizer and any additives. Irradiation of the aromatic aldehyde and cobaloxime catalyst results in the formation of an acyl radical, which undergoes radical addition with alkene or indole and subsequently β-H elimination to afford alkenyl ketone. The reaction features cheap and readily available raw materials, a broad substrate scope, and mild conditions, even for late-stage derivatization of bioactive compounds.

Recent advances in the sulfonylation of alkenes with the insertion of sulfur dioxide via radical reactions.

Recent advances in the sulfonylation of alkenes via the insertion of sulfur dioxide are summarized. The sulfur dioxide surrogate of DABCO·(SO2)2 or inorganic sulfites is used in the transformation through a radical process. Two strategies for the sulfonylation of alkenes with the insertion of sulfur dioxide have been developed. In most cases, the vicinal difunctionalization of alkenes is initiated by the arylsulfonyl radical generated in situ. Although the sulfonylation of alkenes with the insertion of sulfur dioxide is efficient, the asymmetric versions are still challenging and less to be ex

Multigram-Scale Asymmetric Alkene Reduction Catalyzed by a Thermostable Flavin Ene-Reductase.

Chiral-substituted carbonyl compounds can be sustainably obtained via asymmetric alkene reduction catalyzed by ene-reductases from the Old Yellow Enzyme (OYE) family. Yet OYEs are seldom implemented in scale-up reactions due to low turnover numbers (TONs). Herein, we demonstrate multigram 150 g/L scale reactions with the thermostable OYE from Thermus scotoductus (0.2 wt %) for the asymmetric reduction of monoterpenes. Best results were achieved with (S)-carvone (7.5 g), affording a record TON of 123,000, with 90% isolated yield and >99% enantiomeric excess of (2R,5S)-dihydrocarvone, and an env
